Isolation and microbial transformation of tea sapogenin from seed pomace of Camellia oleifera with anti-inflammatory effects

In the current study,tea saponin,identified as the primary bioactive constituent in seed pomace of Camellia oleifera Abel.,was meticulously extracted and hydrolyzed to yield five known sapogenins:16-O-tiglogycamelliagnin B(a),camelliagnin A(b),16-O-angeloybarringtogenol C(c),theasapogenol E(d),theasapogenol F(e).Subsequent biotransformation of compound a facilitated the isolation of six novel metabolites(a1−a6).The anti-inflammatory potential of these compounds was assessed using pathogenassociated molecular patterns(PAMPs)and damage-associated molecular patterns molecules(DAMPs)-mediated cellular inflammation models.Notably,compounds b and a2 demonstrated significant inhibitory effects on both lipopolysaccharide(LPS)and high-mobility group box 1(HMGB1)-induced inflammation,surpassing the efficacy of the standard anti-inflammatory agent,carbenoxolone.Conversely,compounds d,a3,and a6 selectivity targeted endogenous HMGB1-induced inflammation,showcasing a pronounced specificity.These results underscore the therapeutic promise of C.oleifera seed pomace-derived compounds as potent agents for the management of inflammatory diseases triggered by infections and tissue damage.

INTRODUCTION.In recent years,with the steady development of offshore energy exploration technology,submarine gas seepage sites,and their products are gradually being identified and analyzed.The submarine cold seep is a seepage phenomenon in which hydrocarbon gases stored deep in the seafloor are transported to seawater by gushing or seepage under the action of tectonic compression(Roy et al.,2019;Tinivella and Giustiniani,2016).

Due to its highly insidious and rapid progression,deep tissue pressure injury(DTPI)is a clinical challenge.Our previous study found that DTPI may be a skeletal muscle injury dominated by macrophage immune dysfunction due to excessive iron accu-mulation.Decellularized extracellular matrix(dECM)hydrogel promotes skeletal muscle injury repair.However,its role in po-larizing macrophages and regulating iron metabolism in DTPI remains unclear.Here,porcine dECM hydrogel was prepared,and its therapeutic function and mechanism in repairing DTPI were investigated.The stimulus of dECM hydrogel toward RAW264.7 cells resulted in a significantly higher percentage of CD2o6+macrophages and notably decreased intracellular divalent iron levels.In mice DTPI model,dECM hydrogel treatment promoted M1 to M2 macrophage conversion,improved iron metabolism and reduced oxidative stress in the early stage of DTPI.In the remodeling phase,the dECM hydrogel remarkably enhanced revascularization and accelerated skeletal muscle repair.Furthermore,the immunomodulation of dEcM hydrogels in vivo was mainly involved in the P13k/Akt signaling pathway,as revealed by GO and KEGG pathway analysis,which may ameliorate the iron deposition and promote the healing of DTPI.Our findings indicate that dECM hydrogel is promising in skeletal muscle repair,inflammation resolution and tissue injury healing by effectively restoring macrophage immune homeostasis and normalizing iron metabolism.

Multi-omics analysis reveals the molecular regulatory network underlying the prevention of Lactiplantibacillus plantarum against LPS-induced salpingitis in laying hens

Background Salpingitis is one of the common diseases in laying hen production, which greatly decreases the economic outcome of laying hen farming. Lactiplantibacillus plantarum was effective in preventing local or systemic inflammation, however rare studies were reported on its prevention against salpingitis. This study aimed to investigate the preventive molecular regulatory network of microencapsulated Lactiplantibacillus plantarum(MLP) against salpingitis through multi-omics analysis, including microbiome, transcriptome and metabolome analyses.Results The results revealed that supplementation of MLP in diet significantly alleviated the inflammation and atrophy of uterus caused by lipopolysaccharide(LPS) in hens(P < 0.05). The concentrations of plasma IL-2 and IL-10 in hens of MLP-LPS group were higher than those in hens of LPS-stimulation group(CN-LPS group)(P < 0.05). The expression levels of TLR2, MYD88, NF-κB, COX2, and TNF-α were significantly decreased in the hens fed diet supplemented with MLP and suffered with LPS stimulation(MLP-LPS group) compared with those in the hens of CN-LPS group(P < 0.05). Differentially expressed genes(DEGs) induced by MLP were involved in inflammation, reproduction, and calcium ion transport. At the genus level, the MLP supplementation significantly increased the abundance of Phascolarctobacterium, whereas decreased the abundance of Candidatus_Saccharimonas in LPS challenged hens(P < 0.05). The metabolites altered by dietary supplementation with MLP were mainly involved in galactose, uronic acid, histidine, pyruvate and primary bile acid metabolism. Dietary supplementation with MLP inversely regulates LPSinduced differential metabolites such as Lyso PA(24:0/0:0)(P < 0.05).Conclusions In summary, dietary supplementation with microencapsulated Lactiplantibacillus plantarum prevented salpingitis by modulating the abundances of Candidatus_Saccharimonas, Phascolarctobacterium, Ruminococcus_torques_group and Eubacterium_hallii_group while downregulating the levels of plasma metabol

Preparation of high-purity fluorite and nanoscale calcium carbonate from low-grade fluorite

Flotation separation of calcite from fluorite is a challenge on low-grade fluorite flotation that limits the recovery and purity of fluorite concentrate.A new acid leaching–flotation process for fluorite is proposed in this work.This innovative process raised the fluor-ite’s grade to 97.26wt%while producing nanoscale calcium carbonate from its leachate,which contained plenty of calcium ions.On the production of nanoscale calcium carbonate,the impacts of concentration,temperature,and titration rate were examined.By modifying the process conditions and utilizing crystal conditioning agents,calcite-type and amorphous calcium carbonates with corresponding particle sizes of 1.823 and 1.511μm were produced.The influence of the impurity ions Mn^(2+),Mg^(2+),and Fe^(3+)was demonstrated to reduce the particle size of nanoscale calcium carbonate and make crystal shape easier to manage in the fluorite leach solution system compared with the calcium chloride solution.The combination of the acid leaching–flotation process and the nanoscale calcium carbonate preparation method improved the grade of fluorite while recovering calcite resources,thus presenting a novel idea for the effective and clean usage of low-quality fluorite resources with embedded microfine particles.

Geophysical Evidence for Carbonate Platform Periphery Gravity Flows in the Xisha Islands,South China Sea

High-resolution bathymetric and partial seismic data were utilized to determine the characteristics and distribution of mass-transport deposits(MTDs)in the Xisha area,North of the South China Sea.Many isolated carbonate platforms are found in this area,providing mass-wasting materials as a sediment source for MTDs.The MTDs in the study area were divided into three types(slide,slump,and debris flow)based on their rheological properties.The MTDs were mostly distributed at the toe of the carbonate platform slope,where a large amount of carbonate debris accumulated.A total of 32 MTDs were identified on the carbonate platform slopes.A systematic assessment of the MTD morphometric parameters showed that their total area covered 2225.2 km2,with the largest MTD covering 354.1 km2.These MTDs differ largely from the MTDs of other areas,especially in terms of sediment compositions and deposit processes.Gullies and channels on the slope and reefs on the top of the platform act as sediment conduits and carbonate-producing sources,respectively,playing vital roles in the distribution and generation of gravity flow deposits.The development model proposed in this work states that reefs disintegrate into carbonate debris and mix with seawater to form gravity flows,which scour the seafloor and accumulate at the downslope,eventually resulting in failure.Sediments from these failures turn into gravity flows,flowing into submarine canyons and channels.The results of this research further our understanding of the development pattern of MTDs in a carbonate setting.

Substrate Effect on the Structural and Electrical Properties of LaNiO_(3) Thin Films

Epitaxial LaNiO_(3)(LNO)thin films prepared from the sols modified with polyethyleneimine(PEI)were grown on single-crystal LaAlO_(3),(LaAlO_(3))_(0.3)(SrAlTaO_(6))_(0.7),and SrTiO_(3) substrates,respectively,using a simple polymer assisted deposition(PAD).The epitaxial structure,surface morphologies and transport of the LNO films were studied by X-ray diffraction(θ/2θ symmetric scan,ω-scan,and in-planeφ-scan),the field emission scanning electron microscopy,and a standard dc four-probe method.It is found that,compared with that of LNO bulk,the c-axis parameter of the LNO film increases under compressive strain and decreases under tensile strain.All the LNO films exhibit metal properties in the temperature-dependent resistivity.The resistivity of the LNO films shows an increasing trend with the lattice mismatch strain changing from compressive to tensile.It is suggested that the oxygen vacancy compensated by more Ni^(2+)changed from Ni^(3+)in the film increases with the strain changing from compressive to tensile,which results in the increase of the resistivity.

Identification and Characterization of Long Non-Coding RNAs Involved in Sex-Related Gene Regulation in Kelp Saccharina japonica

Long non-coding RNAs(lncRNAs)regulate a variety of biological processes,including sexual reproduction and differentiation.Saccharina japonica,a commercially important brown alga in China,shows remarkable sexual dimorphism in haploid gametophytes.The sex of Saccharina japonica gametophytes is determined by UV sexual system.However,no results have been reported on the lncRNAs involved in the sex-related gene regulation of S.japonica.This study identified a number of lncRNAs and assessed their expression levels in male and female gametophytes.Among them,a total of 405 lncRNAs and 211 mRNAs showed differential expressions.Furthermore,the functions of target genes of differentially expressed lncRNAs(DELs)differed from those of differentially expressed genes(DEGs),suggesting that lncRNA may interact with other functional proteins,in addition to DEGs,to involve sex regulation in S.japonica.There were 32 and 90 potential cis-regulatory and trans-regulatory interactions between DELsDEGs,respectively.Five of these lncRNAs(LNC_002974,LNC_021059,LNC_038466,LNC_051584,and LNC_027400)interacted with putative male sex determination region(SDR)genes,suggesting that they act as regulators in gametophytes'sex regulation potentially.Findings from this study contribute to our understanding of the roles of lncRNAs in sex differentiation and lay the foundation for functional studies of candidate lncRNAs in the future.

新中国成立前后,跨海解放东南海岛成为中共重要工作。为此,中共和解放军需应对诸多环境挑战,包括跨越汪洋大海险阻,应对风浪、潮汐袭扰,适应海岛生存环境。为应对这些挑战,中共开展大规模民间船工动员工作,但在此过程中也遇到不少具体难题,如沿海船工和船只因战争遭受严重损失,船工和船只动员、运用殊为不易,木帆船作战稳定性不强等。为解决这些问题,中共一方面完善支前机构、注重思想动员、保障基本权益等,另一方面着力训练军队水手、赶制机帆船、加强海洋环境知识学习、提升海岛后勤保障,从而为继续推进岛屿的解放创造基础。

“形”“心”“神”是庄子哲学乃至道家哲学的重要概念,关涉人的生命构成问题。庄子学派对生命构成的看法复杂、多样。《庄子》内篇对于生命的理解就是“形”和“身”,“心”为形体的一部分,“精”“神”为形体的要素。而在外杂篇中,“形”只是生命的一部分成为比较普遍的认识,“神”(或精、精神)与“形”并立而形成生命整体。这一变化之所以发生,有其思想背景,也是思想发展逻辑的需要。“心”作为“形”的一部分,无法彻底摆脱“形”的束缚,“神”取代“心”与“形”并立可以解决这一问题;宗教意味的“神”观念由外向内转化,需要在人的生命中得到安置,战国时期思想界对“心”的精神性的阐发、“心”为“形”之主的观念的成熟,为“神”的进入做好了理论准备。
