DEAD-box helicase 17(DDX17)is a typical member of the DEAD-box family with transcriptional cofactor *** DDX17 is abundantly expressed in the myocardium,its role in heart is not fully *** generated cardiomyocyte-specific Ddx17-knockout mice(Ddx17-cKO),cardiomyocyte-specific Ddx17 transgenic mice(Ddx17-Tg),and various models of cardiomyocyte injury and heart failure(HF).DDX17 is downregulated in the myocardium of mouse models of heart failure and cardiomyocyte ***-specific knockout of Ddx17 promotes autophagic flux blockage and cardiomyocyte apoptosis,leading to progressive cardiac dysfunction,maladaptive remodeling and progression to heart *** of DDX17 expression in cardiomyocytes protects cardiac function under pathological *** studies showed that DDX17 can bind to the transcriptional repressor B-cell lymphoma 6(BCL6)and inhibit the expression of dynamin-related protein 1(DRP1).When DDX17 expression is reduced,transcriptional repression of BCL6 is attenuated,leading to increased DRP1 expression and mitochondrial fission,which in turn leads to impaired mitochondrial homeostasis and heart *** also investigated the correlation of DDX17 expression with cardiac function and DRP1 expression in myocardial biopsy samples from patients with heart *** findings suggest that DDX17 protects cardiac function by promoting mitochondrial homeostasis through the BCL6-DRP1 pathway in heart failure.

Objective Hydrogen sulfide(HS)is considered to be a new gaseous molecular alongside with NO and *** of evidences indicate that HS performs a wide range of physiological and pathological ***, the central cardiovascular effects of HS are not fully *** present study,we tested the hypothesis that HS in CNS may inhibit NADPH oxidase and decrease the production of ROS,thus mediate central cardiovascular effects. Methods Experiments were completed in Wistar-Kyoto(WKY)rats and spontaneous hypertensive rats(SHR).We used morphological and molecular biological techniques to explore the mechanisms of central cardiovascular effects of *** The following observations were made:(1)Expressions of CBS of RVLM were increased in WKY rats compared with SHR rats.(2.Intracerebroventricular(ICV)SAM(1 nmol/10μL)significantly decreased mean arterial pressure(MAP);ICV NaHS by low concentration(4 nmol/10μL)significantly decreased mean arterial pressure(MAP), while high concentration(2. nmol/10μL)could increase mean arterial pressure(MAP).(3)Microinjection of NaHS (400 pmol/0.1μL)into the RVLM notably decreased MAP.(4)The level of ROS in RVLM of SHR rats was higher than WKY rats,and after ICV with NaHS(4 nmol/10μL),the level of ROS decreased compared with SHR rats. Moreover,we also certified that the decrease of ROS associated with the inhibition of the activity of NADPH oxidase. Conclusion The results of present study suggest that HS in CNS may lower blood pressure by inhibiting the activity of NADPH oxidase in the RVLM in SHR.

Conclusion Our findings demonstrate that toll-like receptor 4 contributes to the bone cancer pain-induced mechanical allodynia.

Objective:The aim of this study was to investigate the effect of adenosine AR in the rostral ventrolateral medulla(RVLM) of rats with acute myocardial ischemia(AMI) on cardiovascular regulation and whether it was involved in the mechanism of the electroacupuncture(EA) on AMI *** and Methods:Experiment was performed on male Sprague-Dawley(SD) rats weighing 2.0-2.0 *** animals were randomly divided into sham, AMI and AMI+EA PC6 groups.A rat model of AMI was made by ligation of the left anterior descending branch of the coronary *** was treated at bilateral"Neiguan"acupoints(PC 6) for 30 min and continued 5 *** that,the technique of immunohistochemistry was carried out for examining the alteration in expression of adenosine A receptor(AR) in the ***,western blot and real-time PCR were also used to investigate the protein and mRNA level of adenosine AR in the same *** of CGS2.680,an agonist of adenosine AR,into the RVLM was performed to observe changes of the mean arterial blood pressure(MBP) and heart rate (HR).Results:The number of adenosine AR immunoreactive positive neurons in the RVLM of the AMI group was more than that of the sham group(P2.680 into the RVLM produced a fall in MBP and HR compared with microinjection of aCSF in the each *** attenuation in MBP and HR was significantly greater in AMI group than that in sham group(P<0.05),whereas the attenuation in AMI+EA PC 6 group was less as compared with that in AMI group(P<0.05).Conclusion:We concluded that adenosine and its corresponding receptor AR play depressor and bradycardia roles in the *** addition, adenosine AR of the RVLM participates in the central regulation of cardiovascular activ

Background and Aims:We asked if comprehensive bile acid profiling could provide insights into the physiopathology of ABCB4-mutated patients and evaluated the prognostic value of taurine-conjugated tetrahydroxylated bile acid(tauro-THBA)in ***:Serum bile acid profiles were evaluated in 13 ABCB4-mutated patients with 65 healthy controls by ultra-high-performance liquid chromatography/multiple-reaction monitoring-mass spectrometry(UPLC/MRM-MS).The concentration of tauro-THBA was compared between ABCB4-mutated patients with different *** areas under the curve(AUCs)of tauro-THBA were compared between ABCB11-mutated patients with native liver survival and those who died or underwent liver transplantation before 3 years of age by receiver operating characteristic curve(ROC),with another patient cohort for further ***:The overall hydrophobicity indices of bile acids in ABCB4-mutated patients(12.99±3.2. m)were significantly lower than those of healthy controls(14.02.1.74 m,p11-mutated patients with different prognoses(p=0.0192..Of the 15 patients with good prognosis,14 were classified correctly and four of the five patients with a poor prognosis were classified correctly(14:15 vs.1:5,p=0.005)with tauro-THBA as a ***:Tauro-THBA concentration may be a biomarker for predicting the clinical outcome in low gammaglutamyl transferase intrahepatic cholestasis patients.

Background:Currently,the need for new therapeutic strategies involving programmed cell death protein-1(PD-1)monoclonal antibodies in the second-line setting of small cell lung cancer(SCLC)is *** study aimed to evaluate the efficacy and safety of anlotinib plus penpulimab as a second-line treatment for patients with SCLC who progressed after first-line platinum-based ***:This study included the patients from Cohort 4 of a single-arm,open-label,multicenter,phase II clinical trial.A safety run-in phase was performed under anlotinib(10/12.mg quaque die[QD],days 1–14)plus penpulimab(2.0 mg intravenously[IV],day 1)in a 2.-day cycle,followed by the formal trial in which the patients received anlotinib(12.mg QD,days 1–14)plus penpulimab(2.0 mg IV,day 1)in a 2.-day *** primary endpoint of the safety run-in phase was *** primary endpoint of the formal trial phase was the objective response rate(ORR).Results:From April 2.,2.2.,to November 2.,2.2.,2. patients were enrolled from 11 hospitals,including 2.in the safety run-in phase and 19 in the formal trial *** the formal trial phase,the ORR was 42.1.(8/19;95%confidence interval[CI]:17.7–66.6%).The median progression-free survival was 4.8 months(95%CI:2.9–11.3 months),and the median overall survival was 13.0 months(95%CI:4.6–not applicable[NA]months).The incidence of grade 3 treatment-related adverse events(TRAEs)was 52.4%(11/2.),and the incidence of treatment-related serious adverse events(AEs)was 2..6%(6/2.).Two AE-related deaths *** most common AEs were hypertension(57.1%,12.2.),hypothyroidism(42.9%,9/2.),and hypertriglyceridemia(38.1%,8/2.).Conclusions:In patients with SCLC who progressed after first-line platinum-based chemotherapy,the second-line anlotinib plus penpulimab treatment demonstrates promising anti-cancer activity and a manageable safety profile,which warrants further investigation.

Vascular endothelial growth factor(VEGF) promotes neurogenesis in the subventricular zone(SVZ),hippocampal dentate gyrus(DG),and the striatum of adult mammalian ***,effect of VEGF on cortical neurogenesis after stroke remains ***,we intraventricularly injected human VEGF-expressive plasmids(phVEGF) into rat brains after a transient middle cerebral artery occlusion(MCAO) and examined effects of VEGF on stroke-stimulated cortical *** results showed that 5’-bromodeoxyuridine positive(BrdU,a DNA-replication marker) cells were found in ipsilateral frontal cortex at 2.weeks of reperfusion and declined over *** of phVEGF did not alter such a decline,but it significantly increased BrdU cells compared with the control plasmid(pEGFP) injection at 2.4 and 8 weeks after ***-induced cortical neurogenesis was indicated by the presence of newborn immature(BrdU-Tuj 1), newborn mature(BrdU-MAP-2.,and newborn GABAergic(BrdU-GAD67) *** these neurons declined within 8 weeks after MCAO in the *** of phVEGF significantly increased BrdU-Tuj 1 neurons at 2.weeks,and BrdU-MAP-2.neurons and BrdU-GAD67 neurons at 4 and 8 weeks after *** length and branch numbers of BrdU-MAP-2.newborn neurons in the cortex were detected in both *** of phVEGF significantly increased neurite length and branch numbers of BrdU-MAP-2.newborn *** results demonstrate that VEGF enhances ischemia-induced cortical neurogenesis and maturation of newborn *** study further indicates that neuroprotection of VEGF against ischemic injury may associated with enhancement of neurogenesis in adult mammalian brains.

由区域选择性生长方法(即分子在预图案表面的自组装技术)制备的有机微纳米图案有望在光电和全彩显示方面发挥巨大作用.而制备多色图案薄膜的有机器件依赖于两种及多种有机分子的成核分配,特别是混合沉积方法的应用有效提高了多色薄膜成核的效率.本文利用动力学蒙特卡罗方法研究了两种分子间的结合能强弱对混合沉积两种分子以实现相分离式生长的影响.结果表明:当分子间结合能较弱时,两种分子表现出完全不同的相分离生长行为,绝大部分的非平面和平面分子分别沉积在电极顶部和侧面;当分子间结合能较强时,电极顶部及侧面都出现了两类分子的掺杂生长,导致了分子相分离生长的失效.研究证明了分子间的结合能会影响多种有机分子选区生长分离的效果,因此混合沉积两种有机分子时,通过适当调整分子间的结合能,在电极顶部和侧面可分别获得较纯结晶的单色薄膜,从而促进了多色有机微纳米图案在微显示等领域的进一步应用.

针对现有方法在风光预测时气象因素考虑不全面且未考虑风光功率关联性的问题，提出一种风光功率短期预测方法。首先，以云模型表征风光出力不确定性，逆向云结合灰色关联度分析不同气象特征对输出功率的影响程度，并设立选取标准及综合评分指标；其次，采用集合经验模态分解（EEMD）将选取相似日的功率数据分解为子序列；最后，将子序列和气象数据作为基于折射学习策略（RL）的灰狼算法（GWO）优化的改进长短期记忆网络（LSTM）模型的预测输入进行训练，对待预测日的子序列分别预测，并叠加得到短期区域风光发电功率的预测。以中国西北某风光联合电场数据为例，对该模型进行验证，结果表明，相比于现有预测模型，该文所提方法考虑了天气因素，具有较高的预测精度，能够较好地为区域风光联合电场的功率预测提供参考。
