Glutamate oxidase-integrated biomimetic Fe3O4 hybrids as cascade nanozymes for glutamate detection

Herein,a new nanozyme/natural enzyme hybrid biosensor was established for ultrasensitive l-glutamic acid(L-Glu)detection.The Fe3 O4 nanoparticles with peroxidase-like activity and stability was used as a nanozyme and carrier for immobilizing l-Glutamate oxidase(GLOD)through Schiffff base reaction to construct a chem-enzyme cascade detector.The resultant Fe3 O4-GOLD exhibited a wide linear range(10−500μM)and a low detection limit of 6.03μM for L-Glu detection.Furthermore,the Fe3 O4-GOLD exhibited excellent pH stability,thermal stability,reusability and storage stability.After repeated nine cycles,Fe3 O4-GOLD still retained 70%of its initial activity.Meanwhile,Fe3 O4-GOLD maintained 50%of its initial activity after storage for 20 days,while free GLOD only retained 20%of its initial activity.This strategy of integrating biomimetic Fe 3 O4 and natural enzymes for cascade catalysis makes it possible to design an efficient and stable chemo-enzyme composite catalysts,which are promising for applications in biosensing and biomimetic catalysis.

Physical Modeling of Fold-and-Thrust Belt Evolution and Triangle Zone Development:Dabashan Foreland Belt(Northeast Sichuan basin,China) as an Example

Triangle zones, generally found in foreland fold-and-thrust belts, serve as favorable objects of petroleum exploration. Taking the Dabashan foreland belt as an example, we studied the formation and development of triangle zones, and investigated the effect of d^collements and the mechanical contrast of lithology by employing the method of physical modeling. Four experimental models were conducted in the work. The results showed that ＇sand wedges＇ grew episodically, recorded by deformational length, height and slope angle. The height versus shortening rate presented an S-shape curve, and uplifting occurred successively in the direction of the foreland belt. During the formation of the triangle zone, layer-parallel shortening took place at the outset; deformation decoupling then occurred between the upper and lower brittle layers, divided by a middle-embedded silicone polymers layer. The upper brittle layers deformed mainly by folding, while the lower sand layers by thrusting. As shortening continued, the geometry of a triangle zone was altered. We consider that the triangle zone in the Dabashan foreland belt was modified from an early one based on available seismic profiles and the experimental results. In addition, dccollements and mechanical contrast impose significant influence on structural development, which can directly give rise to structural discrepancies. More d^collements and obvious mechanical contrast between brittle layers can promote the coupling between the upper and lower brittle layers. Basal d^collement controls the whole deformation and decreases the slope angle of the wedge, while roof d^collement determines whether a triangle zone can be formed.

Homologous feature point extraction is a key problem in the optical and synthetic aperture radar （SAR） image registration for islands. A new feature point extraction method using a threshold shrink operator and non-subsampled wavelet transform （TSO-NSWT） for optical and SAR image registration was proposed. Moreover, the matching for this proposed feature was different from the traditional feature matching strategies and was performed using a similarity measure computed from neighborhood circles in low-frequency bands. Then, a number of reliably matched couples with even distributions were obtained, which assured the accuracy of the registration. Application of the proposed algorithm to SPOT-5 （multi-spectral） and YG-1 （SAR） images showed that a large number of accurately matched couples could be identified. Additionally, both of the root mean square error （RMSE） patterns of the registration parameters computed based on the TSO-NSWT algorithm and traditional NSWT algorithm were analyzed and compared, which further demonstrated the effectiveness of the proposed algorithm. The algorithm can supply the crucial step for island image registration and island recognition.

Effects of Combined Stress from Salt and Herbicide on Microbial Remediation of Soil Contaminated by n-Hexadecane and Phenanthrene

In this study,physicochemical,cluster,and high-throughput sequencing analyses were used to investigate the joint effects of salt and herbicide(glyphosate)stress factors on the microbial remediation of soil contaminated by n-hexadecane and phenanthrene.Based on the soil’s physical and chemical properties,differences in microbial quantity and enzyme activities were analyzed among the samples,and how that influenced distribution of soil community structure was focused upon.After 120 days of indoor simulated remediation,the cumulative degradation rate of n-hexadecane decreased by 94.92%(blank control),96.96%(low concentration salt and glyphosate stress group),65.07%(high concentration salt and glyphosate stress group),while that of phenanthrene decreased by 87.33%(blank control),86.25%(low concentration salt and glyphosate stress group),58.45%(high concentration salt and glyphosate stress group).The combined stress of salt and herbicides was capable of lowering the reduction efficiency of organic matter,total nitrogen,and total phosphorus,and also restricting the growth of microorganisms and enzyme activities.Cluster analysis results indicated that the non-stress group was similar to the low-concentration compound stress group during different remedial period,whereas both of those differed starkly from the high-concentration compound stress group.High-throughput sequencing revealed that the dominant soil bacteria phyla shifted from Firmicutes to Actinobacteria within 120 days of remediation.

OBJECTIVE: To investigate the effects of Gubi prescription on the expression of caveolin-1, and the phosphoinositide 3 kinase/protein kinase B(PI3 K/Akt) and Fas signal pathways in rats with knee osteoarthritis(KOA).METHODS: Forty KOA model rats were established using a modification of Hulth's method. Rats were divided into five groups by the random number method: model, positive drug(Vicolli group), and high-, medium-, and low-dose Gubi prescription groups(n = 8/group). In the sham surgery group(n = 8), only anterior and posterior cruciate ligaments of rats were exposed during surgery. A normal group(n = 8) consisted of rats with no treatment. Rats were intragastrically administered corresponding drugs once every day for eight consecutive weeks. Then, rat synovial membranes were extracted and histomorphological changes were recorded. m RNA expression was measured by q-PCR.Serum superoxide dismutase(SOD), malondialdehyde(MDA), nitric oxide(NO), and interleukin 1β(IL-1β) levels were measured. Western blotting determined the effects of Gubi prescription on protein expressions of caveolin-1, Bax, Bcl-2, Fas, and caspase-3 in chondrocytes from KOA rats. The knee cartilage of rats was excised and cultured under aseptic conditions. After coincubation of chondrocytes with Gubi prescription-containing serum, IL-1β, and si RNA, Western blotting was used to determine the protein expressions of caveolin-1, Bax, Bcl-2, Fas, and caspase-3.RESULTS: The morphological score of the articular synovium in the model group was significantly higher than in the normal group(P < 0.01). The morphological score in the high-and mediumdose Gubi prescription groups was lower than in the model group(P < 0.05). Chondrocytes from the decoction-containing serum group had a lower expression of Bax(P < 0.05), and higher expressions of Bcl-2(P < 0.05) and caspase-3(P < 0.05) compared with the model group. 目的了解糖尿病足截肢患者创伤后成长现状,探讨其影响因素。方法采用便利抽样法选择2021年5-12月天津市2所三级甲等医院319例已行糖尿病足截肢术的住院患者为研究对象,采用一般人口学资料与疾病资料调查表、疾病不确定感量表、家庭关怀度量表、医学应对方式量表和创伤后成长量表对其进行调查。结果糖尿病足截肢患者的创伤后成长得分(64.29±15.99)分,疾病不确定感、应对方式(面对)、家庭关怀度、家庭人均月收入、身体质量指数(>29.9 kg/m^(2))、职业(个体)、截肢次数(2~3次)、应对方式(回避)、糖尿病足病程(>3~6月)是糖尿病足截肢患者创伤后成长的影响因素(均P<0.05),共解释创伤后成长总变异的47.50%。结论糖尿病足截肢患者的创伤后成长处于中等水平,创伤后成长受多个因素影响;护理人员应采取个性化措施,进一步帮助患者获得创伤后成长。

针对现有的机载数据单木分割方法对林型的普适度不高,尤其在高郁闭度阔叶林地带提取精度偏低的问题,选用海南省海口市热带阔叶林地带的光谱影像和LiDAR数据,先采用基于距离阈值的单木分割方法,利用高分光谱影像分割得到的树冠边缘,对初始探测树顶点进行位置约束。获得单木顶点的精确定位后,采用基于种子点的单木分割方法分割,完成了阔叶林的单木提取。结果显示,与已有的基于单木间相对间距单木分割方法相比,本研究通过选取最佳分割尺度结合光谱影像进行精确定位,改善了原有单一尺度分割方法导致的过分割现象,将单木识别精确率由0.67提升至0.92。该方法在使用遥感对森林单木进行分割工作中,可以更好地识别单木,对不同林型适用度较高,可以为后续的单木信息提取工作提供数据基础。

采用室内培养皿发芽法,研究外源添加聚乙二醇、碳酸钠、碳酸氢钠对花楸胚胎萌发及幼苗生理指标的影响。结果表明,聚乙二醇处理花楸胚胎后使萌发率提高15%~25%,碳酸钠、碳酸氢钠处理使花楸胚胎的萌发率分别降低了88%和85%。聚乙二醇可缓解盐碱胁迫对花楸胚胎萌发的抑制,使胁迫下胚胎的萌发率提高1~2倍。聚乙二醇可提高盐碱胁迫下花楸胚胎中抗氧化酶活性,使胁迫下胚胎中超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶活性(CAT)分别提高16.45%、75%、80%。聚乙二醇使胁迫下胚胎中的丙二醛(MDA)含量降低41.44%。研究显示,聚乙二醇能够有效缓解盐碱胁迫对花楸胚胎的抑制作用,并提高花楸胚胎中的抗氧化酶活性,增强花楸的抗逆性,为建立PEG引发促进花楸胚胎萌发技术提供科学依据。
