目的探究下咽癌患者血清源性外泌体中角蛋白8(KRT8)小干扰RNA(si-KRT8)对人咽鳞状细胞癌FaDu细胞株5-氟脲嘧啶(5-FU)化疗敏感性的影响和作用机制。方法收集下咽癌患者肿瘤组织以及治疗后耐药患者的血清、肿瘤组织以及癌旁组织。构建FaDu的5-FU耐药细胞株(FaDu/R)用于后续实验。超速梯度离心法分离患者血清中的外泌体并利用透射电镜及蛋白质免疫印迹(Western blot,WB)技术进行鉴定,利用电转染技术将si-KRT8封装至外泌体(Exosome@si-KRT8)中,后续用于处理细胞。实时荧光定量PCR(RT-qPCR)及WB方法检测KRT8分别在不同组织、电转染后外泌体及不同处理条件下FaDu细胞中的表达水平。CCK-8、原位末端转移酶标记技术及迁移侵袭技术和WB检测Exosome@si-KRT8对FaDu/R细胞活力、凋亡及上皮间质转化(epithelial-mesenchymal transition,EMT)的影响。结果下咽癌耐药组织及FaDu/R细胞中的KRT8表达水平相较于癌旁组织和肿瘤组织以及正常FaDu细胞均升高(t=15.79,P<0.01)。分离的患者血清外泌体呈现双层膜结构,且CD63和TSG101蛋白均表达,电转染si-KRT8后,外泌体中KRT8表达量下降(t=6.70,P<0.01)。Exosome@si-KRT8能够抑制FaDu/R细胞中KRT8的蛋白表达水平(t=123.50,P<0.01)。与5-FU组及5-FU+Exosome组相比,Exosome@si-KRT8能够抑制FaDu/R细胞的活力(t=17.07,P<0.01),促进其凋亡水平,并且抑制FaDu/R细胞耐药相关蛋白的表达(t P-gp=103.20,t MDR1=238.60,P<0.01)。此外,Exosome@si-KRT8还能抑制FaDu/R细胞的迁移侵袭能力(t=42.30,t=122.00,P<0.01),并在促进E-cadherin的表达同时抑制N-cadherin的表达水平(t=130.80,t=83.90,P<0.01)。结论下咽癌患者血清来源的外泌体封装si-KRT8能够增强下咽癌细胞的化疗敏感性,且其作用机制可能与抑制细胞上皮间质转化有关。

目的探讨以针刀为主的综合治疗方案治疗后外侧型颈椎间盘突出症的临床疗效。方法选择2020年1月—2022年1月泉州市正骨医院针灸推拿科收治的82例后外侧型颈椎间盘突出症患者为研究对象,给予针刀结合手法、药物、牵引等综合疗法治疗。记录临床疗效、治疗前后的疼痛情况、颈椎功能障碍情况、生活质量、不良反应。结果(1)痊愈20例(24.2%),显效36例(43.9%),有效19例(23.2%),无效7例(8.5%),总体有效率为91.5%。(2)治疗前,患者平均视觉模拟评分法(visual analogue scale,VAS)评分为(5.268±0.969)分;治疗后,VAS评分为(2.317±0.910)分,差异有统计学意义(P<0.05)。(3)治疗前,颈椎功能障碍指数量表(neck disability index,NDI)评分为(434.965±4.182)分;治疗后,NDI评分为(12.122±4.448)分,差异有统计学意义(P<0.05)。(4)治疗前,患者平均生活质量调查问卷(European organization for research and treatment of cancer quality of life questionnaire core30 items,QLQ-C30)评分为(50.622±10.426)分;治疗后,QLQ-C30评分为(82.136±3.915)分,差异有统计学意义(P<0.05)。(5)治疗期间1例患者因针刀对颈椎的刺激而发生血肿,不良反应发生率为1.22%。结论针刀为主综合治疗后外侧型颈椎间盘突出症的临床疗效肯定,能有效缓解疼痛,改善患者颈椎功能障碍,是治疗此疾病的一种较理想的疗法。

In this paper,we study the joint bandwidth allocation and path selection problem,which is an extension of the well-known network utility maximization(NUM)problem,via solving a multi-objective minimization problem under path cardinality ***,such a problem formulation captures various types of objectives including proportional fairness,average delay,as well as load *** addition,in order to handle the"unsplittable flows",path cardinality constraints are added,making the resulting optimization problem quite challenging to solve due to intrinsic nonsmoothness and *** all existing works deal with such a problem using relaxation techniques to transform it into a convex optimization ***,we provide a novel solution framework based on the linearized alternating direction method of multipliers(LADMM)to split the original problem with coupling terms into several *** then derive that these subproblems,albeit nonconvex nonsmooth,are actually simple to solve and easy to implement,which can be of independent *** some mild assumptions,we prove that any limiting point of the generated sequence of the proposed algorithm is a stationary *** simulations are performed to demonstrate the advantages of our proposed algorithm compared with various baselines.

Dynamic evolutions of ecological carrying capacity in poor areas using ecological footprint model at Ruyang County of China

In this study,the ecological footprint(EF)from 2004 to 2013 of a poor county chosen from Central China was measured and analyzed by using the methodology and theory of *** results showed that in the past 10 years,the county’s ecological footprint per capita was growing gradually,increasing from 0.7053 hm^(2)in 2004 to 1.4473 hm^(2)in 2013,with growth clearly accelerated in recent *** the same period,the ecological carrying capacity per capita reduced from 0.6351 hm^(2)to 0.5018 hm^(2).Additionally,regional development had been in a state of ecological deficit,per capita ecological deficit increased from 0.0702 hm^(2)to 0.9456 hm^(2).This led to contradictions between regional socio-economic development and sustainability of natural ecosystems,meaning the development of the region is *** to reduce the ecological deficit were proposed to coordinate regional industrialization,urbanization,ecological environmental protection,and to promote the sustainable development in the region(*** industrial structure and consumption structure,promoting economic transformation,scientific planning and management of land).The results could provide a scientific basis for studies on ecological carrying capacity of the poor areas in Central China.

Genetic and pathogenic characterization of new infectious bronchitis virus strains in the GVI-1 and GI-19 lineages isolated in central China

Avian infectious bronchitis(IB)is a highly contagious infectious disease caused by infectious bronchitis virus(IBV),which is prevalent in many countries worldwide and causes serious harm to the poultry *** present,many commercial IBV vaccines have been used for the prevention and control of IB;however,IB outbreaks occur *** this study,two new strains of IBV,SX/2106 and SX/2204,were isolated from two flocks which were immunized with IBV H120 vaccine in central *** and recombination analysis indicated that SX/2106,which was clustered into the GI-19 lineage,may be derived from recombination events of the GI-19 and GI-7 strains and the LDT3-A *** analysis showed that SX/2204 belongs to the GVI-1 lineage,which may have originated from the recombination of the GI-13 and GVI-1 strains and the H120 *** virus cross-neutralization test showed that the antigenicity of SX/2106 and SX/2204 was different from *** experiments found that both SX/2106 and SX/2204 could replicate effectively in the lungs and kidneys of chickens and cause disease and death,and H120 immunization could not provide effective protection against the two IBV *** is noteworthy that the pathogenicity of SX/2204 has significantly increased compared to the GVI-1 strains isolated previously,with a mortality rate up to 60%.Considering the continuous mutation and recombination of the IBV genome to produce new variant strains,it is important to continuously monitor epidemic strains and develop new vaccines for the prevention and control of IBV epidemics.

Background Chicken is one of the most numerous and widely distributed species around the world,and many studies support the multiple ancestral origins of domestic *** research regarding the yellow skin phenotype in domestic chickens(regulated by BCO2)likely originating from the grey junglefowl serves as crucial evidence for demonstrating the multiple origins of ***,beyond the BCO2 gene region,much remains unknown about the introgression from the grey junglefowl into domestic ***,in this study,based on wholegenome data of 149 samples including 4 species of wild junglefowls and 13 local domestic chicken breeds,we explored the introgression events from the grey junglefowl to domestic *** We successfully detected introgression regions besides BCO2,including two associated with growth trait(IGFBP2 and TKT),one associated with angiogenesis(TIMP3)and two members of the heat shock protein family(HSPB2 and CRYAB).Our findings suggest that the introgression from the grey junglefowl may impact the growth performance of ***,we revealed introgression events from grey junglefowl at the BCO2 region in multiple domestic chicken breeds,indicating a phenomenon where the yellow skin phenotype likely underwent strong selection and was ***,our haplotype analysis shed light on BCO2 introgression event from different sources of grey junglefowl into domestic chickens,possibly suggesting multiple genetic flows between the grey junglefowl and domestic *** In summary,our findings provide evidences of the grey junglefowl contributing to the genetic diversity of domestic chickens,laying the foundation for a deeper understanding of the genetic composition within domestic chickens,and offering new perspectives on the impact of introgression on domestic chickens.

Background Understanding of the ratio of photosynthetic photon flux density（Qp） to global solar radiation（Rs）(Qp/Rs) is crucial for applying Rsto ecology-related *** studies reported Qp/Rsand its variations based on measurements from a single observatory tower,instead of multi-site-based measurements over complex *** may neglect spatial heterogeneity in the terrain,creating a gap in an understanding of how terrain affects Qp/Rsand how this effect interacts with meteorological *** Here the Qingyuan Ker Towers（three towers in a valley with different terrains:T1,T2,and T3） were utilized to measure Qpand Rsover mountainous forests of Northeast *** airborne LiDAR system was used to generate a digital elevation model,and sky view factor of sectors（SVFs） divided from the field of view of tower's pyranometer was calculated as a topographic factor to explain the variations of Qp/*** The results identified significant differences in Qp/Rsof the three towers at both daily and half-hour scales,with larger differences on clear days than on overcast ***/Rswas positively correlated with SVFsof T1 and T3,while this correlation was negative with that of *** effect of SVFson Qp/Rsinteracted with clearness index,water vapor pressure and solar zenith *** forest-based importance assessment demonstrated that explanation（R2） on Qp/Rswas improved when SVFswas included in the predictor variable set,indicating that incorporating terrain effects enhances the prediction accuracy of Qp/*** improvement in the R2values was more pronounced on clear days than on overcast days,suggesting that the effect of terrain on Qp/Rsdepended on sky *** All findings suggested that Qp/Rsis affected by terrain,and integrating terrain information into existing Qp/Rsmodels is a feasible solution to improve Qp/Rsestimates in mountainous areas.

随着全球价值链的迅速发展,中间品通过复杂的供应商网络和管理模式融入全球价值链,成为跨国公司价值链投资决策的重要影响因素。本文基于TiVA数据库的数据,探讨不同制造成本对价值链参与度和价值链地位的影响。研究结果显示:处于价值链低端的国家,其中间品质量改进会降低其价值链参与度,提升价值链地位;处于价值链高端的国家,其中间品质量改进则会提升价值链参与度,降低其价值链地位。研究结果对于提高我国中间品质量,提升我国在全球价值链中的地位具有重要意义和价值。基于研究结果,应进一步提升我国企业生产技术和创新能力,重视高技能劳动力的培养,加快传统劳动密集型制造业转型升级。
