OBJECTIVE: To reveal the potential underlying mechanism of the Shenqi Tiaoshen formula(参芪调肾方, SQTS) in the treatment of chronic obstructive pulmonary disease(COPD) by utilizing network pharmacology, molecular docking, and experimental verification. METHODS: Multiple open-source databases and research related to Traditional Chinese Medicine or compounds were employed to screen active ingredients and corresponding potential targets of the SQTS. The protein-protein interaction network screened hub genes, the relevant molecular mechanism and gene regulation were initially identified through the Kyoto Encyclopedia of Genes and Genomes(KEGG) and Gene Ontology(GO) analysis, and molecular docking was used to confirm further the interaction of the main components bound to the core targets. In vivo experiments on the COPD combined Qi-deficiency syndrome rat model were performed to verify the intervention effects and predicted potential molecular mechanisms of the SQTS. RESULTS: This study selected 156 active compounds and 326 candidate targets for treating COPD. Quercetin, Nobiletin, Kaempferol, Luteolin, Ginsenoside Rh2 and Formononetin were probably the main active compounds of SQTS in COPD treatment as they affected the most COPD-related targets, and interleukin-1(IL-6), signal transducing activator of transcription 3(STAT3), matrix metalloproteinase-9(MMP9), vascular endothelial growth factor A(VEGFA), protein kinase B(AKT1), hypoxiainducible factor-1α(HIF-1α), and forkhead box O3(FoxO3) were identified as the hub genes associated with its therapeutic effect. KEGG analysis was mainly enriched in the signaling pathways closely related to inflammation, immunity and oxidative stress, such as HIF-1, tumor necrosis factor(TNF), phosphatidylinositol 3 kinase(PI3K)-AKT, FoxO, apoptosis, IL-17, and toll-like receptor. Molecular docking confirmed that the main active components shared a good affinity with the hub genes. Background Despite its prevalence, there is ongoing debate regarding the optimal management strategy for chronic subdural hematoma (CSDH), reflecting the variability in clinical presentation and treatment outcomes. This ambidirectional, nationwide, multicenter registry study aims to assess the efficacy and safety of multimodality treatment approaches for CSDH in the Chinese population.Methods/design A multicenter cohort of CSDH patients from 59 participating hospitals in mainland China was enrolled in this study. The treatment modalities encompassed a range of options and baseline demographics, clinical characteristics, radiographic findings, and surgical techniques were documented. Clinical outcomes, including hematoma resolution, recurrence rates, neurological status, and complications, were assessed at regular intervals during treatment, 3 months, 6 months, 1 year, and 2 years follow-up.Result Between March 2022 and August 2023, a comprehensive cohort comprising 2173 individuals who met the criterion was assembled across 59 participating clinical sites. Of those patients, 81.1% were male, exhibiting an average age of 70.12 ± 14.53 years. A historical record of trauma was documented in 48.0% of cases, while headache constituted the predominant clinical presentation in 58.1% of patients. The foremost surgical modality employed was the burr hole (61.3%), with conservative management accounting for 25.6% of cases. Notably, a favorable clinical prognosis was observed in 88.9% of CSDH patients at 3 months, and the recurrence rate was found to be 2.4%.Conclusion This registry study provides critical insights into the multimodality treatment of CSDH in China, offering a foundation for advancing clinical practices, optimizing patient management, and ultimately, improving the quality of life for individuals suffering from this challenging neurosurgical condition.Trial registration ChiCTR2200057179

This study aims to improve the performances of the high-pressure water descaling technology used in steel hot rolling processes.In particular,a 2050 mm hot rolling line is considered,and the problem is investigated by means of a fluid–structure interaction(FSI)method by which the descaling effect produced by rolling coils with different section sizes is examined.Assuming a flat fan-shaped nozzle at the entrance of the R1R2 roughing mill,the outflow field characteristics and the velocity distribution curve on the strike line(at a target distance of 30–120 mm)are determined.It is found that the velocity in the center region of the water jet with different target distances is higher than that in the boundary region.As the target distance increases,the velocity of the water jet in the central region decreases.Through comparison with experimental results,it is shown that the simulation model can accurately predict the impact position of the high-pressure water on the impact plate,thereby providing a computational scheme that can be used to optimize the nozzle space layout and improve the slabs’descent effect for different rolling specifications.

dichloromethane(DCM)has been listed as a toxic and harmful water pollutant,and its re moval needs attention.Microbial electrolysis cells(MECs)are viewed as a promising alterna tive for pollutant removal,which can be strengthened from two aspects:microbial inocula tion and acclimation.In this study,the MEC for DCM degradation was inoculated with the ac tive sludge enhanced by Methylobacterium rhodesianum H13(strain H13)and then acclimated in the form of a microbial fuel cell(MFC).Both the introduction of strain H13 and the initi ation in MFC form significantly promoted DCM degradation.The degradation kinetics were fitted by the Haldane model,with V_(max),K_(h),K_(i)and v_(max)values of 103.2 mg/L/hr,97.8 mg/L268.3 mg/L and 44.7 mg/L/hr/cm^(2),respectively.The cyclic voltammogram implies that DCM redox reactions became easier with the setup of MEC,and the electrochemical impedance spectrogram shows that the acclimated and enriched microbes reduced the charge transfe resistance from the electrode to the electrolyte.In the biofilm,the dominant genera shifted from Geobacter to Hyphomicrobium in acclimation stages.Moreover,Methylobacterium played an increasingly important role.DCM metabolism mainly occurred through the hydrolytic glutathione S-transferase pathway,given that the gene dcmA was identified rather than the dhlA and P450/MO.The exogenous electrons facilitated the reduction of GSSG,directly o indirectly accelerating the GSH-catalyzed dehalogenation.This study provides support fo the construction of an efficient and stable MEC for DCM removal in water environment.

Registration system reform and initial public offering ownership preference:Evidence from China

Whether registration system reform(RSR)can curb administrative intervention and create a fair market environment has long been a concern in China.We explore this issue from the perspective of the initial public offering(IPO)preference based on the entropy balancing method,and findings are as follows.First,an IPO ownership preference exists under approval system.That is,state-owned enterprises(SOEs)are more likely than private enterprises to obtain IPO approval.However,RSR significantly changes this preference,especially for non-politically connected private enterprises.Second,the post-IPO market performance of SOEs is inferior to that of private enterprises under approval system,thus excluding the view that approval is prioritized for SOEs because of superior performance.Third,compared with SOEs,private enterprises are more likely to switch the issuance system from approval system to registration system;this change makes it easier for them to obtain IPO approval,indicating that private enterprise owners perceive the registration channel to be fairer.Fourth,the effect of RSR on IPO ownership preference mainly occurs in companies in three major urban agglomerations in China—the Pearl River Delta,the Yangtze River Delta,and the Beijing–Tian jin–Hebei region—and in technology-intensive industries.Collectively,our findings reveal that RSR cultivates a fairer IPO approval process.

To further improve the performance of passive intelligent reflecting surface(IRS)-assisted communication systems and mitigate the serious path loss due to “double-fading ” of IRS-assisted links, an active IRS-aided simultaneous wireless information and power transfer(SWIPT) system is investigated. This paper jointly optimizes the transmit beamforming at the base station(BS) and the phase shifts at the active IRS in order to maximize the power collected by the energy harvesting receiver under both perfect and imperfect channel state information(CSI)states, subject to the signal-to-interference-noise ratio constraint of the information decoding receiver, and the power constraints of the BS/IRS. Under perfect CSI, the alternating optimization algorithm is utilized for obtaining the transmit beamforming at the BS and the phase shifts at the active IRS. For each subproblem, we first transform nonconvex objective function and constraints into convex ones by performing a first-order Taylor expansion. Then, each subproblem is solved by using the interior point method. Given that obtaining perfect CSI is impractical, two robust beamforming designs are proposed for imperfect CSI case. Under the bounded CSI error model, we first transform the non-convex optimization problem into two semidefinite programming subproblems, and then solve each subproblem based on S-procedure and sequential rank-one constraint relaxation(SROCR) techniques. Under the stochastic CSI error model, the alternating optimization method is applied in an iterative manner based on Bernstein-type inequality and SROCR technique. Simulation results show that both robust and non-robust schemes for active IRS-assisted SWIPT systems can achieve extremely superior performance over conventional passive IRS-assisted systems under the same overall power budget.

The highly conserved target of rapamycin(TOR)pathway plays an important role in aging across species.Previous studies have established that inhibition of the TOR complex 1(TORC1)significantly extends lifespan in Caenorhabditis elegans.However,it has not been clear whether TORC1 perturbation affects aging in a spatiotemporal manner.Here,we apply the auxin-inducible degradation tool to knock down endogenous DAF-15,the C.elegans ortholog of regulatory associated protein of TOR(Raptor),to characterize its roles in aging.Global or tissue-specific inhibition of DAF-15 during development results in various growth defects,whereas neuron-specific knockdown of DAF-15 during adulthood significantly extends lifespan and healthspan.The neuronal DAF-15 deficiency-induced longevity requires the intestinal activities of DAF-16/FOXO and PHA-4/FOXA transcription factors,as well as the AAK-2/AMP-activated protein kinaseαcatalytic subunit.Transcriptome profiling reveals that the neuronal DAF-15 knockdown promotes the expression of genes involved in protection.These findings define the tissue-specific roles of TORC1 in healthy aging and highlight the importance of neuronal modulation of aging.

Chemokine platelet factor 4 accelerates peripheral nerve regeneration by regulating Schwann cell activation and axon elongation

Schwann cells in peripheral nerves react to traumatic nerve injury by attempting to grow and regenerate.Howeve r,it is unclear what factors play a role in this process.In this study,we searched a GEO database and found that expression of platelet factor 4 was markedly up-regulated after sciatic nerve injury.Platelet factor is an important molecule in cell apoptosis,diffe rentiation,survival,and proliferation.Further,polymerase chain reaction and immunohistochemical staining confirmed the change in platelet factor 4 in the sciatic nerve at different time points after injury.Enzyme-linked immunosorbent assay confirmed that platelet factor 4 was secreted by Schwann cells.We also found that silencing platelet factor 4 decreased the proliferation and migration of primary cultured Schwann cells,while exogenously applied platelet factor 4 stimulated Schwann cell prolife ration and migration and neuronal axon growth.Furthermore,knocking out platelet factor 4 inhibited the prolife ration of Schwann cells in injured rat sciatic nerve.These findings suggest that Schwann cell-secreted platelet factor 4 may facilitate peripheral nerve repair and regeneration by regulating Schwann cell activation and axon growth.Thus,platelet factor 4 may be a potential therapeutic target for traumatic peripheral nerve injury.

Porous heterogeneous lyophobic systems(HLSs)find potential applications in energy restoring,dissipating,and absorbing.However,the development of controllable HLSs still lacks rational structure design of nanoporous materials matching the molecular sizes of adopted liquids.Besides that,thoroughly understanding the underlying transportation mechanism in the confined nano channels is greatly challenging.In this work,a series of Co/Zn bimetallic zeolitic imidazolate frameworks(ZIFs)with tunable structures were synthesized via regulating the Co to Zn ratios and employed to investigate the intrusion–extrusion of liquid water in confined nanopores.Structural characterizations confirm the heterometallic coordination in the Co/Zn-doped frameworks.Water intrusion–extrusion experiments unlock the relationship between the intrusion pressure and the nanopore size and realize the evolution of the HLSs between molecular spring and shock-absorber.In addition,cycling tests indicate the reversible structure change of Co/Zn ZIFs encountering pressure-induced water intrusion.In combination with molecular dynamics simulations,we present that the water multimers intrude into nanopores of ZIFs in chain-like forms along with dissociation of hydrogen bonds(HBs).Water molecules in the pre-intrusion state exhibit reduced HBs in response to the increase of pressure and linear structure with 1.6–3.0 HBs on average.After transition to the post-intrusion situation,the associative configuration of water tends to exhibit the tetrahedral structure.Herein,we highlight the roles of pore size and HB in synergically dominating the pressure-induced intrusion–extrusion of liquid water in hydrophobic nanopores.Furthermore,the present work can also guide the development of functional guest–host systems based on porous architectures.
