Effect of Rhizobacterium Rhodopseudomonas palustris Inoculation on Stevia rebaudiana Plant Growth and Soil Microbial Community

There is an increasing concern that the continuous use of chemical fertilizers might lead to harmful effects on soil ecosystem.Accordingly, a biocompatible approach involving inoculation of beneficial microorganisms is presented to promote plant growth and simultaneously minimize the negative effect of chemical fertilizers. In this study, Rhodopseudomonas palustris, a plant growth-promoting rhizobacterium(PGPR), was inoculated into both fertilized and unfertilized soils to assess its influence on Stevia rebaudiana plant growth and microbial community in rhizosphere soils in a 122-d field experiment. Soil enzyme assays(dehydrogenase, urease, invertase, and phosphomonoesterase), real-time quantitative polymerase chain reaction(RT-_qPCR), and a high-throughput sequencing technique were employed to determine the microbial activity and characterize the bacterial community. Results showed that the R.palustris inoculation did not significantly influence Stevia yields and root biomass in either the fertilized or unfertilized soil. Chemical fertilization had strong negative effects on soil bacterial community properties, especially on dehydrogenase and urease activities.However, R. palustris inoculation counteracted the effect of chemical fertilizer on dehydrogenase and urease activities, and increased the abundances of some bacterial lineages(including Bacteroidia, Nitrospirae, Planctomycetacia, Myxococcales, and Legionellales). In contrast, inoculation into the unfertilized soil did not significantly change the soil enzyme activities or the soil bacterial community structure. For both the fertilized and unfertilized soils, R. palustris inoculation decreased the relative abundances of some bacterial lineages possessing photosynthetic ability, such as Cyanobacteria, Rhodobacter, Sphingomonadales, and Burkholderiales. Taken together, our observations stress the potential utilization of R. palustris as PGPR in agriculture, which might further ameliorate the soil microbial properties in the long run.

Cytokinins(CKs)function in plant development and during stress responses,but their role in drought tolerance in wheat is unknown.In the present study,24 isopentenyltransferase(IPT)genes,encoding ratelimiting enzymes in CK biosynthesis were identified in the wheat genome.The chromosomal locations and structures of the genes,protein properties,and phylogenetic relationships were characterized.ATP/ADP TaIPT genes showed tissue-specific expression.TaIPT2,TaIPT7,and TaIPT8 expression was rapidly induced by 0.5–1 h drought treatments,which decreased to low levels after 2 h drought treatment,as did most other TaIPT genes.TaIPT8-5a/5b/5d triple mutants showed decreased levels of tZtype CK under normal and drought conditions and reduced drought tolerance,which,however,did not manifest as phenotype alterations.By contrast,transgenic wheat plants with drought-induced TaIPT8showed increased drought tolerance.Our study provides a foundation for further investigation of TaIPT genes and novel insights into the role of CKs in the drought response of wheat.

Taking swash plate axial piston pump as the research object,the mechanism of fluid vibration and transfer rule are analyzed.The pump shell can be assumed as the ultimate recipient of vibration transmission,the path model and differential equations from the fluid to the shell are established.The parameters of the path model are determined by the simulation software,and the mathematical model is solved by the simulation software.And time/frequency domain analysis of vibration acceleration of shell is presented.Based on the different influence of various parameters in the transfer path model on transfer characteristics and vibrational recipients,the time-varying parameters are studied by using sensitivity analysis theory,and the influence of the structural parameters on the vibration characteristics of vibration subject is quantitatively analyzed.The research in the paper provides theoretical basis for vibration analysis and structure parameter optimization of axial piston pump.

1 Introduction The coal bearing basin in the east of Northeast China is many, and the little or middle fault depression of low or middle rank coal bearing basin is the main, including Jixi, Boli, Shuangyashan, Hegang, Hulin and other basins （TANG Shuheng, 2000）. Several coal bearing basins have seen well show of CBM（Tablel）. But the research of the coal bed gas system is not carried out at present.

Dear Editor,Akabane disease is an arthropod-borne viral disease caused by the Akabane virus(AKAV)that can affect cattle,sheep,and goats.Inapparent infections in adults can lead months later to abortions,stillbirths,congenital defects,and death in newborns(Kurogi et al.,1976).Wild ruminants can also be affected,but abnormalities of the offspring in these species have not been reported.One report suggests that this virus can also cause clinical signs in pigs(Yanase et al.,2018).

煤岩孔隙结构特征是评价煤层储集能力和选层压裂的重要参数。综合应用低温氮吸附方法、核磁共振技术和氩离子抛光成像等方法,利用分形理论定量表征孔隙的非均质性,并探讨分形维数的影响因素。结果表明,八连城矿区煤岩有机质孔主要为植物组织孔、粒间孔和气孔,矿物质孔为溶蚀孔和黏土矿物孔。Ⅰ类曲线显示煤岩发育狭缝状孔和楔形孔。Ⅱ类曲线表明煤岩瓶型孔发育。核磁共振双峰型T2谱表明吸附孔较为发育,连通性差。三峰型显示渗流孔和裂隙发育,孔渗条件好。孔隙直径在2~100 nm时,水分含量和比表面积与D1表现为正相关关系。D2与灰分含量、平均孔径呈正相关和负相关。孔隙直径在0.1~10.0μm时,核磁共振法获得DN1与吸附孔表面积呈正相关,DN2与渗流孔的孔体积呈正相关;DM和溶蚀孔分形维数Dc,分别受到黏土矿物和长石含量的控制。因此,氮吸附Ⅰ型曲线煤层和三峰型核磁T2图谱煤层利于煤层气的开发。

The soybean PLATZ transcription factor GmPLATZ17 suppresses drought tolerance by interfering with stress-associated gene regulation of GmDREB5

Plant AT-rich sequence and zinc binding(PLATZ)transcription factors are a class of plant specific zincdependent DNA-binding proteins that function in abiotic stress response and plant development.In this study,31 GmPLATZ genes were identified in soybean.GmPLATZ17 was down-regulated by drought and exogenous abscisic acid.Transgenic Arabidopsis and soybean hairy roots overexpressing GmPLATZ17 showed drought sensitivity and inhibition of stress-associated gene transcription.In contrast,suppressed expression of GmPLATZ17 led to increased drought tolerance in transgenic soybean hairy roots.The GmPLATZ17 protein was verified to interact physically with the GmDREB5 transcription factor,and overexpression of GmDREB5 increased drought tolerance in soybean hairy roots.Interaction of GmPLATZ17 with GmDREB5 was shown to interfere with the DRE-binding activity of GmDREB5,suppressing downstream stress-associated gene expression.These results show that GmPLATZ17 inhibits drought tolerance by interacting with GmDREB5.This study sheds light on PLATZ transcription factors and the function of GmPLATZ17 in regulating drought sensitivity.

煤矸石-粉煤灰用作井下充填材料实验研究

黄金矿山由于尾矿氰化物含量较高无法用于井下采空区充填,但随着井下采空区逐渐扩大,亟需充填材料用于回填,研究利用煤矸石-粉煤灰用作骨料与水泥混合充填至采空区。岩质煤矸石的普氏硬度为2.74,易于破碎加工。电厂干排粉煤灰,细度为18.6%,颗粒较细。将煤矸石破碎至20 mm以下用作粗骨料,粉煤灰为细骨料,当煤矸石-粉煤灰比为7∶3时,混合骨料级配与Fuller理论曲线接近,级配良好。以水泥为胶凝材料配制1∶7、1∶9两种灰砂比的充填材料,混合料浆浓度为83%时,均能满足采空区管道高浓度输送的流动度要求。测定不同养护龄期下充填材料的单轴抗压强度,两种灰砂比材料在浓度为83%、82%下,28 d的强度均超过4 MPa,最高可达6.03 MPa,可满足采空区顶底部或中心部位的充填强度要求。

Research and application of uniform material counting method based on machine vision

Manufacturing and agricultural industries use manual methods to count materials. This leads to low accuracy and inefficiency. This paper proposes a secondary counting method that combines main and differential counting. The area-fill identification algorithm is applied to mark the counted materials. To verify the effectiveness of the proposed counting algorithm, numbers of countings are conducted for different materials, such as the screws, hole gaskets, beans, jujube, etc. The results show that the counting accuracy reaches 98% for materials with size of 2—20 mm. The method has delivered a high-efficiency and high-accuracy automatic intelligent counting, with a wide range of application prospects and reference value.
