KNOX (knotted1-like homeobox, KNOX)是植物特有的一类具有同源异型结构域的转录因子，广泛参与植物的生长发育过程。为探究谷子(Setaria italica) KONXs基因的潜在功能，本研究采用生物信息学方法，在谷子全基因组水平对SiKNOX基因家族进行了鉴定和分析，并利用RT-qPCR检测其在不同组织中的表达量。结果显示，在谷子全基因组水平上一共鉴定到11个SiKNOXs基因，系统进化分析分为classⅠ和classⅡ两组，classⅠ包含5个SiKNOXs,classⅡ包含6个SiKNOXs。亚细胞定位预测发现所有SiKNOXs蛋白均定位在细胞核。染色体定位分析发现，11个SiKNOXs基因不均等地分布在谷子的7条染色体上。基因复制分析发现，有5个SiKNOXs基因组成了4个片段复制基因对。基因结构组成分析发现，所有的SiKNOXs由5～6个外显子组成；蛋白保守结构域分析结果显示，SiKNOXs的蛋白保守结构域由10个基序组成，其中motif 2、3、5、6分别构成了KNOX1、KNOX2、ELK、HD结构域。在SiKNOXs基因的启动子区域分布着许多与生物过程密切相关的顺式作用元件。SiKNOXs基因在谷子不同组织部位(包括根,茎,叶,穗,种子)均能检测到，且大部分SiKNOXs基因具有组织特异性表达特点。本研究为深入探讨KNOX基因家族调控谷子生长发育的功能提供了基础资料。

目的探讨使用ImageJ软件对HE染色结果进行质控评估及在定量评价中的应用。方法选择淋巴结质控蜡块以3μm厚度连续切片并连续HE染色5天,将染色的切片进行扫描后,使用ImageJ软件测量每张切片苏木素及伊红的染色强度,对测得的数据进行统计学分析。结果根据测量结果,苏木素染色强度值与染色天数呈正相关线性关系,R^(2)=0.8689,苏木素染色强度值可以较好的评估染液的状态,强度值越高提示苏木素染色效力越差,而伊红染色由于拟合度R^(2)过小,R^(2)=0.01924,通过强度值不适宜评估染液的状态。结论使用ImageJ软件可以对切片染色结果进行定量分析,可以客观的评价苏木素的染色结果及染液状态,为切片质量评估提供可靠的数据支撑。

The partial opening of the working gate for a discharge tunnel is used more and more widely in the various conditions, such as releasing discharge to meet the needs of the various stages under construction, or of the operations at low reservoir levels. In this article the hydraulic characteristics were investigated of the partial opening of the working gate based on the physical model experiments of the discharge tunnel for the Longtan Hydropower Project, including the discharge coefficient, the surface profile of the free flow section, the bottom pressure distribution of the tunnel with an aerator, the cavity length just from the ramp, and the wall pressure of the curve section of the outlet. Some new issues were discussed and the properties were presented of the partial opening of the gate.

Water-wings, induced by the middle-pier placed in the inlet of a discharge tunnel, have harmful effects on the operation of the discharge tunnel. Based on dimensional analysis and physical model tests, the hydraulic characteristics of water-wings were investigated and their causes were analyzed in this study. The results show that the height and length of the water-wings increased with the increase of three factors, i.e., the Froude number Frd of the outlet of the pressure section, the depth Hc of the water surface concave at the end of the middle-pier, and the impact length Li of the two flows after this end.

Adhesive forces exist between soil and the surfaces of soil-engaging components; they increase working resistance and energy consumption. This paper tries to find an approach to reduce the adhesion and resistance of bulldozing plate. A simplified mechanical model of adhesion and resistance between soil and a non-smooth bulldozing plate is proposed. The interaction force between moist soil and a non-smooth bulldozing plate is analyzed. The pressure and friction distribution on the bulldozing plate are computed, and the anti-adhesive effect of a corrugated bulldozing plate is simulated numerically. Numerical results show that the wavy bulldozing plate achieves an effective drag reduction in moist soil. The optimal wavy shape of the corrugated bulldozing plate with the minimal resistance is designed. The basic principle of reducing soil adhesion of the non-smooth surface is discovered.

Assembly and co-occurrence patterns of rare and abundant bacterial sub-communities in rice rhizosphere soil under short-term nitrogen deep placement

Nitrogen(N)deep placement has been found to reduce N leaching and increase N use efficiency in paddy ***,relatively little is known how bacterial consortia,especially abundant and rare taxa,respond to N deep placement,which is critical for understanding the biodiversity and function of agricultural *** this study,lllumina sequencing and ecological models were conducted to examine the diversity patterns and underlying assembly mechanisms of abundant and rare taxa in rice rhizosphere soil under different N fertilization regimes at four rice growth stages in paddy *** results showed that abundant and rare bacteria had distinct distribution patterns in rhizosphere *** bacteria showed ubiquitous distribution;while rare taxa exhibited uneven distribution across all *** processes dominated community assembly of both abundant and rare bacteria,with dispersal limitation playing a more vital role in abundant bacteria,and undominated processes playing a more important role in rare *** N deep placement was associated with a greater influence of dispersal limitation than the broadcast N fertilizer(BN)and no N fertilizer(NN)treatments in abundant and rare taxa of rhizosphere soil;while greater contributions from homogenizing dispersal were observed for BN and NN in rare *** analysis indicated that abundant taxa with closer relationships were usually more likely to occupy the central position of the network than rare ***,most of the keystone species were rare taxa and might have played essential roles in maintaining the network ***,these findings highlighted that the ecological mechanisms and co-occurrence patterns of abundant and rare bacteria in rhizosphere soil under N deep placement.
