A Survey on Methods for Predicting Polyadenylation Sites from DNA Sequences,Bulk RNA-seq,and Single-cell RNA-seq

Alternative polyadenylation(APA)plays important roles in modulating mRNA stability,translation,and subcellular localization,and contributes extensively to shaping eukaryotic transcriptome complexity and proteome diversity.Identification of poly(A)sites(pAs)on a genomewide scale is a critical step toward understanding the underlying mechanism of APA-mediated gene regulation.A number of established computational tools have been proposed to predict pAs from diverse genomic data.Here we provided an exhaustive overview of computational approaches for predicting pAs from DNA sequences,bulk RNA sequencing(RNA-seq)data,and single-cell RNA sequencing(scRNA-seq)data.Particularly,we examined several representative tools using bulk RNA-seq and scRNA-seq data from peripheral blood mononuclear cells and put forward operable suggestions on how to assess the reliability of pAs predicted by different tools.We also proposed practical guidelines on choosing appropriate methods applicable to diverse scenarios.Moreover,we discussed in depth the challenges in improving the performance of pA prediction and benchmarking different methods.Additionally,we highlighted outstanding challenges and opportunities using new machine learning and integrative multi-omics techniques,and provided our perspective on how computational methodologies might evolve in the future for non-30 untranslated region,tissuespecific,cross-species,and single-cell pA prediction.

Precipitation Behavior and Mechanical Properties of a 16Cr-25Ni Superaustenitic Stainless Steel Weld Metal During Post-weld Heat Treatment

A 16Cr-25Ni superaustenitic stainless steel weld metal for austenitic stainless steel/ferrite heat-resistance steel dissimilar metal weld was designed and prepared through tungsten inert-gas welding.The precipitate evolution and its correlation with mechanical properties were investigated during post-weld heat treatment(PWHT)at 690℃ for up to 12 h.The primary precipitates in the as-welded weld metal were identified as Mo-rich M6C carbides in the interdendritic region and semicontinuous fine-sized M23C6 carbides along grain boundary.After PWHT,three types of precipitates coexisted in the interdendritic region:primary M6C carbides,newly precipitated Mo-rich M2X carbonitrides and some of the secondary M23C6 carbides.Additionally,mass secondary M23C6 carbides formed and coarsened along grain boundary.No undesirable intermetallic phases formed during the whole period.The M2X and interdendritic M23C6 improved the strength of the weld metal after PWHT,but the elongation and impact toughness degraded,which were mainly owing to the intergranular M23C6 carbides that changed the fracture mode from ductile transgranular mode to mixed mode of transgranular and intergranular fracture.Meanwhile,the coarsening of M2X carbonitrides may lead to the elongation loss during 8 h to 12 h.Evolution of impact toughness was also related to the M2X carbonitrides,which made the crack easier to propagate compared with austenitic matrix and contributed to the decline of impact toughness.However,due to the sluggish precipitation of M2X carbonitrides with longer holding time,the decreasing trend became slow from 4 to 12 h.The results showed that PWHT should be controlled within 8 h to obtain better combination of strength and ductility.

Self‑Assembly MXene‑rGO/CoNi Film with Massive Continuous Heterointerfaces and Enhanced Magnetic Coupling for Superior Microwave Absorber

MXene, as a rising star of two-dimensional(2 D) materials, has been widely applied in fields of microwave absorption and electromagnetic shielding to cope with the arrival of the 5 G era. However, challenges arise due to the excessively high permittivity and the difficulty of surface modification of few-layered MXenes severely, which infect the microwave absorption performance. Herein, for the first time, a carefully designed and optimized electrostatic selfassembly strategy to fabricate magnetized MXene-r GO/Co Ni film was reported. Inside the synthesized composite film, r GO nanosheets decorated with highly dispersed Co Ni nanoparticles are interclacted into MXene layers, which effectively suppresses the originally self-restacked of MXene nanosheets, resulting in a reduction of high permittivity. In addition, owing to the strong magnetic coupling between the magnetic Fe Co alloy nanoparticles on the r GO substrate, the entire MXener GO/Co Ni film exhibits a strong magnetic loss capability. Moreover, the local dielectric polarized fields exist at the continuous heterointerfaces between 2 D MXene and r GO further improve the capacity of microwave loss. Hence, the synthesized composite film exhibits excellent microwave absorption property with a maximum reflection loss value of-54.1 d B at 13.28 GHz. The electromagnetic synergy strategy is expected to guide future exploration of high-efficiency MXene-based microwave absorption materials.

Geological factors controlling deep geothermal anomalies in the Qianjiaying Mine,China

Here,the geological factors controlling deep geothermal anomalies in mines were studied based on the geotemperature,lithologic thermal conductivity,and related geological data collected from the Qianjiaying Mine,China.A simulation of the change in magma waste heat,conducted using the ANSYS Workbench,revealed the distribution characteristics of geothermal anomalies in this mine and the corresponding geological control factors.The results revealed the following points.(1)First-degree heat hazard level(temperature=31-37℃)occurred in the central and southwestern parts of the mine at an^600-m depth,while second-degree heat hazard level(temperature≥37℃)occurred at an^800-m depth.The geotemperature and geothermal gradient in the southwestern part of the mine were anomalously high.(2)The geotemperatures measured in the mine generally reflected a standard increase with depth,while the geothermal gradient remained unchanged with depth.The geothermal gradient and its average value in the study area were 0.70-4.23 and 2.12C·hm^-1,respectively.(3)A combination of stratum characteristics,geological structure,and groundwater characteristics led to geothermal anomalies in mines;additionally,the waste heat from magma had no significant effect on the geothermal field.

Fractionation mechanism of iron isotopes in highly fractionated granites from the Xinxian Pluton,Western Dabie Orogen,Central China

Iron isotopes are important for tracing the magmatic process.The fractionation of iron isotopes in granite is up to 0.55‰.In this study,Wangjiagou(XWJ)granite and Tayueping(XTY)granite in the Xinxian pluton of the Western Dabie orogen were evaluated.Both the XTY and XWJ granite belong to monzogranites,with high SiO2(74.42-76.82 wt.%)contents.The granites are depleted of Nb and Ti but enriched with Pb and K,and they display negative Eu anomalies(Eu/Eu^(*)=0.40-0.52)on REE plots that are normalized by chondrite.Theδ^(56)Fe values of the XTY granites vary from 0.19±0.03‰to0.27±0.04‰,and theδ^(56)Fe values of the XWJ granites are 0.34±0.02‰and 0.36±0.01‰,respectively.Both the XTY and the XWJ granites belong to highly fractionated granites due to their SI(solidification index),DI(differentiation index),and content of CaO.Evidence from the iron isotopes shows that neither fluid exsolution,alteration,weathering,nor partial melting can explain the enrichment of the heavy iron isotopes.The results modeled using the Rayleigh equation showed that fractional crystallization can produceΔ^(56)Femelt-crystalwith the value of0.08-0.15‰.In conclusion,fractional crystallization was the main factor controlling the fractionation of iron isotopes,and the change of melt composition may also lead to the enrichment of heavy iron isotopes in the residual melt.

Meso-mechanical model of concrete under a penetration load

The influence of concrete components on projectile penetration is significant.To study the relationship between the equivalent mechanical properties and components of concrete under a penetration load,concrete is simplified as a two-phase composite of coarse aggregate and mortar,and a meso-mechanical model is established,including the equivalent equation of state model,the equivalent confining pressure strength model and the equivalent dynamic tensile strength model,considering shear stress,large deformation and pore compression.Tests of the mechanical properties of mortar,concrete and limestone were conducted;the results show that the equivalent mechanical properties of concrete calculated by the meso-mechanical model are consistent with the test results,and the equivalent mechanical properties of concrete with different volume fractions of coarse aggregate are obtained.Meso-scale and macro-scale numerical simulations of a projectile penetrating into concrete are carried out,the penetration depths obtained by meso-scale and macro-scale numerical simulations are consistent for different volume fractions of coarse aggregate and different velocities of the projectile,which verifies the rationality of the meso-mechanical model.

Characteristics of short double stranded RNA against hepatitis C virus: a literature-based analysis

Objective: To describe the characteristics of short interfering double stranded RNA (dsRNA) against hepatitis C virus (HCV) and to find out the determining factors in design for desirable inhibitory efficacy. Methods: The data were collected and analyzed by retrieval of 229 published short dsRNAs designed for degradation of HCV RNA. Results: Statistical analyses showed that the most frequently involved short dsRNAs were directing against 5′NTR/core and genotype 1b, accounting for 64.2% and 69.9%, respectively. Inhibitory efficacy varied with the structural characteristics of short dsRNAs, of which the most potential were those directed against HCV core region with inhibitory efficacy of 70.2%. Moreover, the mean inhibitory efficacy of short dsRNAs with GC contents from 30% to 52% was higher than that of those with GC contents out of this range. Conclusion: Based on this pooled data in a relatively large sample, the present results provided clues to design for short dsRNAs with more potent inhibitory efficacy.

On an open problem of Guo-Skiba

Let G be a finite group, and let A be a proper subgroup of G. Then any chief factor H/AG of G is called a G-boundary factor of A. For any G- boundary factor H/AG of A, the subgroup （A ∩ H）/AG of G/AG is called a G-trace of A. In this paper, we prove that G is p-soluble if and only if every maximal chain of G of length 2 contains a proper subgroup M of G such that either some G-trace of M is subnormal or every G-boundary factor of M is a p′- group. This result give a positive answer to a recent open problem of Guo and Skiba. We also give some new characterizations of p-hypercyclically embedded subgroups.

High adsorption selectivity of activated carbon and carbon molecular sieve boosting CO_(2)/N_(2) and CH_(4)/N_(2) separation

Flue gas and coal bed methane are two important sources of greenhouse gases.Pressure swing adsorption process has a wide range of application in the field of gas separation,and the selection of adsorbent is crucial.In this regard,in order to assess the better adsorbent for separating CO_(2) from flue gas and CH_(4) from coal bed methane,adsorption isotherms of CO_(2),CH_(4) and N_(2) on activated carbon and carbon molecular sieve are measured at 303.15,318.15 and 333.15 K,and up to 250 kPa.The experimental data fit better with Langmuir 2 compared to Langmuir 3 and Langmuir-Freundlich models,and Clausius-Clapeyron equation was used to calculate the isosteric heat.Both the order of the adsorbed amount and the adsorption heat on the two adsorbents are CO_(2)>CH_(4)>N_(2).The adsorption kinetics are calculated by the pseudo-first kinetic model,and the order of adsorption rates on activated carbon is N_(2)-CH_(4)>CO_(2),while on carbon molecular sieve,it is CO_(2)-N_(2)>CH_(4).It is shown that relative molecular mass and adsorption heat are the primary effect on kinetics for activated carbon,while kinetic diameter is the main resistance factor for carbon molecular sieve.Moreover,the adsorption selectivity of CH_(4)/N_(2) and CO_(2)/N_(2) were estimated with the ideal adsorption solution theory,and carbon molecular sieve performed best at 318.15 K for both CO_(2) and CH_(4) separation.The study suggested that activated carbon is a better choice for separating flue gas and carbon molecular sieve can be a strong candidate for separating coal bed methane.
