Molecular Targets of Combined Natural Antioxidants Against Diabetes in Streptozotocin-induced Diabetic Mice

Natural antioxidants, due to the different chemical structures and their unique biological protection effects on islet cell, have been received much attention in applied chemistry field for developing a new diabetic drug. However, the molecular targets of the islet cell protection have been remained obscure. For investigating molecular targets of combined natural antioxidant’s（CNA） roles in against diabetes, type 1.diabetic mouse model was made by multiple injection of low doses of streptozotocin into C57BL mice. Two combinations of natural antioxidants were orally supplemented to the mice. Blood glucose level, glutathione peroxidase（GSH-Px） and superoxide dismutase （SOD） activities, and malondialdehyde（MDA） contents were measured via biochemical assays. Tumor necrosis factor α（TNF-α）, interferon γ（IFN-γ） and Interleukin-1.（IL-1.） in CD4＋ Peripheral blood mononuclear cells（PBMC） were evaluated with flow cytometry（FCM）. In situ hybridization（ISH） and immunohistochemistry（IHC） were used to detect insulin gene expression. The results demonstrate both the combinations significantly： increased GSH-px and SOD, but decreased MDA levels in pancreas（p〈0.05―0.01.; up-regulated insulin expression at both mRNA and protein levels in islet cells（p〈0.05―0.01.; down-regulated TNF-α and IL-1. expression in PBMC（p〈0.05―0.01.. Our data suggest that the CAN may regulate multiple key molecular targets related to β cell function, including reduction of oxidative stress and pre-inflammatory cytokines, and upregulate insulin gene expression to reduce the development of diabetes.

Melittin,a classical antimicrobial peptide,is a highly potent antitumor ***,its significant toxicity seriously hampers its application in tumor *** this study,we developed novel melittin analogs with pH-responsive,cell-penetrating and membranelytic activities by replacing arginine and lysine with *** conjugation with camptothecin(CPT),CPT-AAM-1.and CPT-AAM-2.were capable of killing tumor cells by releasing CPT at low concentrations and disrupting cell membranes at high concentrations under acidic ***,we found that the C-terminus of the melittin analogs was more suitable for drug conjugation than the ***-AAM-1.significantly suppressed melanoma growth in vivo with relatively low ***,the present study demonstrates that the development of antitumor drugs based on pH-responsive antimicrobial peptide-drug conjugates is a promising strategy.

The Songshugang deposit is a large Ta-Nb deposit in South china,with Ta-Nb mineralization associated genetically with the granite and pegmatite.A diversity of quartz from topaz-albite granite,quartz-mica pegmatite,quartz-feldspar pegmatite,and quartz-fluorite pegmatite at Songshugang was studied by CL and LA-ICP-MS in order to constrain enrichment mechanisms of Nb and Ta and to find geochemical indicators of quartz for rare metal *** image illuminates a canvas of complexity,the quartz from topaz-albite granite,quartz-mica pegmatite,quartz-feldspar pegmatite,and quartz-fluorite pegmatite,exhibits numerous dark CL streaks,patches,and a series of healed *** textures suggest that the rocks were fractured because of deep crustal pressure,and underwent later hydrothermal metasomatism and quartz *** quartz from quartz-fluorite pegmatite present limited patches or fractures but distinct growth bands,indicating that the melt fluid composition during the formation of quartz at this stage varies greatly and is less aff ected by mechanical *** LA-ICP-MS analysis of quartz shows that there is a positive correlation between Al and Li in the quartz from topaz-albite granite,quartz-mica pegmatite,quartz-feldspar pegmatite,to quartz-fluorite pegmatite,indicating that Al mainly enters the quartz lattice through charge compensation substitution mechanism with ***,our data deviate from the theoretical Li:Al mass ratio of~1.3.89 in quartz,indicating that there may be competition between H^(+)and Li in a water-rich magmatic *** quartz from topaz-albite granite is enriched in K and Na elements,and the quartz from quartz-fluorite pegmatite is enriched in fluorite with a low Ca content in quartz,further elucidating that these rocks were subjected to hydrothermal *** topaz-albitite granite to quartz-fluorite pegmatite,Al,Li and Ge content and Al/Ti,Ge/Ti,Sb/Ti ratios in quartz gradually increased,but

A two-chamber microbial fuel cell(MFC)with algal-film cathode was *** showed good electric-generating performance with three electric-generating stages:start-up,development,and *** average output voltage reached~0.41. 抗草甘膦转基因油菜和野芥菜回交1.子2.-子6代在无除草剂选择压下的适合度转基因作物的抗性基因能否成功渗入野生或近缘杂草取决于携带转基因回交后代的适合度。为深入了解转基因油菜(Brassica napus)向野芥菜(wild ***)基因流动的生态风险,本文研究了在低密度(5株/m^(2.)和高密度(1.株/m^(2.)下,以及单种和混合种植(野芥菜:BC1.2.-BC1.6R=1.1.下抗草甘膦转基因油菜与野芥菜回交1.子2.到子6代(BC1.2.-BC1.6R)的适合度;并分析回交后代的适合度成分、种植密度和种植方式之间的相关性。研究结果表明:在单种低密度下,与野芥菜相比,回交1.子2.(BC1.2.)和子3代(BC1.3R)具有较低的适合度,而回交1.子4代-子6代(BC1.4R-BC1.6R)具有较高的适合度;无论是单种还是混种,高密度下所有回交后代的适合度均低于野芥菜。相关性分析结果显示,种植密度和种植方式对回交后代(BC1.2.-BC1.6R)的适合度成分有显著影响。这些结果表明,转基因从栽培油菜渗入到野芥菜很大程度上取决于回交后代的特定生长环境。
