The cyclic guanosine monophosphate-adenosine monophosphate synthase-stimulator of interferon(IFN)genes(cGAS-STING)signaling pathway is crucial for sensing abnormal DNA accumulation in the *** binds to abnormal DNA,cGAS catalyzes the production of second messenger cyclic dinucleotides,followed by the activation of downstream *** activation induces the expression of type I interferon and other inflammatory cytokines,ultimately initiating an immune *** to the involvement of the cGAS-STING pathway in various diseases,including infection,tumor,autoimmune disease and kidney disease,ongoing research is focused on developing drugs and treatment methods to target and regulate this *** the development of nanotechnology,nanomedicines targeting cGAS-STING signaling are of great significance in clinical applications due to their targeted delivery,controlled drug release,improved solubility,multifunctionality,and enhanced *** comprehensive review focuses on the most recent progress of nanoplatforms targeting cGAS-STING in disease therapy,aiming to provide references and guidelines for further design and optimization of nanomedicines.

Corrosion of reinforcement induced by chloride invasion is extensively considered as the dominating deterioration mechanism of reinforced concrete(RC)structures,leading to serious safety hazards and tremendous economic ***,it still lacks well dispersive and cost-efficient nanomaterials to improve the anti-chloride-corrosion ability of RC ***,specific carbon dots(CDs)with high dispersity and low cost are deliberately designed,successfully prepared by hydrothermal processing,and then firstly applied to immensely enhance chloride binding performance of cement,thereby contributing to suppressing the corrosion of ***,the tailored CDs are composed of the carbon core with highly crystalline sp^(2)C structures and oxygen-containing groups connecting on the carbon core;The typical equilibrium test confirms that with respect to that of the blank cement paste,the chloride binding capacity of cement paste involving 0.2 wt%(by weight of cement)CDs is increased by 109% after 14-day exposure to 3 mol/L Na Cl solution;according to comprehensive analyses of phase compositions,the chloride binding mechanism of CDs-modified cement is rationally attributed to the fact that the incorporation of CDs advances the formation of calcium silicate hydrate(C-S-H)gels and Friedel's salt(Fs),thus enormously enhancing the physically adsorbed and chemically bound chloride ions of cement *** work not only firstly provides a novel high-dispersity and low-cost nanomaterial toward the durability enhancement of RC structures,but also broadens the application of CDs in the field of engineering,conducing to stimulating their industrialization development.

Microbially induced carbonate precipitation(MICP)is a promising technique for the autonomous healing of concrete *** this study,the effect of pH on MICP was *** results indicate that the MICP process was inhibited when the pH was higher than *** vaterite and calcite were produced when the pH was*** concrete aggregates(RCA)coated with sodium silicate have been proposed as protective carriers for microbial healing *** the presence of the coated RCA resulted in a loss of the splitting tension strength of the concrete,the loaded healing agents were highly efficient in self-healing *** incorporated with 20%RCA loaded with healing agents exhibited the best self-healing *** the initial crack widths were between 0.3 and 0.4 mm,the 7-d mean healing rate was approximately 90%.At 28 d,the crack area filling ratio was 86.4%,while its water tightness recovery ratio was 74.4%and 29.8%,respectively,for rapid and slow *** study suggests that RCA coated with sodium silicate is an effective method for packaging microbial healing agents and has great potential for developing cost-effective self-healing concrete.

Dear Editor,This letter proposes a parameter-free multiple kernel clustering(MKC)method by using shifted Laplacian *** MKC can effectively cluster nonlinear data,but it faces two main challenges:1)As an unsupervised method,it is up against parameter problems which makes the parameters intractable to tune and is unfeasible in real-life applications;2)Only considers the clustering information,but ignores the interference of noise within Laplacian.

Primary sclerosing cholangitis(PSC)is an autoimmune cholangiopathy characterized by chronic inflammation of the biliary epithelium and periductal fibrosis,with no curative treatment available,and liver transplantation is inevitable for end-stage *** placentalmesenchymal stem cell(hpMSC)-derived exosomes have demonstrated the ability to prevent fibrosis,inhibit collagen production and possess immunomodulatory properties in autoimmune liver ***,we prepared hpMSC-derived exosomes(Exo^(MSC))and further investigated the anti-fibrotic effects and detailed mechanism on PSC based on Mdr2^(−/−)mice and multicellular organoids established from PSC *** results showed that Exo^(MSC) ameliorated liver fibrosis in Mdr2^(−/−)mice with significant collagen reduction in the preductal area where Th17 differentiation was inhibited as demonstrated by RNAseq analysis,and the percentage of CD4+IL-17A+T cells was reduced both in Exo^(MSC)-treated Mdr2^(−/−)mice(Mdr2^(−/−)-Exo)in vivo and Exo^(MSC)-treated Th17 differentiation progressed in ***,Exo^(MSC) improved the hypersecretory phenotype and intercellular interactions in the hepatic Th17 microenvironment by regulating PERK/CHOP signaling as supported by multicellular ***,our data demonstrate the antifibrosis effect of Exo^(MSC) in PSC disease by inhibiting Th17 differentiation,and ameliorating the Th17-induced microenvironment,indicating the promising potential therapeutic role of Exo^(MSC) in liver fibrosis of PSC or Th17-related diseases.

目的建立间接竞争酶联免疫吸附(indirect competitive enzyme-linked immunosorbent assay,ic-ELISA)法用于保健食品中糖皮质激素类药物多残留检测。方法将可的松、地塞米松分别与丁二酸酐衍生制备免疫半抗原,倍他米松衍生制备包被半抗原,采用活泼酯法与载体蛋白偶联制备人工抗原,采用紫外(ultraviolet,UV)吸收光谱鉴定及基质辅助激光解析串联飞行时间质谱(matrix assisted laser desorption-tandem time of flight-mass spectrometry,MALDI-TOF-MS)测定人工抗原的偶联比。通过动物免疫,制备针对糖皮质激素类药物的兔多克隆抗体,建立保健食品中糖皮质激素类药物多残留检测ic-ELISA法。结果地塞米松丁二酸酐衍生物为免疫半抗原,倍他米松丁二酸酐衍生物为包被半抗原。通过优化实验条件,确定了最佳的包被原质量浓度为1.25μg/m L,以及最佳的抗体稀释倍数为9000倍。在此条件下,本研究建立的ic-ELISA法对11种糖皮质激素类药物的半抑制质量浓度为0.68~40.17 ng/m L,展现了良好的检测灵敏度。使用甲醇超声提取,样品提取液通过标准稀释液稀释500倍消除基质效应,11种目标分析物在4种剂型保健食品中的添加回收率为81.2%~118.7%,变异系数低于14.00%。结论本研究成功制备糖皮质激素类药物的广谱性多克隆抗体,并建立了保健食品中糖皮质激素类药物多残留检测ic-ELISA法,可用于保健食品中糖皮质激素类药物的快速筛查。
