Levels of Crotonaldehyde and 4-hydroxy-(E)-2-nonenal and Expression of Genes Encoding Carbonyl-Scavenging Enzyme at Critical Node During Rice Seed Aging

The critical node(CN) is an important stage during seed aging, which is related to effective genebank conservation. Previous studies have demonstrated that proteins undergo carbonylated modification at the CN in rice, indicating oxidative damage. However, the levels of reactive carbonyl species(RCS) and the associated scavenging system at the CN are largely unknown. In this study, we optimized methods for the extraction and analysis of RCS from dry rice embryos. In order to acquire seeds at the CN, rice seeds were subjected to natural conditions for 7, 9, 11 and 13 months, and the seed germination rates were reduced to 90%, 82%, 71% and 57%, respectively. We chose the stage with seed germination rate of 82% as the CN according to the rice seed vigor loss curve. The levels of crotonaldehyde and 4-hydroxy-(E)-2-nonenal(HNE) were significantly increased at the CN. In addition, genes encoding carbonyl-scavenging enzyme, including Os ALDHs and Os AKRs, were significantly down-regulated at the CN, and reductions in the expression of Os ALDH2-2, Os ALDH2-5, Os ALDH3-4, Os ALDH7, Os AKR1 and Os AKR2 in particular could be responsible for RCS accumulation. Thus, the accumulations of crotonaldehyde and HNE and down-regulation of genes encoding carbonyl-scavenging enzyme might be related to an accelerating loss of seed viability at the CN.

Despite extensive studies on cultivated rice, the population structure and genetic diversity of the indica photoperiod-and thermo-sensitive genic male sterility(P/TGMS) lines in China remains unclear. Using 48 simple sequence repeat(SSR) markers, we genotyped a panel of 208 indica P/TGMS lines and confirmed three subgroups, named indica-I, indica-II and indica-III, in indica P/TGMS lines. Further diversity analysis indicated indica-II had the highest genetic diversity. The genetic differentiation between indica-II and indica-III was demonstrated as the largest among the three subgroups. Moreover, indica/japonica component identification was detected that five P/TGMS lines possess indica components less than 0.900. These results improve our knowledge on the genetic background for P/TGMS lines in China and will be beneficial for hybrid rice breeding programs.

Genetic diversity of rice cultivars (Oryza sativa L.) in China and the temporal trends in recent fifty years

To understand geographical distribution of the genetic diversity of rice cultivars {Oryza sativa L.) and its trends in recent fifty years in China, 453 ’ accessions were analyzed by 36 microsatellites loci and 42 phenotypic traits. Results revealed that the genetic diversity by SSRs is highly consistent with that by phenotypic traits and the genetic diversity of indica cultivars was higher than that of japonica cultivars; the genetic diversity of cultivars declined from 1950s to 1980s and then increased greatly; among the six rice ecological zones (REZs), genetic diversity of REZTl was the highest and those of REZV and REZVT were the lowest at both DNA and phenotypic level. Jiangsu and Jiangxi provinces in the middle and lower reaches of Yangtze River and Sichuan province in southwest of China were the areas with the highest genetic diversity. Breeders in REZV which is an important japonica rice area but with very low genetic diversity should explore more gene resources to widen the genetic backgrounds of cultivars.

During the logging work of the foreign cooperation mudlogging projects, the description of drillling cuttings (sample) required by the foreign companies is quite different from what we did at home. This paper is intended to give some ideas of description of sample at wellsite with the reference of the guidance of standards of several foreign companies and the working experiences of the author, also some problems that the geologists should pay attention to during the description of sample at wellsite.

Y faulted depression is located in the southeast of Songliao basin, which is another important area explored for natural gas in deep Formation after Xujiaweizi. But it is lowly explored and is not likely to make a major breakthrough in nature gas exploration for shallow and medium layers. The data on the geochemistry and geology indicate that the deep source rocks, especially the Jurassic mudstone has great potentials of natural gas generation. Based on this, the paper examines the conditions of natural gas generation and further points out the favorable prospective gas - bearing area. The results show that the amount of natural gas generated from the Jurassic mudstone in the Y faulted depression is 8. 42x1012m3 and the favorable gas - generating area is the body of the faulted depression.
