Berbamine selectively induces apoptosis of human acute promyelocytic leukemia cells via survivin-mediated pathway

Background Currently, resistance and relapse are still major problems in acute promyelocytic leukemia （APL） cases. Thus, new agents that override the resistance are crucial to the development of curative therapies for APL. In this study, we investigated the effects of berbamine on the proliferation of APL cell line NB4 and its possible mechanisms. Methods NB4 cells were treated with berbamine at different concentrations （0-64 μg/ml） for 72 hours. MTT assay was used to determine proliferation inhibition of NB4 cells. Cell apoptosis was evaluated by both flow cytometry （FCM） and morphological examination. PML/RAR-α and survivin mRNAs were measured by nested-RT-PCR and RT-PCR, respectively. Activated-caspase 3 was determined by FCM. Results Berbamine greatly inhibited the proliferation of NB4 cells in dose- and time-dependent manners, and its IC50 value was 3.86 μg/ml at 48 hours. Both morphological observations and FCM results showed that berbamine induced apoptosis of NB4 cells with concomitant increase of activated caspase-3 and decrease of survivin mRNA. After treatment with berbamine at 8 μg/ml for 48 hours, the percentage of apoptotic cells increased from 2.83% to 58.44% （P〈0.01）, and the percentage of cells with activated-caspase 3 elevated from 2.06% to 70.89% （P〈0.01）, whereas, level of survivin mRNA was reduced to 38.24% of control （P〈0.01）. However, no significant change was observed in PML/RAR-α mRNA.Conclusions Berbamine induces caspase-3-dependent apoptosis of leukemia NB4 cells via survivin-mediated pathway, suggesting that berbamine may be a novel potential agent against APL with a mechanism distinct from that of all-trans retinoic acid （ATRA） and arsenic trioxide （ATO）.

Investigation on Steelmaking with Hot Metal Containing Low Silicon Content in Large Converter

There are some problems in steelmaking with hot metal containing low silicon content such as difficulty in slag formation, less slag for dephosphorization and slag adhesion on oxygen lance and hood. To overcome these problems, experiments wcrc conducted and some improvements were obtained, such as adding appropriate flux, increasing the lance position slightly during steelmaking and using effective multi-outlet nozzle. Moreover, to keep normal heating rate, the ore and scrap charge should be reduced due to less chemical heat input in steelmaking.

Separation of phosphatidylcholine from soybean phospholipids by simulated moving bed

A simulated moving bed (SMB), equipped with eight silica-gel columns, was used to separate phosphatidylcholine (PC) from soybean phospholipids. The effects of flow rate in Sections 2 (Q2) and 3 (Q3), switching time, feed flow rate and feed concentration on the operating performance parameters: purity, recovery, productivity and desorbent consumption were studied. Operating conditions leading to more than 90% purity in both outlet streams have been identified, together with those achieving optimal performance. Regions leading to complete separation are observed and explained theoretically. As the mass-transfer effect was not considered, the triangle theory only gives initial guesses for the optimal operating conditions.

Tight sandstone gas(hereafter"tight gas")has become a subject of unconventional gas exploration *** large-scale development and use of tight gas resources in the USA,in particular,facilitated the rapid rebound of natural gas production in the USA,in addition to driving the rapid development of tight gas *** the eastern Ordos Basin,the Upper Paleozoic feature includes multiple layers of gas,a shallow depth,and notable potential for exploration and ***,the reservoirs in the area are relatively tight,exhibit strong heterogeneity,and possess a complex micropore structure,thus restricting the eff ective economic development of oil and ***,research on the primary parameters controlling pore throat structure and the seepage capability of low-permeability reservoirs will be beneficial for the effcient exploration and development of natural gas in the eastern Ordos *** parameters of reservoir porosity and percolation ability,as well as permeability,were analyzed using systematic sampling of the of the Upper Paleozoic Benxi,Taiyuan,and Shanxi Formations in the eastern Ordos Basin,constant-rate mercury injection experiments,nuclear magnetic resonance analysis,and gas–water-phase experimental *** results indicate that reservoir porosity is controlled by the effective pore volume and number,whereas permeability is controlled by the largest throat radius,rather than the *** effective pore volume controls the movable fluid saturation,while reservoir percolation capability is controlled by the effective pore volume,irreducible water saturation,and size of the gas–water two-phase seepage zone.

针对某SUV车型低频路噪轰鸣声问题,实车道路排查测试发现备胎舱与车内轰鸣声存在强相关,并结合前期的仿真分析结果,识别出备胎舱是引起车内轰鸣声主要原因。通过充分利用截面几何力学特性指导结构优化,提高备胎舱刚度,使其模态频率与问题频率避频,达到改善驾乘人员的乘坐舒适度。同时也为后续车型开发过程中提供借鉴,备胎舱结构设计开发,不仅要满足承重的性能指标要求,还要兼顾NVH性能指标,特别是在共平台车型开发过程中,此性能指标制定尤为重要,如果舒适性能指标制定合理,可以有效避免共用底盘件不同车型由于配置产生的其他NVH问题,也可以间接减少物理试验的投入,降低开发成本。

Selective photosynthesis of imines from biomass-derived aldehydes over Ni/TiO_(2)

Photocatalytic reductive amination of biomass-derived aldehydes is a desirable way to selectively upgrade biomass into value-added nitrogen-containing chemicals under mild ***,it is challenging to produce imines in high selectivity because of the undesirable side reactions caused by the activity of functional ***,we demonstrate the highly reactive and selective production of imines from biomass derived aldehydes via the photocatalytic reductive amination,using a defective TiO_(2)supported nickel *** employment of methanol as the hydrogen donor and ammonia solution as the nitrogen source avoids the use of high-pressure H_(2)and expensive amines,rendering the current catalytic process safe,economical,and environmentally *** depth investigations attribute the improved separation and transfer of photogenerated charge carriers to the presence of oxygen vacancies and decorated Ni nanoparticles,thereby accelerating the production of imines from benzaldehyde amination(conversion,95.8%;seleetivity,95.2%).Furthermore,the developed system could be easily translated to the photocatalytic conversions of various biomass derived aldehydes,which provided an example of a cost-effective and sustainable approach for the valorization of biomass derived feedstocks.

Serrated flow behavior of GH536 superalloy under different loading rates at room temperature

Serrated flow phenomenon has been widely observed among different metals due to the potential influence on their *** tensile tests of nickel-based superalloy GH536 were carried out at loading rates of 0.06,0.60,3.60,and 36.00 mm·min^(-1)at room temperature,*** tensile stress-strain curves demonstrate repetitive and discontinuous yielding behavior,namely serrated *** is observed that the stress-strain curves were dominated by type B serration,and original annealing twin boundaries(TBs)were distorted to various degrees under comparatively different lower tensile *** contrast,the TBs almost disappear,and the type B serrations become smoother and regular under the higher loading *** phenomenon can be attributed to the interactions between TBs and dislocations during tensile *** effects of the mobile and immobile dislocations illustrate the unsteady amplitude of serrations observed in stress-strain *** electron microscope images of tested pieces reveal the interaction of dislocation and TB,with dislocation tangling around the TBs.

Mixed D-vine copula-based conditional quantile model for stochastic monthly streamflow simulation

Copula functions have been widely used in stochastic simulation and prediction of ***,existing models are usually limited to single two-dimensional or three-dimensional copulas with the same bivariate block for all *** address this limitation,this study developed a mixed D-vine copula-based conditional quantile model that can capture temporal *** model can generate streamflow by selecting different historical streamflow variables as the conditions for different months and by exploiting the conditional quantile functions of streamflows in different months with mixed D-vine *** up-to-down sequential method,which couples the maximum weight approach with the Akaike information criteria and the maximum likelihood approach,was used to determine the structures of multivariate Dvine *** developed model was used in a case study to synthesize the monthly streamflow at the Tangnaihai hydrological station,the inflow control station of the Longyangxia Reservoir in the Yellow River *** results showed that the developed model outperformed the commonly used bivariate copula model in terms of the performance in simulating the seasonality and interannual variability of *** model provides useful information for water-related natural hazard risk assessment and integrated water resources management and utilization.

Microplastics(MPs)have become a significant concern for their potential ***,the correlation between the size of plastic particles and their toxicity remains ***,we investigate the toxic effects of different sizes(80 nm,800 nm,8μm and 80μm)polystyrene MPs(PS-MPs)on the model organism Nile tilapia(Oreochromis niloticus).The results of bioluminescent imaging indicate that the 80 nm PS-MPs are more likely to invade the body.H&E staining shows severe damage on the intestinal villi and distinct hepatic steatosis in the 80 nm *** U labeling shows that the proliferation activity of intestinal and liver cells reduces significantly in the 80 nm *** gut microbiome analysis shows a severe imbalance of gut microbiota homeostasis in the 80 nm *** analysis of liver transcriptomics and metabolomics shows that the liver lipid metabolism is disordered in the 80 nm *** conclusion,this study confirms that the 80 nm PS-MPs are more likely to induce intestinal and liver *** the above lay the foundation for further study on the pathological damage of MPs to other organisms.
