Malignant Transformation and Abnormal Expression of Eukaryotic Initiation Factor in Bronchial Epithelial Cells Induced by Cadmium Chloride

Objective To analyze the relationship between malignant transformation and abnormal expression of eukaryotic initiation factor 3 （eIF3 p36） in human bronchial epithelial （16HBE） cells induced by cadmium chloride （CdCl2）. Methods 16HBE cells were treated several times with different concentrations of CdCl2. Tumorigenic potential of transformed cells was identified by assays for anchorage-independent growth in soft agar and for tumorigenicity in nude mice after the 35th passage. Total RNA was isolated from 16HBE cells induced by CdC12, including non-transformed, Cd-transformed, and Cd-tumorigenic cell lines. Special primers for eIF3 p36 were designed and the expression of eIF3 mRNA in different cell lines was detected with fluorescent quantitative-polymerase chain reaction technique （FQ-PCR）. Results The 35th passage of 16HBE cells transformed by CdCl2 exhibited overlapping growth. Compared with the non-transformed cells, colonies of transformed cell lines in soft agar showed statistically significant increases and dose-dependent effects （P〈0.01）. All Cd-induced transformed cell lines formed rumors in nude mice within 2 weeks of inoculation, but none of the mice injected with non-transformed cells showed tumors even after 3 weeks. All tumors were pathologically identified as poorly differentiated squamous cell carcinoma. The eIF3 p36 genes in different stages of 16HBE cells transformed by CdCl2 were elevated as compared with the non-transformed control （P〈0.01）, and the eIF3 expression increased with the degree of cell malignancy. Conclusion CdCl2 is capable of inducing morphological transformation in 16HBE cells and transformed cells are potentially tumorigenic. Over-expression of eIF3 p36 is positively correlated with malignant transformation of 16HBE cells induced by CdCl2 and may be one of the molecular mechanisms potentially responsible for carcinogenesis due to Cd.

Effects of chromium nanoparticle dosage on growth,body composition,serum hormones and tissue chromium in Sprague-Dawley rats

This 6-week study was conducted to evaluate the effects of seven different levels of dietary chromium (Cr) (0,75,150,300,450,600,and 1200 ppb Cr) in the form of Cr nanoparticle (CrNano) on growth,body composition,serum hormones and tissue Cr in Sprague-Dawley (SD) rats. Seventy male SD rats (average initial body weight of (83.2±4.4) g) were randomly assigned to seven dietary treatments (n=10). At the end of the trial,body composition was assessed via dual energy X-ray absorptiometry (DEXA). All rats were then sacrificed to collect samples of blood,organs and tissues for determination of serum hormones and tissue Cr contents. The results indicated that lean body mass was significantly increased (P<0.05) due to the addition of 300 and 450 ppb Cr from CrNano. Supplementation of 150,300,450,and 600 ppb Cr decreased (P<0.05) percent body fat significantly. Average daily gain was increased (P<0.05) by addition of 75,150,and 300 ppb Cr and feed efficiency was increased (P<0.05) by supplementation of 75,300,and 450 ppb Cr. Addition of 300 and 450 ppb Cr decreased (P<0.05) the insulin level in serum greatly. Cr contents in liver and kidney were greatly increased (P<0.05) by the addition of Cr as CrNano in the dosage of from 150 ppb to 1 200 ppb. In addition,Supplementation of 300,450,and 600 ppb Cr significantly increased (P<0.05) Cr content in the hind leg muscle. These results suggest that supplemental CrNano has beneficial effects on growth performance and body composition,and increases tissue Cr concentration in selected muscles.

All-perovskite tandem solar cells have the potential to surpass the theoretical efficiency limit of single junction solar cells by reducing thermalization ***,the challenges encompass the oxidation of Sn2+to Sn4+and uncontrolled crystallization kinetics in Sn-Pb perovskites,leading to nonradiative recombination and compositional heterogeneity to decrease photovoltaic efficiency and operational ***,we introduced an ionic liquid additive,1-ethyl-3-methylimidazolium iodide （EMIMI） into Sn-Pb perovskite precursor to form low-dimensional Sn-rich/pure-Sn perovskites at grain boundaries,which mitigates oxidation of Sn2+to Sn4+and regulates the film-forming dynamics of Sn/Pb-based perovskite *** optimized single-junction Sn-Pb perovskite devices incorporating EMIMI achieved a high efficiency of 22.87%.Furthermore,combined with wide-bandgap perovskite sub-cells in tandem device,we demonstrate 2-terminal all-perovskite tandem solar cells with a power conversion efficiency of 28.34%,achieving improved operational stability.

粘贴钢板加固钢筋混凝土受弯构件,加固前原构件截面已存在应变和应力,而新粘钢板在新增荷载下开始受力,桥梁构件具有不同受力阶段的特点。现有《公路桥梁加固设计规范》(JTJ/TJ22—2008)中未给出T形截面梁粘贴钢板加固受弯构件正截面承载能力计算公式,而且也不能反映各阶段材料的受力特点。根据粘贴钢板加固钢筋混凝土T形截面梁发生适筋破坏,建立了正截面极限承载力计算公式,并对粘贴钢板的应力取值进行了分析,在常用的粘贴钢板厚度内,可直接取钢板的抗拉强度设计值。并根据平截面假定,建立了各阶段混凝土、钢筋和钢板应力的计算公式,材料的最终应力按叠加原理计算得到。算例分析表明,建立的正截面抗弯承载力计算公式可行,可供桥梁加固设计参考。

目的探讨水中环氧氯丙烷的气相色谱测定法。方法水中环氧氯丙烷经二氯甲烷萃取,高倍浓缩,经FFAP大口径毛细管分离,氢焰离子化检测器检测。结果该方法线性范围为0.502～10.04μg/ml,r=0.9992。检测限为0.0005mg/ml。回收率为90.3%～99.9%,RSD为3.5%～4.3%。结论该方法简便,灵敏,准确,适用于输配水设备以及涂料浸泡水中痕量环氧氯丙烷的检测。

Direct and rapid detection of serum amino acid and monoamine neurotransmitters to assist the diagnosis of panic disorder

The measurement of amino acid and monoamine neurotransmitters（NTs） in serum plays a central role in the identification and monitoring of clinical panic disorder（PD）. However, the strong polarity and chemical heterogeneity of NTs make direct extraction and rapid detection extremely challenging. Herein, we developed a strategy that integrated solid phase extraction based on magnetic Fe3O4coated with polydopamine（Fe3O4@PDA） and direct analysis of mass spectrometry in real time（DART-MS） for quantification of 10 NTs. We systematically investigated the optimal conditions of this strategy to achieve good accuracy and precision. Compared with existing magnetic solid phase extraction methods for NTs detection in biological samples, this strategy could adsorb more targets while having similar adsorption efficiencies, and the total process time was reduced by an average of 69.42%. We rapidly determined the metabolic abnormality of 6 types of NTs in the serum of PD patients and demonstrated their diagnostic accuracy. In addition, an outstanding feature of PD in which tryptophan was more metabolized by the KYN pathway and the 5-hydroxytryptamine pathway was inhibited could be elucidated by this strategy. Our results highlighted the potential ability of Fe3O4@PDA coupled with DART-MS to assist in the clinical diagnosis of PD.

以草酸二乙酯和水合肼为原料,经取代、中和反应合成草酰肼二硝酸盐(OHDN),总收率为97%,并通过红外光谱、核磁共振、元素分析等进行了结构表征;研究了草酰肼在稀硝酸中的水解反应,确定了水解产物为草酸和硝酸肼;优化了OHDN合成条件,确定适宜的反应条件:温度为40℃,时间为2h;同时研究了OHDN在高温和室温水溶液中的稳定性,结果表明,OHDN在高温水溶液中化学稳定性差,而在室温水溶液中较为稳定;培养获得了OHDN·2H_2O单晶,结构分析表明:OHDN·2H_2O晶体结构属单斜晶系,空间群为P2(1)/n,a=0.4639(13)nm,b=1.0992(3)nm,c=1.0570(3)nm,V=0.5301(2)nm^3,Z=2,Dc=1.755 g·cm^(-3),F(000)=292,R_1=0.0855,wR_2=0.0895。
