Proteome profiling of spinal cord and dorsal root ganglia in rats with trinitrobenzene sulfonic acid-induced colitis

AIM: To investigate proteomic changes in spinal cord and dorsal root ganglia (DRG) of rats with trinitrobenzene sulfonic acid (TNBS)-induced colitis. METHODS: The colonic myeloperoxidase (MPO) activity and tumor necrosis factor-(TNF- ) level were determined. A two-dimensional electrophoresis (2-DE)-based proteomic technique was used to profile the global protein expression changes in the DRG and spinal cord of the rats with acute colitis induced by intracolonic injection of TNBS. RESULTS: TNBS group showed significantly elevated colonic MPO activity and increased TNF-level. The proteins derived from lumbosacral enlargement of the spinal cord and DRG were resolved by 2-DE; and 26 and 19 proteins that displayed significantly different expression levels in the DRG and spinal cord were identified respectively. Altered proteins were found to be involved in a number of biological functions, such as inflammation/immunity, cell signaling, redox regulation, sulfate transport and cellular metabolism. The over-expression of the protein similar to potassium channel tetramerisation domain containing protein 12 (Kctd 12) and low expression of proteasome subunit type-1 (psma) were validated by Western blotting analysis. CONCLUSION: TNBS-induced colitis has a profound impact on protein profiling in the nervous system. This result helps understand the neurological pathogenesis of inflammatory bowel disease.

Objective: To report the clinical maternal and fetal outcomes of pregnant women with coronavirus disease 2019 (COVID-19), along with any associated pregnancy complications, in Hong Kong, China, and to assess the impact of COVID-19 vaccination on these ***: This prospective registry-based observational study included pregnant women who were recruited through convenient sampling and had a laboratory-confirmed severe acute respiratory syndrome coronavirus 2 infection with a cycle threshold (Ct) value result available on admission to eight local hospitals in Hong Kong, China. Data on clinical symptoms, laboratory results, medical treatments, delivery timing and mode, and pregnancy complications were extracted from the Hospital Authority’s electronic medical record system. Maternal, fetal, and pregnancy outcomes were compared between unvaccinated pregnant women with COVID-19 and those who had received at least one dose of COVID-19 vaccine before diagnosis. Nonparametric continuous variables and categorical variables were analyzed using the Mann-WhitneyU test and the Pearson’s chi-squared test respectively. AP value less than 0.05 was considered statistically ***: A total of 164 pregnant women were included, of whom 78 (47.56%) were nulliparous. COVID-19 was diagnosed before 28 weeks’ gestation in 30 (18.29%), while 134 (81.71 %) were diagnosed at or after 28 weeks’ gestation. Sixty-two (37.80%) women received at least one dose of COVID-19 vaccine. There were no significant differences between vaccinated and unvaccinated groups in the time interval between COVID-19 diagnosis and delivery, the Ct value, and the gestational age at infection onset or delivery (P > 0.05). The majority of women were symptomatic at diagnosis regardless of vaccination status (55 (88.71%) in vaccinated groupvs. 78 (76.47%) in unvaccinated group (P = 0.052). 前期研究表明,肾髓质鞘氨醇1-磷酸受体1(sphingosine-1-phosphate receptor 1,S1PR1)介导钠排泄,高盐摄入可增加S1PR1水平,醋酸脱氧皮质酮(deoxycorticosterone acetate,DOCA)可阻断高盐诱导的肾髓质S1PR1,在集合管中敲除S1PR1可加重DOCA盐诱导的高血压。该研究验证在肾髓质中过表达S1PR1基因是否可减轻DOCA-盐小鼠模型的钠潴留和高血压。方法:给雄性C57BL/6J小鼠肾髓质转染对照或表达S1PR1的质粒后予DOCA-盐处理。

Aseismic Reliability Analysis Approach for Offshore Jacket Platform Structures

By means of nonlinear pushover collapse analysis approach, the aseismic reliability analyses of two offshore jacket platforms in the Bohai Gulf in China are studied according to their ocean location and environmental loadings there. On the basis of those analyses, an aseismic reliability analysis approach is presented. The results show that the aseismic reliability of those platforms is high. Also it is proved that this aseismic reliability analysis approach is simple, practical and reliable.

SYNTHESIS AND PROPERTIES OF SOME NEW POLYIMIDES CONTAINING BITHIAZOLE RINGS

Some new polyimides containing bithiazole rings were prepared by reacting 2,2'-diamino-4,4'-bisthiazole with different aromatic dianhydrides. The polyimides obtained had inherent viscosities of 0.37-0.82 dL/g. Thermogravimetric analysis of the polyimides showed good thermal stability, the temperature at 5% weight loss being from 450 degrees C to 560 degrees C. The permeability of two polymer membranes to H-2, O-2 and N-2 was determined.

Nucleation and Oriented Textured Growth of Diamond Films on Si(100) via Electron Emission in Hot Filament Chemical Vapor Deposition

Oriented textured diamond films were obtained on Si(100) substrate via electron emission in hot filament chemical vapor deposition (HFCVD). A de bias voltage relative to the filament was applied to the tungsten electrode between the substrate and the filament. The nucleation and subsequent growth of diamond films were characterized by scanning electron microscopy and Raman spectroscopy. The experimental results showed that the electron emission from the diamond coating on the electrode played a critical role during the nucleation. The maximum value of nucleation density was up to 10(11) cm(-2) on pristine Si surface at emission current of 250 mA. The effect of the electron emission on the reactive gas composition was analyzed by in situ infrared absorption, indicating that the concentration of CH3 and C2H2 near the substrate surface was extremely increased. This may be responsible for the enhanced nucleation by electron emission.
