本研究基于代谢组学结合生物信息学的策略分析热毒宁注射液与青霉素注射液联用引发类过敏反应(pseudo-allergic reactions,PARs)的潜在致敏原及作用机制。所有动物实验操作和福利均遵循河南中医药大学第一附属实验动物伦理与动物福利委员会要求(批号:YFYDW2.2.002.。基于UPLC-Q-TOF/MS技术结合UNIFI软件在热毒宁与青霉素混合注射液中共鉴定得到2.种化合物。进一步基于分子对接技术筛选出其10种与类过敏原受体MrgprX2.动剂位点具有强烈结合活性的潜在致敏原。采用UPLC-Q-TOF/MS技术进行代谢组学分析发现花生四烯酸、磷脂酰胆碱、磷脂酰丝氨酸、前列腺素类以及白三烯类等3.个内源性代谢物为热毒宁注射液与青霉素注射液联用引发PARs的差异代谢物。通过对“潜在致敏原-靶标-差异代谢物”交互网络分析获得二者联用时药液中的绿原酸类(绿原酸、新绿原酸、隐绿原酸、异绿原酸A等)以及β-内酰胺类致敏原可能主要通过调控PLD1、PLA2.12.和CYP1A1这三种上游信号靶标,主要激活花生四烯酸代谢通路,促使肥大细胞的脱颗粒,释放下游的内源性炎性介质,继而诱发PARs。

采用检测血清生化指标与血浆代谢组学特征关联探索热毒宁注射液(Reduning,RDN)与青霉素注射液(penicillin G,PG)序贯用药风险“时间窗”,以期降低二者联用引发不良反应发生风险。所有动物实验操作和福利均遵循河南中医药大学第一附属实验动物伦理与动物福利委员会要求(批号:YFYDW2.2.002.。利用ELISA检测大鼠血清中生化指标的含量变化检测相关过敏指标,判定出RDN联用PG能引起由补体途径激活后的类过敏反应(pseudo-allergic reactions,PARs)。进一步对二者设置不同时间给药间隔(1.5、2.3.5、4、6和8 h PG+RDN)进行考察,发现在3.5 h以内序贯给药,均能引起明显的PARs;而给药间隔超过4 h后PARs则明显减轻。同时采用LC-MS进行血浆代谢组学分析,平行比较大鼠的血清生化指标水平与血浆代谢轮廓特征,筛选出2.个差异代谢物在3.5 h前后呈现出与生化指标相似或相反的变化趋势。并富集到花生四烯酸代谢、甾体激素生物合成代谢、亚油酸代谢、甘油磷脂代谢、色氨酸代谢等10条类过敏相关通路。综上所述,RDN和PG序贯使用所导致的药物不良反应确实存在风险“时间窗”现象,在“时间窗”之后的间隔用药能有效降低不良反应发生风险。

The Circular Electron Positron Collider(CEPC)is a large scientific project initiated and hosted by china,fostered through extensive collaboration with international *** complex comprises four accelerators:a 3. 中子辐射俘获反应在反应堆运行、核装置设计及核天体物理研究中起重要的作用.4πBaF_(2.探测装置有着高时间分辨能力、低中子灵敏度、高探测效率等优点,适合开展中子辐射俘获反应截面数据的测量.中国原子能科学研究院核数据重点实验室建立了伽马全吸收装置(Gamma total absorption facility,GTAF),该装置用2.块六棱BaF_(2.晶体和12.五棱BaF_(2.晶体构成了外径2. cm,内径10 cm的球壳,覆盖了95.2.的立体角.利用GTAF在中国散裂中子源Back-n束线上,测量了197Au(n,γ)的反应截面数据.测量数据通过能量筛选、PSD方法、晶体多重性筛选进行了初步本底扣除,随后结合对^(nat)C及空样品的测量数据对本底进行了分析及扣除,获得了197Au俘获反应的产额,利用SAMMY程序拟合得到了^(197)Au在1—100 e V的共振能量、中子共振宽度和伽马共振宽度参数.实验测量结果与ENDF/B-VIII.0数据库符合良好,其共振参数存在一定差异,分析原因可能与GTAF能量分辨率、Back-n的中子能谱测量精度、以及实验本底扣除方法相关,这也是下一步工作的重点.

There exist differences in the epidemiological characteristics,clinicopathological features,tumor biological characteristics,treatment patterns,and drug selections between gastric cancer patients from the Eastern and Western *** Chinese Society of Clinical Oncology(CSCO)has organized a panel of senior experts specializing in all sub-specialties of gastric cancer to compile a clinical guideline for the diagnosis and treatment of gastric cancer since 2.16 and renews it *** into account regional differences,giving full consideration to the accessibility of diagnosis and treatment resources,these experts have conducted expert consensus judgment on relevant evidence and made various grades of recommendations for the clinical diagnosis and treatment of gastric cancer to reflect the value of cancer treatment and meeting health economic indexes in *** 2.2. The ^(2.2.Th(n,f)cross section is very important in basic nuclear physics and applications based on the Th/U fuel *** the time-of-flight method and a multi-cell fast-fission ionization chamber,a novel measurement of the^(2.2.Th(n,f)cross sec-tion relative to^(2.5)U in the 1–2.0 MeV range was performed at the china Spallation Neutron Source Back-n white neutron source(Back-n).The fission event-neutron energy spectra of^(2.2.Th and^(2.5)U fission cells were measured in the single-bunch *** 2.2.h/2.5U fission cross-sectional ratios were obtained,and the measurement uncertainties were 2.5–3.7%for energies in the 2.2. MeV range and 3.6–6.2.for energies in the 2.–2.0 MeV ***^(2.2.Th(n,f)cross section was obtained by introducing the standard cross section of^(2.5)U(n,f).The results were compared with those of previous theoreti-cal calculations,measurements,and *** measured 2.2.h fission cross section agreed with the main eva{2.ation results in terms of the experimental uncertainty,and 2.2.h fission resonances were observed in the 1–3.MeV *** present results provide^(2.2.Th(n,f)cross-sectional data for the eva{2.ation and design of Th/U cycle nuclear systems.
