A PCA-based method for ancestral informative markers selection in structured populations

Identification of population structure can help trace population histories and identify disease genes. Structured association (SA) is a commonly used approach for population structure identification and association mapping. A major issue with SA is that its performance greatly depends on the informa-tiveness and the numbers of ancestral informative markers (AIMs). Present major AIM selection meth-ods mostly require prior individual ancestry information, which is usually not available or uncertain in practice. To address this potential weakness, we herein develop a novel approach for AIM selection based on principle component analysis (PCA), which does not require prior ancestry information of study subjects. Our simulation and real genetic data analysis results suggest that, with equivalent AIMs, PCA-based selected AIMs can significantly increase the accuracy of inferred individual ancestries compared with traditionally randomly selected AIMs. Our method can easily be applied to whole genome data to select a set of highly informative AIMs in population structure, which can then be used to identify potential population structure and correct possible statistical biases caused by population stratification.

Objective To investigate the effects of acitretin on T helper cell(Th)1/Th2.balance and Th17 cells in psoriasis vulgaris(PV)*** A total of 13 men and 17 women with PV were investigated.10 mg of acitretin was administered twice a day for 8 weeks for intervention *** levels of interferon-gamma(IFN-γ),interleukin(IL)-4 and IL-17 were measured by enzyme-linked immunosorbent assay.T,Th1,Th2.and Th17 cells in skin biopsies were counted with double-labeled *** Area and Severity Index(PASI)score was calculated before and 8 weeks after *** Before treatment PV patients had higher serum levels of IFN-γ and IL-17,and increased T,Th1 and Th17 cells in skin *** treatment,both serum levels of IFN-γ and IL-17,and T,Th1 and Th17 cells infiltrating in PV skin decreased ***1/Th2.balance was restored to ***,their IL-4 and Th2.cells showed no significant change throughout the *** Acitretin exerts influence on dermal Th1/Th2.balance and Th17 cell infiltration,so does it on production of systematic inflammatory cytokines IFN-γ and IL-17 in PV ***,Th2.cells and its derivative cytokine—IL-4 are not affected.

Electro-optical effect-based liquid crystal devices have been extensively used in optical modulation techniques, in which the Kerr coefficient reflects the sensitivity of the liquid crystals and determines the strength of the device's operational electric field. The Peterlin–Stuart theory and the O'Konski model jointly indicate that a giant Kerr coefficient could be obtained in a material with both a large geometrical anisotropy and an intrinsic polarization, but such a material is not yet reported. Here we reveal a ferroelectric effect in a monolayer two-dimensional mineral vermiculite. A large geometrical anisotropy factor and a large inherent electric dipole together raise the record value of Kerr coefficient by an order of magnitude, till 3.0 × 10-4mV-2. This finding enables an ultra-low operational electric field of10～2.10～4V m-1and the fabrication of electro-optical devices with an inch-level electrode separation, which has not previously been practical. Because of its high ultraviolet stability（decay 1% under ultraviolet exposure for 1000 hours）, large-scale production, and energy efficiency, prototypical displayable billboards have been fabricated for outdoor interactive scenes. This work provides new insights for both liquid crystal optics and two-dimensional ferroelectrics.

Objective To observe the expression of hypoxia inducible factor-1α (HIF-1α) in the retina of rabbits with acute high intraocular pressure and to investigate the mechanism of systemic domestic recombinant human erythropoietin (rhEPO) protecting the retina from ischemia-reperfusion injury. Methods First,control group and model group were established in rabbit eyes. The acute high intraocular pressure model was established by saline perfusion into anterior chamber,and then hypodermic injection of domestic rhEPO was made. HIF-1α protein in the retina was observed by immunohistochemical staining method on days 1,3,7 and 14 after retinal ischemia-reperfusion,respectively. Results No cells with HIF-1α positive expression were observed in the retina of the control group. Cells with HIF-1α positive expression in the model group outnumbered those in the control group (P1). The resemblance pattern occurred in EPO group but its degree was slightly greater than that in the model group from day 3 after ischemia-reperfusion (P1α in the retina with acute high intraocular pressure,which may be one of the mechanisms that rhEPO protects the retina from ischemia-reperfusion injury.

Application of Factor Difference Scheme to Solving Discrete Flow Equations Based on Unstructured Grid

A second-order mixing difference scheme with a limiting factor is deduced with the reconstruction gradient method and applied to discretizing the Navier-Stokes equation in an unstructured *** transform of nonorthogonal diffusion items generated by the scheme in discrete equations is *** Delaunay triangulation method is improved to generate the unstructured *** computing program based on the SIMPLE algorithm in an unstructured grid is compiled and used to solve the discrete equations of two types of incompressible viscous *** numerical simulation results of the laminar flow driven by lid in cavity and flow behind a cylinder are compared with the theoretical solution and experimental data *** the former case,a good agreement is achieved in the main velocity and drag coefficient *** the latter case,the numerical structure and development of vortex under several Reynolds numbers match well with that of the *** is indicated that the factor difference scheme is of higher accuracy,and feasible to be applied to Navier-Stokes equation.

Supramolecular Macrocycle Regulated Single-Atom MoS2.Co Catalysts for Enhanced Oxygen Evolution Reaction

The development of active water oxidation catalysts for water splitting has stimulated considerable interest. Herein, the design and building of single atom Co sites using a supramolecular tailoring strategy are reported, that is,the introduction of pillar[4]arene[1]quinone （P4A1Q） permits mononuclear Co species stereoelectronically assembled on MoS2.atrix to construct an atomically dispersed MoS2.Co catalyst with modulated local electronic structure, definite chemical environment and enhanced oxygen evolution reaction performance. Theoretical calculations indicate that immsobilized single-Co sites exhibit an optimized adsorption capability of oxygencontaining intermediates, endowing the catalyst an excellent electrocatalytic oxygen evolution reaction activity, with a low overpotential of 370 mV at10 mA cm-2.nd a small Tafel slope of 90 mV dec-1. The extendable potential of this strategy to other electrocatalysts such as MoS2.Ni and MoS2.Zn, and other applications such as the hydrogen evolution reaction was also demonstrated. This study affords new insights into the rational design of single metal atom systems with enhanced electrocatalytic performance.

The low-abundantly expressed genes composed the majorities of the mRNAs expressed in the central nervous system (CNS), and were thought to be important for the normal brain functions. Through differential screening a low-abundance cDNA sublibrary with mRNA from neuropathic pain of chronic constriction injury (CCI) model, we have identified a novel rat gene, rat spinal-cord expression protein 4 gene (RSEP4). The total length of RSEP4 cDNA is 2.06 bp, with a 501 nucleotide open reading frame (ORF) that encodes a 167 amino acid polypeptide. Northern blot revealed that RSEP4 was expressed specifically in the CNS. In situ hybridization showed that the mRNA of RSEP4 was strongly expressed in the CA1, CA2. CA3 and DG regions of hippocampus, the Purkinje cells of cerebellum, and the small sensory neurons of dorsal horn and large motor neurons of ventral horn of spinal cord. Over-expression of RSEP4-EGFP fusion protein in the human embryonic kidney 2.3T cells showed that RSEP4 protein was mainly localized in the cell cytoplasm. These results suggest that RSEP4 may play some roles in the CNS.
