The cornea is a highly specialized and unique organ in the human body. Its main function is to project light from the external environment onto the retina, and it has a specific transparency to perform its function properly. The transparency and integrity of the cornea is of vital importance. The corneal wound, especially laceration deep to Bowman＇s membrane and stroma, which will inevitably cause scar formation, may cause the degeneration or even loss of sight.

In Alzheimer’s disease and ischemic stroke,intranasal insulin can act as a neuroprotective ***,whether intranasal insulin has a neuroprotective effect in intracerebral hemorrhage and its potential mechanisms remain poorly *** this study,a mouse model of autologous blood-induced intracerebral hemorrhage was treated with 0.5,1,or 2 IU insulin via intranasal delivery,twice per day,until 24 or 72 hours after *** with saline treatment,1 IU intranasal insulin treatment significantly reduced hematoma volume and brain edema after cerebral hemorrhage,decreased blood-brain barrier permeability and neuronal degeneration damage,reduced neurobehavioral deficits,and improved the survival rate of *** levels of p-AKT and p-GSK3βwere significantly increased in the perihematoma tissues after intranasal insulin *** findings suggest that intranasal insulin therapy can protect the neurological function of mice after intracerebral hemorrhage through the AKT/GSK3βsignaling *** study was approved by the Ethics Committee of the North Sichuan Medical College of China(approval ***(A)2019(01))on January 7,2019.

Advanced intrahepatic cholangiocarcinoma(ICC)has a dismal ***,we report the efficacy and safety of combining toripalimab,lenvatinib,and gemcitabine plus oxaliplatin(GEMOX)as first-line therapy for advanced *** patients with pathologically confirmed advanced ICC received intravenous gemcitabine(1 g/m2)on Days 1 and 8 and oxaliplatin(85 mg/m2)Q3W for six cycles along with intravenous toripalimab(240 mg)Q3W and oral lenvatinib(8 mg)once daily for one *** expression of programmed death-ligand 1(PD-L1)and genetic status was investigated in paraffin-embedded tissues using immunohistochemistry and whole-exome sequencing(WES)*** primary endpoint was the objective response rate(ORR).Secondary outcomes included safety,overall survival(OS),progression-free survival(PFS),disease control rate(DCR)and duration of response(DoR).As of July 1,2022,the median follow-up time was 23.5 months,and the ORR was 80%.Twenty-three patients achieved partial response,and one achieved complete ***(21/30)with DNA damage response(DDR)-related gene mutations showed a higher ORR,while patients(14/30)with tumor area positivity≥1(PD-L1 staining)showed a trend of high ORR,but without significant *** median OS,PFS,and DoR were 22.5,10.2,and 11.0 months,*** DCR was 93.3%.Further,56.7%of patients experienced manageable grade≥3 adverse events(AEs),commonly neutropenia(40.0%)and leukocytopenia(23.3%).In conclusion,toripalimab plus lenvatinib and GEMOX are promising first-line regimens for the treatment of advanced ICC.A phase-III,multicenter,double-blinded,randomized study to validate our findings was approved by the National Medical Products Administration(NMPA,No.2021LP01825).

Background Human amniotic epithelial cells （HAECs）, which have several characteristics similar to stem cells, therefore could possibly be used in cell therapy without creating legal or ethical problems. In this study, we transplanted HEACs into the injured spinal cord of rats to investigate if the cells can improve the rats＇ hindlimb motor function. Methods HAECs were obtained from a piece of fresh amnion, labeled with Hoechst33342, and transplanted into the site of complete midthoracic spinal transections in adult rats. The rats （n=21） were randomly divided into three groups： Sham-operation group （n=7）, cells-graft group （n=7）, and PBS group （n=7）. One rat of each group was killed for histological analysis at the second week after the transplantation. The other six rats of each group were killed for histological analysis after an 8-week behavioral testing. Hindlimb motor function was assessed by using the open-field BBB scoring system. Survival rate of the graft cells was observed at second and eighth weeks after the transplantation. We also detected the myelin sheath fibers around the lesions and the size of the axotomized red nucleus. A one-way ANOVA was used to compare the means among the groups. The significance level was set at P〈0.05. Results The graft HAECs survived for a long time （8 weeks） and integrated into the host spinal cord without immune rejection. Compared with the control group, HAECs can promote the regeneration and sprouting of the axons, improve the hindlimb motor function of the rats （BBB score： cells-graft group 9.0 ± 0.89 vs PBS group 3.7± 1.03, P〈0.01）, and inhibit the atrophy of axotomized red nucleus [cells-graft group （526.47±148.42）μm^2 vs PBS group （473.69±164.73） μm^2, P〈0.01]. Conclusion Transplantation of HAECs can improve the hindlimb motor function of rats with spinal cord injury.

The back-streaming white-neutron beamline(Back-n)of the China Spallation Neutron Source is an essential neutronresearch platform built for the study of nuclear data,neutron physics,and neutron *** types of cross-sectional neutron-reaction measurements have been performed at Back-n since early *** measurements have shown that a significant number of gamma rays can be transmitted to the experimental stations of Back-n along with the neutron *** gamma rays,commonly referred to as in-beam gamma rays,can induce a non-negligible experimental background in neutron-reaction *** the characteristics of in-beam gamma rays is important for understanding the experimental ***,measuring in-beam gamma rays is challenging because most gamma-ray detectors are sensitive to neutrons;thus,discriminating between neutron-induced signals and those from in-beam gamma rays is *** this study,we propose the use of the black resonance filter method and a CeBr_(3) scintillation detector to measure the characteristics of the in-beam gamma rays of *** types of black resonance filters,^(181)Ta,^(59)Co,^(nat)Ag,and^(nat)Cd,were used in this *** time-of-flight(TOF)technique was used to select the detector signals remaining in the absorption region of the TOF spectra,which were mainly induced by in-beam gamma *** energy distribution and flux of the in-beam gamma rays of Back-n were determined by analyzing the deposited energy spectra of the CeBr_(3) scintillation detector and using Monte Carlo *** on the results of this study,the background contributions from in-beam gamma rays in neutron-reaction measurements at Back-n can be reasonably evaluated,which is beneficial for enhancing both the experimental methodology and data analysis.

Traumatic brain injury （TBI）, which is an important reason of human mortality and morbidity in industrialized countries, still cannot be treated effectively. Since the self-repair capacity of brain is limited, cellular transplantation in TBI may be a therapeutic *** amniotic cells （HACs） from fertilized ovum are able to differentiate into all the tissues of the body. They can not only express the markers of neuronal and glial cells^1 but also synthesize and release the neurotrophic factors.^2 Moreover, transplanted HACs rarely induce immunologic rejection.^3,4 In this study, we investigated whether HAC cultures conditioned by traumatic brain tissue extracts can acquire the phenotype of neuron, and examined the effects of transplanted HACs on the affected hindlimb of rats with TBI.
