Direct conversion of furfural to levulinic acid/ester in dimethoxymethane:Understanding the mechanism for polymerization

This study investigated the conversion of furfural to 5-hydroxymethylfurfural(HMF)and further to levulinic acid/ester in dimethoxymethane under acidic conditions,with the particular focus on understanding the mechanism for polymer *** results showed that furfural could react with dimethoxymethane via electrophilic substitution reaction to form HMF or the ether/acetal of HMF,which were further converted to levulinic acid and methyl *** polymerization of furfural and the cross-polymerization between dimethoxymethane and the levulinic acid/ester produced were the main side reactions leading to the decreased yields of levulinic acid/*** to the other solvent,methanol as the co-solvent helped to alleviate but not totally inhibited the occurrences of the polymerization,as the polymerization reactions via aldol condensation did not eliminate the C=O *** a consequence,the polymerization reactions continued to *** co-solvent used such as guaiacol,dimethyl sulfoxide and acetone interfered with the transformation of furfural to HMF or aided the polymerization *** polymer produced from the reactions between furfural and DMM was different from that produced from levulinic acid/*** former had a higher crystallinity,while the latter was more *** DRIFTS and TG-MS studies showed that the polymer had the carboxylic group,methyl group and the aliphatic structure in the *** removal of these functionalities was accompanied by the aromatization of the *** condensation of DMM with levulinic acid/ester was the key reason for the diminished production of levulinic acid/ester.

AIM:To investigate the anti-proliferation and apoptosisinducing effects of sodium aescinate(SA)on retinoblastoma Y79 cells and its ***:Y79 cells were cultured at different drug concentrations for different periods of time(24,48,and 72 h).The inhibitory effect of SA on proliferation of Y79 cells was detected by the cell counting kit-8(CCK-8)assay,and the morphology of Y79 cells in each group was observed under an inverted *** IC50 of 48 h was selected for subsequent *** pretreatment with SA for 24 and 48 h,cellular DNA distribution and apoptosis were detected by flow ***-time qunatitative polymerase chain reaction(RT-qPCR)and Western blot were used to assess changes in related genes(CDK1,CyclinB1,Bax,Bcl-2,caspase-9,caspase-8,and caspase-3).RESULTS:SA inhibited proliferation and induced apoptosis of Y79 cells in a time-dependent and concentrationdependent *** its intervention in the cell cycle pathway,SA can inhibit the expression of CDK1 and Cyclin B1 at the mRNA and protein levels,and block cells in the G2/M *** caspase-related apoptotic pathways,up-regulation of Bax and down-regulation of Bcl-2 caused caspase-9 to self-cleave and further activate ***’s more,the caspase-8-mediated extrinsic apoptosis pathway was activated,and the activated caspase-8 was released into the cytoplasm to activate caspase-3,which as a member of the downstream apoptotic effect group,initiates a caspase-cascade reaction that induces cell ***:SA inhibits the proliferation of Y79 cells by arresting the cell cycle at the G2/M phase,and induces apoptosis via the caspase-related apoptosis pathway,indicating that SA may have promising potential as a chemotherapeutic drug.

An interlayer is usually employed to tackle the interfacial instability issue between solid electrolytes(SEs)and li metal caused by the side ***,the failure mechanism of the ionic conductor interlayers,especially the influence from electron penetration,remains largely ***,using li1.3Al0.3Ti1.7(PO4)3(LATP)as the model SE and liF as the interlayer,we use metal semiconductor contact barrier theory to reveal the failure origin of li/liF@LATP interface based on the calculation results of density functional theory(DFT),in which electrons can easily tunnel through the liF grain boundary with F vacancies due to its narrow barrier width against electron injection,followed by the reduction of ***,an Al-liF bilayer between li/LATP is found to dramatically promote the interfacial stability,due to the highly increased barrier width and homogenized electric field at the ***,the li symmetric cells with Al-liF bilayer can exhibit excellent cyclability of more than 2,000 h superior to that interlayered by liF monolayer(~860 h).Moreover,the li/Al-liF@LATP/liFePO4 solid-state batteries deliver a capacity retention of 83.2%after 350 cycles at 0.5 *** findings emphasize the importance of tuning the electron transport behavior by optimizing the potential barrier for the interface design in high-performance solid-state batteries.

Calcium(Ca^(2+))plays an important role in determining plant growth and development because it maintains cell wall and membrane ***,understanding the role of Ca^(2+)in carbon and lipid metabolism could provide insights into the dynamic changes in cell membranes and cell walls during the rapid elongation of cotton *** the present study,we found that the lack of Ca^(2+)promoted fiber elongation and rapid ovule expansion,but it also caused tissue browning in the ovule culture ***-sequencing revealed that Ca^(2+)deficiency induced cells to be highly oxidized,and the expression of genes related to carbon metabolism and lipid metabolism was activated *** gene members of nine key enzymes involved in glycolysis were up-regulated,and glucose was significantly reduced in Ca^(2+)deficiency-treated ***^(2+)deficiency adjusted the flowing of glycolysis ***,low K^(+)recovered the expression levels of glycolysis genes and glucose content caused by Ca^(2+)*** ionizationtandem mass spectrometry technology was applied to uncover the dynamic profile of lipidome under Ca^(2+)and K^(+)interacted ***^(2+)deficiency led to the decrease of fatty acid(FA),diacylglycerol(DAG),glycolipid and the significant increase of triacylglycerol(TAG),phospholipid phosphatidylethanolamine(PE),phosphatidylglycerol(PG),and PC(phosphatidylcholine).Low K^(+)restored the contents of FA,phospholipids,and glycolipids,effectively relieved the symptoms caused by Ca^(2+)deficiency,and recovered the development of fiber *** study revealed dynamic changes in transcript and metabolic levels and uncovered the signaling interaction of Ca^(2+)deficiency and low K^(+)in glycolysis and lipid metabolism during fiber development.

Glioblastoma(GBM)is a lethal cancer with limited therapeutic *** cell(DC)-based cancer vaccines provide a promising approach for GBM *** studies suggest that other immunotherapeutic agents may be combined with DC vaccines to further enhance antitumor ***,we report a GBM case with combination immunotherapy consisting of DC vaccines,anti-programmed death-1(anti-PD-1)and poly I:C as well as the chemotherapeutic agent cyclophosphamide that was integrated with standard chemoradiation therapy,and the patient remained disease-free for 69 *** patient received DC vaccines loaded with multiple forms of tumor antigens,including mRNA-tumor associated antigens(TAA),mRNA-neoantigens,and hypochlorous acid(HOCl)-oxidized tumor ***,mRNA-TAAs were modified with a novel TriVac technology that fuses TAAs with a destabilization domain and inserts TAAs into full-length lysosomal associated membrane protein-1 to enhance major histocompatibility complex(MHC)class I and II antigen *** treatment consisted of 42 DC cancer vaccine infusions,26 anti-PD-1 antibody nivolumab administrations and 126 poly I:C injections for DC *** patient also received 28 doses of cyclophosphamide for depletion of regulatory T *** immunotherapy-related adverse events were observed during the *** antitumor CD4t and CD8t T-cell responses were *** patient remains free of disease *** is the first case report on the combination of the above three agents to treat glioblastoma *** results suggest that integrated combination immunotherapy is safe and feasible for long-term treatment in this patient.A large-scale trial to validate these findings is warranted.
