Harnessing Theaflavin-3，3'-Digallate for Virulence Attenuation: A Targeted Approach Against Foodborne Staphylococcus aureus Pathogenicity

Addressing the uncontrolled spread and increase in antibiotic resistance in methicillin-resistant Staphylococcus aureus （MRSA） will require new control strategies， particularly to improve the safety of food. Our results revealed the efficacy of Theaflavin-3，3'-digallate （TFBG）， which is a novel polyphenol derived from tea， in targeting the key regulatory protein MgrA in S. aureus. Through fluorescence anisotropy， we showed that the IC50of TFBG was 26.76 μg/mL. TFBG uniquely counters S. aureus by regulating its virulence factors and adhesion processes rather than by killing the bacteria directly. This compound alters the expression of key virulence factors and modulates the transcription levels of genes related to adhesion in S. aureus， ultimately reducing the bacteria’s ability to adhere to fibrinogen and its hemolytic activity. Our assays confirmed that TFBG directly interacts with the MgrA protein in MRSA， and we identified critical binding sites. Our in vivo studies highlighted the potent efficacy of TFBG. TFBG administration is an innovative approach to improve food safety by diminishing MRSA virulence and reducing its pathogenicity.

The commercialization of sodium-ion batteries is based on developing low-cost,highly stable,and safe cathode and anode ***,the promising hard carbon anode and layered oxide cathode suffer from low sodium-embedded potential near 0.1 V and severe phase transitions,which cause safe problem and short lifespan,***,we design a low-strain bipolar P2-Na_(0.7)Ni_(0.25)Fe_(0.2)Ti_(0.55)O_(2) to solve the mentioned obstacles,whereas(Ni,Fe)and Ti provide charge compensation when it is used as cathode and anode,*** is revealed that the bipolar layered oxide undergoes solid-solution reaction when used as cathode or anode,and exhibits volume-complementary feature in a sodium-ion full-cell,as identified by in-situ X-ray ***,the safe symmetric sodium-ion full-cell exhibits excellent cyclic stability with 91.7%capacity retention after 200 *** work will provide a new horizon for designing safe and stable sodium-ion batteries.

Solving the subset sum problem by the quantum Ising model with variational quantum optimization based on conditional values at risk

The subset sum problem is a combinatorial optimization problem,and its complexity belongs to the nondeterministic polynomial time complete(NP-Complete)*** problem is widely used in encryption,planning or scheduling,and integer *** accurate search algorithm with polynomial time complexity has not been found,which makes it challenging to be solved on classical *** effectively solve this problem,we translate it into the quantum Ising model and solve it with a variational quantum optimization method based on conditional values at *** proposed model needs only n qubits to encode 2ndimensional search space,which can effectively save the encoding quantum *** model inherits the advantages of variational quantum algorithms and can obtain good performance at shallow circuit depths while being robust to noise,and it is convenient to be deployed in the Noisy Intermediate Scale Quantum *** investigate the effects of the scalability,the variational ansatz type,the variational depth,and noise on the ***,we also discuss the performance of the model under different conditional values at *** computer simulation,the scale can reach more than nine *** selecting the noise type,we construct simulators with different QVs and study the performance of the model with *** addition,we deploy the model on a superconducting quantum computer of the Origin Quantum Technology Company and successfully solve the subset sum *** model provides a new perspective for solving the subset sum problem.

The construction of C-N bonds is of great importance in the fields of biology,medicine,chemistry and materials ***,the replacement of organic base from pyridine to piperidine in the Knoevenagel condensation process unexpectedly yields a series of novel organic molecules containing C-N ***,the synthesis method does not require any external transition-metals catalysis,and photo-/***,when the new compound 1b is added as a third component to a well-known binary system of PM6:Y6,the efficiency of the organic solar cell is significantly improved,resulting in an outstanding efficiency of 18.0%,which is one of the highest values reported to date for PM6:Y6-based ternary organic solar cells.

The transition-metal-catalyzed migratory functionalization has emerged as a robust protocol for the building of C–C bonds remote from the “initiation” position. However, a similar strategy for the construction of quaternary carbon centers is still underdeveloped and only a limited number of reports exist. Herein, we report a nickel-catalyzed migratory hydrocyanation of unconjugated dienes to construct remote cyano-substituted quaternary carbon centers. This transformation features exceptional regioselectivity, mild reaction conditions, broad substrate scope and high yields. The synthetic utility of this method has been highlighted by a series of product derivatizations, and the potential of this transformation has been extended to synthesize TRPV1 antagonist and the key intermediate in the total synthesis of quebrachamine. Density functional theory(DFT) studies unveiled that the specific catalytic pocket assumed a significant role in the selective formation of cyano-substituted quaternary carbon centers.

Temporal and Spatial Trends in HIV Positivity Rate for VCT Clinics—China,2015–2022

Introduction:Human immunodeficiency virus(HIV)voluntary counseling and testing(VCT)clinics play a critical role in identifying and diagnosing HIV *** study aimed to describe the trend of HIV positivity rate(HPR)among Chinese VCT clinics between 2015 and ***:This study utilized data from the China Information System for Disease Control and Prevention to analyze the trend in the HPR for VCT clinics from 2015 to *** HPR was calculated by dividing the number of newly-reported HIV cases by the number of HIV tests,multiplied by 100%.To identify temporal and spatial trends in the HPR,we employed joinpoint regression analysis and the Getis-Ord hotspot ***:From 2015 to 2022,VCT clinics in China performed a total of 22,075,386 HIV tests,leading to the identification of 260,353 HIV cases,resulting in a HPR of 1.18%.The HPR consistently declined over the study period,with an average annual percent change(AAPC)of−7.5%(95%confidence interval:−12.6%,−2.2%,P<0.05).The number of HPR hotspots also decreased from 41 in 2015 to 23 in *** HPR hotspots were primarily located in Yunnan,Sichuan,Guangdong,and Guangxi provincial-level administrative divisions(PLADs).Among the 31 PLADs,16 showed a significant decrease in HPR during the study period(AAPC<0,PAAPC<0.05).Conclusions:VCT clinics in China have played a significant role in identifying HIV *** declining HPR observed in these clinics may indicates the progress has been made in some degree in mitigating HIV among high-risk ***,it is crucial to further improving the utilization of VCT clinics for HIV testing.

Construction and manipulation of origami-inspired tubular structures with controlled mechanical buckling for collection and transportation of microspheres based on optically induced electrokinetics

The development of microengineered hydrogels has opened up unlimited possibilities for designing complex structures at the microscale. In this study, we constructed an origami-inspired tubular structure with controlled mechanical buckling based on optically induced electrokinetics(OEK). By inducing a stress gradient in the thickness, a tubular structure can be formed from a poly(ethylene glycol) diacrylate(PEGDA) hydrogel film of various shapes that have been custom fabricated. To achieve an ideal three-dimensional(3D) structure, the amplitude of the tubular structure can be controlled by adjusting the aspect ratios or polymerization time. Furthermore, the tubular structure can be manipulated for the collection and transportation of *** summary, we provide an effective method for designing 3D structures at the micro-nano scale. This forming method holds great potential for achieving various functions in tissue engineering, drug packaging, and transportation in the future.

Epigenetic modifications including DNA methylation, histone modifications, chromatin remodeling, and RNA modifications complicate gene regulation and heredity and profoundly impact various physiological and pathological processes. In recent years, accumulating evidence indicates that epigenetics is vulnerable to environmental changes and regulates the growth, development, and diseases of individuals by affecting chromatin activity and regulating gene expression. Environmental exposure or induced epigenetic changes can regulate the state of development and lead to developmental disorders, aging, cardiovascular disease, Alzheimer's disease, cancers, and so on. However, epigenetic modifications are reversible. The use of specific epigenetic inhibitors targeting epigenetic changes in response to environmental exposure is useful in disease therapy. Here, we provide an overview of the role of epigenetics in various diseases. Furthermore, we summarize the mechanism of epigenetic alterations induced by different environmental exposures, the influence of different environmental exposures, and the crosstalk between environmental variation epigenetics, and genes that are implicated in the body's health. However, the interaction of multiple factors and epigenetics in regulating the initiation and progression of various diseases complicates clinical treatments. We discuss some commonly used epigenetic drugs targeting epigenetic modifications and methods to prevent or relieve various diseases regulated by environmental exposure and epigenetics through diet.
