Quantitative Extraction of p-Coumaric Acid and Ferulic Acid in Different Gramineous Materials and Structural Changes of Residual Alkali Lignin

Ferulic acid(FA)and p-coumaric acid(pCA)in bagasse,wheat straw,corn straw,and corncob were extracted by alkaline hydrolysis and characterized by gas chromatography(GC)and gas chromatography-mass spectrometry(GC-MS).It was found that the FA and most of the pCA in gramineous biomass could be dissociated and released after being treated with 1 M NaOH at 100℃for 4 h.The yields of pCA/FA in bagasse,wheat straw,corn straw,and corncob determined by GC-FID are 39.8/11.5,13.7/11.0,28.0/11.0,and 35.1/14.5 mg/g,respectively.The raw materials and the treated solid residues were characterized by gel-state 2D Heteronuclear Single Quantum Coherence Nuclear Magnetic Resonance(2D HSQC NMR).It was found that only a small amount of lignin was detected in the residue after alkali treatment,indicating that the alkali treatment conditions can effectively cleave the FA and pCA.Additionally,the lignin in the alkali solution was recovered and characterized by 2D HSQC NMR.The FA was not able to be detected by NMR,whereas a small amount of pCA remained in the alkali lignin.This study reveals the structural change of residual lignins during the quantitative isolation of FA and pCA,which is essential for the selective isolation of pCA/FA and valorization of residual alkali lignin.

Background The PACS gene family has been demonstrated to be related to intracellular vesicular trafficking.The phenotypic manifestations caused by the pathogenic variants of PACS include epilepsy,intellectual disability/developmental delay,and malformations,such as facial abnormalities.Methods We identified seven new cases with pathogenic or likely pathogenic PACS variants using next-generation sequencing.Detailed information obtained from these patients was analyzed along with that obtained from previously reported patients.Results With the inclusion of the newly diagnosed cases in this study,103 cases with PACS gene family-related neurological diseases were reported,of which 43 were PACS2-related cases and the remaining were PACSI-related cases.Most patients had seizures,which have been reported to be effectively controlled by several types of anti-seizure medications(ASMs).The most efficacious and frequently prescribed ASMs included sodium valproate(43.3%,13/30),oxcarbazepine/carbamazepine(26.7%,8/30),and levetiracetam(20%,6/30).Almost all patients had intellectual disability/developmental delay.The most common pathogenic missense variants were PACSI p.Arg203Trp and PACS2 p.Glu209Lys.In addition,we report a patient carrying a likely pathogenic copy number variation(CNV)(de novo heterozygous deletion of chr14:105821380-106107443,286 kilobase,destroyed part of the furin-binding region domain and the protein structure after it)with more severe and refractory late-onset epilepsy.Conclusions The clinical phenotypes of the different PACS heterozygous missense variants were similar.The pathogenic variant sites of PACSI and PACS2 were quite limited but located in different regions.A CNV destroying part of the PACS2 gene might also be pathogenic.These findings may provide an important clue for further functional studies on the pathogenic mechanism of neurological disorders related to the PACS gene family.

Molecular insights into the circadian clock in marine diatoms

The circadian clock is a fundamental endogenous mechanism of adaptation that coordinates the physiology and behavior of most organisms with diel variations in the external environment to maintain temporal homeostasis.Diatoms are the major primary producers in the ocean.However,little is known about the circadian clock in marine diatoms compared with other organisms.Here,we investigated circadian clock genes,their expression patterns,and responses to environmental stimuli such as light,nitrogen and phosphorus in two marine diatoms,Skeletonema costatum and Phaeodactylum tricornutum,using a combination of q RT-PCR and bioinformatic analysis.We identified 17 and 18 circadian clock genes in P.tricornutum and S.costatum,respectively.Despite significant evolutionary differences,these genes were similar to those of the higher plant Arabidopsis.We also established a molecular model for the marine diatom circadian clock comprising an input pathway,core oscillator,output pathway,and valve effector.Notably,the expression patterns of core clock genes(circadian clock associated 1(CCA1),late elongated hypocotyl(LHY)and timing of cab 1(TOC1))in both species differed from those of terrestrial plants.Furthermore,the expression of these genes was influenced by variations in ambient light,nitrogen and phosphorus availability.Although marine diatoms and higher plants share common circadian clock components,their clock genes have diverged throughout evolution,likely as a result of adapting to contrasting environments.

Traditional Chinese Medicine(TCM)has been extensively used as a mainstay for treating various pathologies.Combing the pharmacology and systems biology approaches,the network pharmacology(NP)approach was developed to predict the probable mechanism underlying the therapeutic effect of TCM.However,approaches solely based on NP cannot effectively elucidate the curative mechanism in a holistic and reliable manner due to limitations in NP-based methods and complexity of TCM components.Thus,integration strategies combining NP with other approaches are increasingly being used.Since the interdisciplinary research in TCM has received much attention in the advent of the big data era of which the NPbased integration strategy is broadly used,the strategy is clearly elaborated in the present review.We summarized several NP-based integration strategies and their applications in TCM studies,including multi-omics approach,gut microbiota study,chemical information analysis,data-mining,and network toxicology study.

With the rapidly growing abuse of drones, monitoring and classification of birds and drones have become a crucial safety issue. With similar low radar cross sections(RCSs), velocities, and heights, drones are usually difficult to be distinguished from birds in radar measurements. In this paper, we propose to exploit different periodical motions of birds and drones from highresolution Doppler spectrum sequences(DSSs) for classification.This paper presents an elaborate feature vector representing the periodic fluctuations of RCS and micro kinematics. Fed by the Doppler spectrum and feature sequence, the long to short-time memory(LSTM) is used to solve the time series classification.Different classification schemes to exploit the Doppler spectrum series are validated and compared by extensive real-data experiments, which confirms the effectiveness and superiorities of the proposed algorithm.

Although multifunctional aerogels are expected to be used in applications such as portable electronic devices,it is still a great challenge to confer multifunctionality to aerogels while maintaining their inherent microstructure.Herein,a simple method is proposed to prepare multifunctional NiCo/C aerogels with excellent electromagnetic wave absorption properties,superhydrophobicity,and self-cleaning by water-induced NiCo-MOF self-assembly.Specifically,the impedance matching of the three-dimensional(3D)structure and the interfacial polarization provided by CoNi/C as well as the defect-induced dipole polarization are the primary contributors to the broadband absorption.As a result,the prepared NiCo/C aerogels have a broadband width of 6.22 GHz at 1.9 mm.Due to the presence of hydrophobic functional groups,CoNi/C aerogels improve the stability in humid environments and obtain hydrophobicity with large contact angles>140°.This multifunctional aerogel has promising applications in electromagnetic wave absorption,resistance to water or humid environments.

Human immunodeficiency virus(HIV)infection is a significant and pressing concern for public health worldwide.[1]Midway through the 1990s,the widespread use of antiretroviral therapy(ART)has resulted in a significant paradigm shift in the management of HIV infection,thereby altering its prognosis from a lifethreatening disease to a chronic and managed health condition.According to previous studies,people who are living with HIV(PLWH)that are on current ART have shown a decrease in both the morbidity and mortality associated with HIV infection,along with a decreased risk of transmitting the virus to others.

Particulate pollution is a global risk factor that seriously threatens human health.Fine particles(FPs)and ultrafine particles(UFPs)have small particle diameters and large specific surface areas,which can easily adsorb metals,microorganisms and other pollutants.FPs and UFPs can enter the human body in multiple ways and can be easily and quickly absorbed by the cells,tissues and organs.In the body,the particles can induce oxidative stress,inflammatory response and apoptosis,furthermore causing great adverse effects.Epidemiological studies mainly take the population as the research object to study the distribution of diseases and health conditions in a specific population and to focus on the identification of influencing factors.However,the mechanism by which a substance harms the health of organisms is mainly demonstrated through toxicological studies.Combining epidemiological studies with toxicological studies will provide a more systematic and comprehensive understanding of the impact of PM on the health of organisms.In this review,the sources,compositions,and morphologies of FPs and UFPs are briefly introduced in the first part.The effects and action mechanisms of exposure to FPs and UFPs on the heart,lungs,brain,liver,spleen,kidneys,pancreas,gastrointestinal tract,joints and reproductive system are systematically summarized.In addition,challenges are further pointed out at the end of the paper.This work provides useful theoretical guidance and a strong experimental foundation for investigating and preventing the adverse effects of FPs and UFPs on human health.

Microwave dielectric ceramics(MWDCs)with low dielectric constant and low dielectric loss are desired in contemporary society,where the communication frequency is developing to high frequency(sub-6G).Herein,Nd_(2)(Zr_(1−x)Ti_(x))_(3)(MoO_(4))_(9)(NZ_(1−x)T_(x)M,x=0.02-0.10)ceramics were prepared through a solid-phase process.According to X-ray diffraction(XRD)patterns,the ceramics could form a pure crystal structure with the R3c(167)space group.The internal parameters affecting the properties of the ceramics were calculated and analyzed by employing Clausius-Mossotti relationship,Shannon’s rule,and Phillips-van Vechten-Levine(P-V-L)theory.Furthermore,theoretical dielectric loss of the ceramics was measured and analyzed by a Fourier transform infrared(IR)radiation spectrometer.Notably,when x=0.08 and sintered at 700℃,optimal microwave dielectric properties of the ceramics were obtained,including a dielectric constant(ε_(r))=10.94,Q·f=82,525 GHz(at 9.62 GHz),and near-zero resonant frequency temperature coefficient(τ_(f))=−12.99 ppm/℃.This study not only obtained an MWDC with excellent properties but also deeply analyzed the effects of Ti^(4+)on the microwave dielectric properties and chemical bond characteristics of Nd_(2)Zr_(3)(MoO_(4))_(9)(NZM),which laid a solid foundation for the development of rare-earth molybdate MWDC system.
