Prediction of cell-cell communication patierns of dorsal root ganglion cells: single-cell RNA sequencing data analysis

Dorsal root ganglion neurons transmit peripheral somatic information to the central nervous system, and dorsal root ganglion neuron excitability affects pain perception. Dorsal root ganglion stimulation is a new approach for managing pain sensation. Knowledge of the cell-cell communication among dorsal root ganglion cells may help in the development of new pain and itch management strategies. Here, we used the single-cell RNA-sequencing(scRNA-seq) database to investigate intercellular communication networks among dorsal root ganglion cells. We collected scRNA-seq data from six samples from three studies, yielding data on a total of 17,766 cells. Based on genetic profiles, we identified satellite glial cells, Schwann cells, neurons, vascular endothelial cells, immune cells, fibroblasts, and vascular smooth muscle cells. Further analysis revealed that eight types of dorsal root ganglion neurons mediated proprioceptive, itch, touch, mechanical, heat, and cold sensations. Moreover, we predicted several distinct forms of intercellular communication among dorsal root ganglion cells, including cell-cell contact, secreted signals, extracellular matrix, and neurotransmitter-mediated signals. The data mining predicted that Mrgpra3-positive neurons robustly express the genes encoding the adenosine Adora2b(A2B) receptor and glial cell line-derived neurotrophic factor family receptor alpha 1(GFRα-1). Our immunohistochemistry results confirmed the coexpression of the A2B receptor and GFRα-1. Intrathecal injection of the A2B receptor antagonist PSB-603 effectively prevented histamine-induced scratching behaviour in a dose-dependent manner. Our results demonstrate the involvement of the A2B receptor in the modulation of itch sensation. Furthermore, our findings provide insight into dorsal root ganglion cell-cell communication patterns and mechanisms. Our results should contribute to the development of new strategies for the regulation of dorsal root ganglion excitability.

Dynamic emission characteristics and control strategies of air pollutants from motor vehicles in downtown Beijing,China

This study combined the real-time monitoring and investigation of traffic flows to comprehensively analyze the road traffic flow and vehicle structure in downtown Beijing.A dynamic emission inventory of motor vehicle air pollutants in downtown Beijing in 2021 was established,and the impact of these emissions on air quality was simulated and quantified,and different emission reduction control scenarios were proposed to evaluate their environmental improvement effects and explore measures to mitigate the impact of pollution emissions.The results show that the high traffic flow and the structure of the motor vehicle emissions in downtown Beijing are the main causes of severe motor vehicle pollution.Monitoring data shows that traffic flow in central Beijing is dominated by small passenger vehicles,while the vehicle mix is better than in other regions,with 72.0%of vehicles meeting"National V"or higher emission standards.However,to achieve higher air quality goals,further reducing vehicle emissions is necessary.Based on dynamic traffic flow,the average daily emissions of nitrogen oxides(NOX),particulate matters(PM_(2.5))and volatile organic compounds(VOC)from motor vehicles in central Beijing are 17.7 tons,0.6 tons and 14.0 tons,respectively,accounting for 23.0%of the city’s average daily motor vehicle emissions.If a zero-emission zone for motor vehicles were implemented in central Beijing,the annual average emission level of pollutants will be reduced by 10.4%to 21.0%.The designation of ultra-low emission zones for motor vehicles could be effective in improving the air quality in the center of Beijing.

Primary Amebic Meningoencephalitis Caused by Naegleria fowleri in China: A Case Report

Naegleria fowleri is a thermophilic,free-living ameba that is commonly known as the“brain-eating ameba.”This parasite invades the central nervous system and causes an acute fulminant infection,which results in a fatal form of primary amebic meningoencephalitis(PAM).Here,we report a case of PAM by an unexpected transmission route.Regrettably,the patient deteriorated swiftly and passed away subsequent to diagnosis,despite our exhaustive efforts.Here,we compiled a summary of 11 cases of PAM survivors who have all been treated with amphotericin,rifampicin and triazoles.Beside this regimen,miltefosine was also used successfully to treat PAM in an adolescent.More precise understanding of the survival rate and contributing factors can be established with further validation on ad-ditional cases.

Network Embedding Algorithm for Vulnerability Assessment of Power Transmission Lines Using Integrated Structure and Attribute Information

In power systems,failures of vulnerable lines can trigger large-scale cascading failures,and vulnerability assessment is dedicated to locating these lines and reducing the risks of such failures.Based on a structure and attribute network embedding(SANE)algorithm,a novel quantitative vulnerability analysis method is proposed to identify vulnerable lines in this research.First,a two-layered random walk network with topological and electrical properties of transmission lines is established.Subsequently,based on the weighted degree of nodes in the two-layered network,the inter-layer and intra-layer walking transition probabilities are developed to obtain walk sequences.Then,a Word2Vec algorithm is applied to obtain lowdimension vectors representing transmission lines,according to obtained walk sequences for calculating the vulnerability index of transmissions lines.Finally,the proposed method is compared with three widely used methods in two test systems.Results show the network embedding based method is superior to those comparison methods and can provide guidance for identifying vulnerable lines.

Mixed D-vine copula-based conditional quantile model for stochastic monthly streamflow simulation

Copula functions have been widely used in stochastic simulation and prediction of streamflow.However,existing models are usually limited to single two-dimensional or three-dimensional copulas with the same bivariate block for all months.To address this limitation,this study developed a mixed D-vine copula-based conditional quantile model that can capture temporal correlations.This model can generate streamflow by selecting different historical streamflow variables as the conditions for different months and by exploiting the conditional quantile functions of streamflows in different months with mixed D-vine copulas.The up-to-down sequential method,which couples the maximum weight approach with the Akaike information criteria and the maximum likelihood approach,was used to determine the structures of multivariate Dvine copulas.The developed model was used in a case study to synthesize the monthly streamflow at the Tangnaihai hydrological station,the inflow control station of the Longyangxia Reservoir in the Yellow River Basin.The results showed that the developed model outperformed the commonly used bivariate copula model in terms of the performance in simulating the seasonality and interannual variability of streamflow.This model provides useful information for water-related natural hazard risk assessment and integrated water resources management and utilization.

Living fossils are evidence of long-term sustained ecological success.However,whether living fossils have little molecular changes remains poorly known,particularly in plants.Here,we have introduced a novel method that integrates phylogenomic,comparative genomic,and ecological niche modeling analyses to investigate the rate of molecular evolution of Eupteleaceae,a Cretaceous relict angiosperm family endemic to East Asia.We assembled a high-quality chromosome-level nuclear genome,and the chloroplast and mitochondrial genomes of a member of Eupteleaceae(Euptelea pleiosperma).Our results show that Eupteleaceae is most basal in Ranunculales,the earliest-diverging order in eudicots,and shares an ancient whole-genome duplication event with the other Ranunculales.We document that Eupteleaceae has the slowest rate of molecular changes in the observed angiosperms.The unusually low rate of molecular evolution of Eupteleaceae across all three independent inherited genomes and genes within each of the three genomes is in association with its conserved genome architecture,ancestral woody habit,and conserved niche requirements.Our findings reveal the evolution and adaptation of living fossil plants through large-scale environmental change and also provide new insights into early eudicot diversification.

Evolution of Cenozoic sedimentary architecture in Central and Southern South China Sea basins

The Central and Southern South China Sea(CSSCS) has a complex tectonic dynamic background and abundant oil and gas resources, which has always been a hot topic of academic and industrial attention.However, systematic analyses are still lacking regarding its sediment filling structure and evolution, mostly due to limited borehole penetration and poor quality of seismic reflection data for deeply buried sequences. No consensus has been reached yet on the sedimentary infilling processes, which impeded the reconstruction of the palaeogeography of Southeast Asia and the oil-and-gas exploration undertakings. Here, we illustrate the Cenozoic sedimentary evolution of the CSSCS region by synthesizing relevant data from previous literature and our own observations and displaying the evolution of depositional systems in sequential reconstructions. Besides, the controlling factors of preferred sedimentary scenarios in the CSSCS incorporate the latest interpretations of the spreading of South China Sea(SCS) as well as the demise of the hypothetical Proto-South China Sea(PSCS). The results show that there are three types of sedimentary basins in the CSSCS(foreland,strike-slip, and rift basins) with different sedimentary filling structures. The foreland basins formed a depositional pattern of ‘transition from deep water to shallow water environments', dominated by deep-water depositional systems which were formed before the Early Oligocene with submarine fans developed. Later,the foreland basins were gradually dominated by shallow-water depositional systems with deltas and shallow marine facies. The strike-slip basins showed the depositional architecture of ‘transition from lake to marine environments', i.e. the basins were dominated by lacustrine deposits during the Eocene and evolved into the marine depositional environment since Oligocene with delta developed in the western part of the basin. BACKGROUND Obesity has become a serious public health issue,significantly elevating the risk of various complications.It is a well-established contributor to Heart failure with preserved ejection fraction(HFpEF).Evaluating HFpEF in obesity is crucial.Epicardial adipose tissue(EAT)has emerged as a valuable tool for validating prognostic biomarkers and guiding treatment targets.Hence,assessing EAT is of paramount importance.Cardiovascular magnetic resonance(CMR)imaging is acknowledged as the gold standard for analyzing cardiac function and mor-phology.We hope to use CMR to assess EAT as a bioimaging marker to evaluate HFpEF in obese patients.AIM To assess the diagnostic utility of CMR for evaluating heart failure with preserved ejection fraction[HFpEF;left ventricular(LV)ejection fraction≥50%]by measuring the epicardial adipose tissue(EAT)volumes and EAT mass in obese patients.METHODS Sixty-two obese patients were divided into two groups for a case-control study based on whether or not they had heart failure with HFpEF.The two groups were defined as HFpEF+and HFpEF-.LV geometry,global systolic function,EAT volumes and EAT mass of all subjects were obtained using cine magnetic resonance sequences.RESULTS Forty-five patients of HFpEF-group and seventeen patients of HFpEF+group were included.LV mass index(g/m2)of HFpEF+group was higher than HFpEF-group(P<0.05).In HFpEF+group,EAT volumes,EAT volume index,EAT mass,EAT mass index and the ratio of EAT/[left atrial(LA)left-right(LR)diameter]were higher compared to HFpEF-group(P<0.05).In multivariate analysis,Higher EAT/LA LR diameter ratio was associated with higher odds ratio of HFpEF.CONCLUSION EAT/LA LR diameter ratio is highly associated with HFpEF in obese patients.It is plausible that there may be utility in CMR for assessing obese patients for HFpEF using EAT/LA LR diameter ratio as a diagnostic biomarker.Further prospective studies,are needed to validate these proof-of-concept findings.

Establishing Modular Cell-Free Expression System for the Biosynthesis of Bicyclomycin from a Chemically Synthesized Cyclodipeptide

Cell-free expression systems have emerged as a versatile and powerful platform for metabolic engineering,biosynthesis and synthetic biology studies.Nevertheless,successful examples of the synthesis of complex natural products using this system are still limited.Bicyclomycin,a structurally unique and complex diketopiperazine alkaloid,is a clinically promising antibiotic that selectively inhibits the transcription termination factor Rho.Here,we established a modular cell-free expression system with cascade catalysis for the biosynthesis of bicyclomycin from a chemically synthesized cyclodipeptide.The six cell-free expressed biosynthetic enzymes,including five iron-andα-ketoglutarate-dependent dioxygenases and one cytochrome P450 monooxygenase,were active in converting their substrates to the corresponding products.The co-expressed enzymes in the cell-free module were able to complete the related partial pathway.In vitro biosynthesis of bicyclomycin was also achieved by reconstituting the entire biosynthetic pathways(i.e.,six enzymes)using the modular cell-free expression system.This study demonstrates that the modular cell-free expression system can be used as a robust and promising platformforthe biosynthesis of complex antibiotics.

Nomogram to predict severe retinopathy of prematurity in Southeast China

AIM:To define the predictive factors of severe retinopathy of prematurity(ROP)and develop a nomogram for predicting severe ROP in southeast China.METHODS:Totally 554 infants diagnosed with ROP hospitalized in the Second Affiliated Hospital and Yuying Children’s Hospital of Wenzhou Medical University and hospitalized in Taizhou Women and Children’s Hospital were included.Clinical data and 43 candidate predictive factors of ROP infants were collected retrospectively.Logistic regression model was used to identify predictive factors of severe ROP and to propose a nomogram for individual risk prediction,which was compared with WINROP model and Digirop-Birth model.RESULTS:Infants from the Second Affiliated Hospital and Yuying Children’s Hospital of Wenzhou Medical University(n=478)were randomly allocated into training(n=402)and internal validation group(n=76).Infants from Taizhou Women and Children’s Hospital were set as external validation group(n=76).Severe ROP were found in 52 of 402 infants,12 of 76 infants,and 7 of 76 infants in training group,internal validation group,and external validation group,respectively.Birth weight[odds ratio(OR),0.997;95%confidence interval(CI),0.996-0.999;P<0.001],multiple births(OR,1.885;95%CI,1.013-3.506;P=0.045),and non-invasive ventilation(OR,0.288;95%CI,0.146-0.570;P<0.001)were identified as predictive factors for the prediction of severe ROP,by univariate analysis and multivariate analysis.For predicting severe ROP based on the internal validation group,the areas under receiver operating characteristic curve(AUC)was 78.1(95%CI,64.2-92.0)for the nomogram,32.9(95%CI,15.3-50.5)for WINROP model,70.2(95%CI,55.8-84.6)for Digirop-Birth model.In external validation group,AUC of the nomogram was also higher than that of WINROP model and Digirop-Birth model(80.2 versus 51.1 and 63.4).The decision curve analysis of the nomogram demonstrated better clinical efficacy than that of WINROP model and Digirop-Birth model.The calibration curves demonstrated a good consistency between the actual seve
