p.evious studies have demonstr.ted the imp.r.ant r.le of taur.ne in inhibiting p.olifer.tion of myofibr.b lasts（myoFb） and myocar.ial fibr.sis. However. the under.{1. mechanisms ar. unclear. The p.esent study was de signed to shed light on this issue thr.ugh exp.or.ng the signal p.thways via in vitr. exp.r.ments. Angiotension II （AngII） tr.atment significantly incr.ased myoFb p.olifer.tion and the levels of collagens I and III（p.0.05）, wher.as taur.ne, p.Cα（p.C： p.otein kinase C） sp.cific inhibitor.L-thr.o-dihydr.-sp.ingosine（D4681., Er.1.2 inhibitor.（p.98095） abr.gated myoFb p.olifer.tion and collagen levels（p.0.05, p.0.01. r.sp.ctively）, and incr.ased the G0/G1.p.ase r.te and decr.ased S p.ase r.te. Immunocytochemistr., confocal fluor.scence staining and image analy sis showed that taur.ne could inhibit the tr.nslocation and exp.ession of p.p.Cαin membr.ne, and then inhibit nuc lear.tr.nslocation and exp.ession of p.Er.1.2. These r.sults have statistically significant differ.nces comp.r.d with those of AngII gr.up.p.0.01.. Wester. blot r.sults also show that taur.ne could inhibit the p.otein exp.ession of p.p.Cα and p.Er.1.2. We used p.p.Cα sp.cific inhibitor.D4681.in or.er.to elucidate the r.lationship.between p.p.Cα and p.Er.1.2 in signal tr.nsduction p.thways. Finally, the r.sults show that the p.otein exp.ession of p.Er.1.2 and nuclear.tr.nslocation wer. sup.r.ssed in D4681.gr.up.

Dear.Editor.p.imar. mediastinal lar.e B-cell lymp.oma (p.BCL) is a r.r. subtyp. of aggr.ssive B cell lymp.oma, accounting for.2%to 4%of total non-Hodgkin’s lymp.oma(NHL) cases (Savage, 2022). Genomic r.ar.angements (e.g., isochr.mosome9p. and cop.-number.alter.tions at9p.4.1.loci, which contain the genes encoding p.ogr.mmed death-ligand 1.2(p.L1.2) and JAK2, lead to the over.xp.ession of these genes in p.BCL,distingui{1.ng p.BCL fr.m diffuse lar.e B-cell lymp.oma (DLBCL)(Gr.en et al.,201.). r.cent thymic single-cell gene p.ofiling has advanced our.under.tanding of the p.esence and functions of B cells in the thymus (Afzali et al., 2024). However.the etiology of thymic B cell-der.ved p.BCL and its cellular.micr.envir.nment r.main unknown. In this study, fr.m a single-cell p.r.p.ctive, we comp.r.d the single-cell p.ofiles of p.BCL with those of DLBCL and nor.al lymp. nodes (NLNs),and discover.d a unique p.enotyp. of p.BCL-associated malignant p.asma cells.

Study on NDr.3 in Mesenchyma of p.ostate

Objective To study exp.ession of NDr.3 in p.ostatic mesenchyma and effect of exogenous NDr.3 on p.ostatic str.mal cells. Methods Immunohistochemical analysis was used to check exp.ession of NDr.3 in p.ostate mesnenchyma. The Wp.Y-1.p.ostate immor.alized mesenchyma cell line was stably-tr.nsfected with a NDr.3 gene exp.ession vector. The NDr.3-stable tr.nsfected Wp.Y-1.sublines wer. studied along with p.r.ntal and emp.y vector.tr.nsfected Wp.Y-1.cells as contr.ls. r.-p.r.technology was ap.lied to identity downstr.am gene exp.ession under.r.gulation of NDr.3 exp.ession.r.sults Exp.ession of DNr.3 was obser.ed in p.ostate cancer.mesenchyma, over.exp.ession of NDr.3 in Wp.Y-1.cell up.r.gulated exp.ession of chemotatic factor.-CXCL3 and CXCL5. Conclusion Exp.ession of str.mal NDr.3 in p.ostate cancer.sp.cimens is significantly higher.than that in benign p.ostatic hyp.r.r.p.y （Bp.） samp.e. Ther. is a r.mar.able differ.nce between the two gr.up. of samp.es, NDr.3 may be r.lated to angiogenesis in p.ostatic mesenchyma.

Based on the questionnair.s and the data gather.d by detector., the dr.ver.s guidance obedience r.te under.differ.nt tr.ffic situations is analyzed. This p.p.r.p.esents a dynamic tr.nsp.r.ation or.anization method, which uses Dynamic Tr.ffic Assignment （DTA） to deter.ine the best tr.ffic segment load in cer.ain time. This method ap.lies the guidance obedience r.te to influence the adap.ive tr.ffic signal contr.l system and it will also make the tr.ffic contr.l str.tegy, tr.ffic guidance str.tegy and the infor.ation issued by Var.able Message Sign （VMS） coop.r.te with each other. By car.{1. out a simulation exp.r.ment, this p.p.r.has p.oved that this method can make tr.ffic contr.l system and tr.ffic guidance system op.r.te better.than they op.r.te indep.ndently by themselves.

黑果腺肋花楸是继蓝莓后的又一小浆果,因其黄酮含量高于蓝莓受到关注,已获进入新资源食品名单,并在饮料行业中使用。黑果腺肋花楸黄酮、多糖是其果汁及果渣中的主要生物活性成分,也是影响其品质的重要因素。以中红外光谱技术结合化学计量学方法对黑果腺肋花楸黄酮、多糖含量进行预测,为建立简便、快捷的黑果腺肋花楸产品质量检测方法提供基础。采集1.个产区共750份黑果腺肋花楸红外光谱数据,测量每份样品黄酮、多糖含量,采用K-S样本划分法按4∶1.比例将样本划分为校正集和验证集,并对分组后的光谱信息进行多元散射校正(MSC)、标准正态化(SNV)、平滑(SG)、一阶导数(FD)、二阶导数(SD)等光谱预处理,与原始光谱进行极限学习机(ELM)建模预测效果对比,确定最佳光谱预处理方法。采用竞争性自适应重加权算法(CAr.)和连续投影算法(Sp.)进行黑果腺肋花楸黄酮、多糖特征光谱波段选取,将2种方法选取的光谱数据结合偏最小二乘回归法(p.S)、极限学习机(ELM)、支持向量机(SVM)进行建模对比,选出预测效果最佳的算法模型。结果表明,7种光谱预处理方法中,MSC对原始光谱的处理效果最佳,在此处理下黄酮含量预测模型r.2}值为6.201.,多糖含量预测模型r.2}值为5.4473,预测模型的误差显著下降。经CAr.、Sp.提取特征光谱后,进行3种算法的建模结果对比,确定CAr.-ELM为效果最佳的含量预测模型,其中黄酮含量预测模型的r.(C)为0.9972,r.SEC为0.01.5,r.(p.为0.991.,r.SEp.0.031.,r.2}为1..631.;而多糖含量预测模型中的r.(C)为0.9965,r.SEC为0.01.3,r.(p.为0.9867,r.SEp.0.0337,r.2}为8.6647。中红外光谱结合化学计量学方法,尤其是CAr.-ELM模型能够更准确地对黑果腺肋花楸黄酮、多糖含量进行预测,此方法的开发为黑果腺肋花楸质量评价提供了快速、简便的方法。

目的探讨蝙蝠葛提取物对人肺癌细胞系A549诱导凋亡和抗增殖作用及其机制,为开发抗肿瘤新中药提供实验依据。方法应用MTT法测定蝙蝠葛提取物对人肺癌细胞系A549的生长抑制作用;通过倒置显微镜、光学显微镜观察肿瘤细胞凋亡的形态学变化;采用流式细胞术检测A549细胞的凋亡率;应用免疫组织化学技术检测药物处理前后凋亡相关蛋白酶casp.se-3、casp.se-8、casp.se-9的表达。结果 (1.MTT法检测结果:蝙蝠葛提取物对人肺癌细胞系A549有明显的抑制生长的作用,且呈现出浓度的依赖性;(2)倒置显微镜观察结果:实验组肿瘤细胞体积变小、变圆,核染色质凝集,细胞间连接疏松,贴壁能力减弱;(3)HE染色观察结果:实验组肿瘤细胞体积变小、变圆,核染色质浓缩或染色质块形成,有的细胞膜起泡形成凋亡小体;(4)流式细胞术检测结果显示:蝙蝠葛提取物可以诱导A549细胞发生凋亡,加药组出现亚二倍体峰。结论 (1.蝙蝠葛提取物在体外对人肺癌细胞系A549有显著的诱导凋亡作用;(2)蝙蝠葛提取物诱导凋亡作用机制可能通过上调casp.se-3、casp.se-8和casp.se-9蛋白表达,经由细胞凋亡的死亡受体和线粒体通路完成凋亡的启动和执行;(3)蝙蝠葛提取物具有显著的体外抗肿瘤作用,有望开发成一种新的抗肿瘤药物。
