Alterations of FHIT Gene and P16 Gene in Nickel Transformed Human Bronchial Epithelial Cells

Objective To study the alterations of FHIT gene and P16 gene in malignant transformed human bronchial epithelial cells induced by crystalline nickel sulfide using an immortal human bronchial epithelial cell line, and to explore the molecular mechanism of nickel carcinogenesis. Methods 16HBE cells were treated 6 times with different concentrations of NiS in vitro, and the degree of malignant transformation was determined by assaying the anchorage-independent growth and tumorigenicity. Malignant transformed cells and tumorigenic cells were examined for alterations of FHIT gene and P16 gene using RT-PCR, DNA sequencing, silver staining PCR-SSCP and Western blotting. Results NiS-treated cells exhibited overlapping growth. Compared wkh that of negative control cells, soft agar colony formation efficiency of NiS-treated cells showed significant increases （P〈0.01） and dose-dependent effects. NiS-treated cells could form tumors in nude mice, and a squamous cell carcinoma was confirmed by histopathological examination. No mutation of exon 2 and exons 2-3, no abnormal expression in pl6 gene and mutation of FHIT exons 5-8 and exons 1-4 or exons 5-9 were observed in transformed cells and tumorigenic cells. However, aberrant transcripts or loss of expression of the FHIT gene and Fhit protein was observed in transformed cells and tumorigenic cells. One of the aberrant transcripts in the FHIT gene was confirmed to have a deletion of exon 6, exon 7, exon 8, and an insertion of a 36 bp sequence replacing exon 6-8. Conclusions The FHIT gene rather than the P16 gene, plays a definite role in nickel carcinogenesis. Alterations of the FHIT gene induced by crystalline NiS may be a molecular event associated with carcinogen, chromosome fragile site instability and cell malignant transformation. FHIT may be an important target gene activated by nickel and other exotic carcinogens.

Moxibustion combined with acupuncture increases tight junction protein expression in Crohn's disease patients

A Facile Strategy for Intrinsic Low-Dk and Low-Df Polyimides Enabled by Spirobifluorene Groups

Modified polyimides(MPIs)show great potential towards 5G communication applications,due to its excellent thermal stability,mechanical property and chemical stability as compared to most of *** fluoride groups or porous structure is favorable to ultra-low dielectric constant(D_(k))and dielectric loss(D_(f)).However,the cost of the fluorinated MPIs is high and their synthetic processes are complicated,and porous MPIs suffer poor mechanical ***,increasing the fraction of free volume is a very effective way to lower D_(k)through introducing more ultra-low-D_(k)air ***,most of this kind of MPIs lag far behind the fluorinated MPIs and the porous MPIs in terms of ultra-low D_(f),hindering the application of MPIs in high-speed communication ***,it is highly desirable to develop intrinsic ultra-low-D_(k)/D_(f)MPIs at high frequency with less fluoric groups and nonporous ***,we introduce a facile and effective strategy to lower D_(k)and D_(f)through introducing rigid and large sterically hindered aromatic groups into *** the one hand,their large steric hindrance effect leads to low D_(k)by increasing intrinsic free *** the other hand,the resulting highly stiff polymer chain and strong intermolecular interaction are favorable to reduce D_(f)by inhibiting dipole *** on this strategy,the spirobifluorene groups are *** as-prepared MPIs show excellent dielectric performance with low D_(k)of 2.74–2.76 and low D_(f)of 0.00599 at 10 GHz,to some extent,exceeding the multiple fluorinated MPI with D_(k)/D_(f)of 2.67/0.00663 at 10 GHz.

Advanced intrahepatic cholangiocarcinoma(ICC)has a dismal ***,we report the efficacy and safety of combining toripalimab,lenvatinib,and gemcitabine plus oxaliplatin(GEMOX)as first-line therapy for advanced *** patients with pathologically confirmed advanced ICC received intravenous gemcitabine(1 g/m2)on Days 1 and 8 and oxaliplatin(85 mg/m2)Q3W for six cycles along with intravenous toripalimab(240 mg)Q3W and oral lenvatinib(8 mg)once daily for one *** expression of programmed death-ligand 1(PD-L1)and genetic status was investigated in paraffin-embedded tissues using immunohistochemistry and whole-exome sequencing(WES)*** primary endpoint was the objective response rate(ORR).Secondary outcomes included safety,overall survival(OS),progression-free survival(PFS),disease control rate(DCR)and duration of response(DoR).As of July 1,2022,the median follow-up time was 23.5 months,and the ORR was 80%.Twenty-three patients achieved partial response,and one achieved complete ***(21/30)with DNA damage response(DDR)-related gene mutations showed a higher ORR,while patients(14/30)with tumor area positivity≥1(PD-L1 staining)showed a trend of high ORR,but without significant *** median OS,PFS,and DoR were 22.5,10.2,and 11.0 months,*** DCR was 93.3%.Further,56.7%of patients experienced manageable grade≥3 adverse events(AEs),commonly neutropenia(40.0%)and leukocytopenia(23.3%).In conclusion,toripalimab plus lenvatinib and GEMOX are promising first-line regimens for the treatment of advanced ICC.A phase-III,multicenter,double-blinded,randomized study to validate our findings was approved by the National Medical Products Administration(NMPA,No.2021LP01825).

Co-transformation of HMs-PAHs in rhizosphere soils and adaptive responses of rhizobacteria during whole growth period of rice(Oryza sativa L.)

This study investigated the transformations of heavy metals(HMs) and polycyclic aromatic hydrocarbons(PAHs) in rhizosphere soils and adaptive responses of rhizobacterial community under the real field conditions during four growth stages(e.g., greening, tillering, heading, and maturity) of early rice(zhongjiazao 17) and late rice(zhongyou 9918) in jiangshe village(JSV) and Yangji village(YJV). Results showed that rhizosphere soils of YJV were mildly polluted by Cd and PAHs compared to that of JSV. The relative abundance of bioavailable Cd(bio-Cd) and bioavailable As(bio-As) in rhizosphere soil increased before the heading stage but decreased at the subsequent growth stage, but the content of ΣPAHs in rhizosphere soil decreased gradually during whole growth period. The dominant rhizobacteria genera at YJV(e.g., Bacillus, Massilia, Sphingomonas, and Geobacter) increased at an abundance level from the tillering to heading stage. Rhizobacteria interacted with the above co-pollutant more intensely at the tillering and heading stage, where genes involved in HM-resistance and PAH-degradation appeared to have a significant enhancement. The contents of bioCd and bio-As in rhizosphere soil of early rice were higher than that of late rice at each growth stage, especially at the heading stage. Bio-Cd, ΣPAHs, and organic matter were key factors influencing the community structure of rhizobacteria. Results of this study provide valuable insights about the interactions between HM-PAH co-pollutant and rhizobacterial community under real field conditions and thus develop in-situ rhizosphere remediation techniques for contaminated paddy fields.

Dark photons have been well motivated as strong candidates for dark force carriers and light dark matter in the sub-GeV mass *** with collider experiments,fixed-target experiments provide a complementary approach to searching for dark photons,particularly in the lower mass *** have studied the physics potential of the electron-on-target experiment based on the Shanghai SHINE facility,which provides 10 MHz single electron beam at 8 GeV *** analysis focuses on dark photons being produced via electron and nucleon interaction and then decays to dark matter candidates,which escape detection as missing momentum in the *** experiment takes advantage of using missing momentum to enhance signal versus background separation *** this study,signal samples as a function of dark photon mass and an inclusive background sample with 2.5 billion events are simulated with *** better background estimates,major rare background processes have also been *** paper presents the experiment and detector design,signal and background simulations,analysis strategy,and the prospective study of the experiment *** 9×10^(14) electron-on-target events(about three years running),this experiment is expected to rule out most of the sensitive regions predicted by popular dark photon models.

目的利用蛋白质组学技术识别结晶型硫化镍(NiS)诱发人支气管上皮细胞恶性转化相关蛋白的差异表达,探讨镍致癌的分子机制。方法应用二维凝胶电泳与相应软件分析结晶型NiS诱发细胞恶性转化后蛋白质组变化,基质辅助激光解吸电离-飞行时间-质谱(MALDITOFMS)结合数据库检索鉴定恶性转化相关蛋白。结果获得分辨率和重复性较好的二维凝胶电泳图谱,在相对分子质量14400～94000、等电点3～10范围内分离出约700个不同蛋白质点,对差异表达明显的等电点约6,相对分子质量约25000的蛋白质点进行质谱分析,鉴定出等电点为5.66,相对分子质量为21890的蛋白硫氧还原蛋白过氧化物酶2(Peroxiredoxin2,PDX2),并发现该蛋白在恶性转化细胞中高表达。结论蛋白PDX2可能是参与结晶型NiS诱发人支气管上皮细胞恶性转化过程的重要蛋白分子,与细胞恶性转化有关。
