AIM: To investigate the inhibitory effect of in vivoexpression of expressing plasmid pCH510 of recombinant fibronectin polypeptide (CH50) on hepatocellular carcinoma and the improved therapeutic effect of pCH510 in combination with chemotherapeutic agents and Hsp70-H22 hepatocarcinoma antigen peptide on ***: Mice were inoculated with H22 hepatccarcinoma cells. The chemotactic effect of the expression of plasmid pCH510 on immunocytes was observed after in vivo transfection, tissue slicing and HE staining. Inhibitory effect of transfection with pCH510 on routine tumor originatedfrom different inoculative doses was observed. The inhibitory effect of immediate transfection with pCH510 after chemotherapy on tumor was compared with that of transfection 5 days after chemotherapy. The change of function and amount of mouse peritoneal macrophages and the peripheral blood immunocytes resulted from administration of chemotherapeutic agents were detected. The peptides mixture was prepared from H22 hepatocarcinoma cells, pCH510 + Hsp70-H22 antigen peptides were injected into tumor-bearing mice with or without chemotherapy, to observe the inhibitory effects on ***: At the tumor tissue site injected with pCH510,there were a great number of immunocytes which mainly were macrophages, lymphocytes and *** of plasmid pCH510 inhibited significantly the murine tumor induced by different inoculative doses. The inhibitory effect was negatively correlated with the inoculative dose. The therapeutic effect was not improved by immediate transfection with pCH510 after chemotherapy, but was significantly improved by transfection with pCH510 5 days after chemotherapy. Chemotherapeutic agent decreased the number of immunocytes and suppressed their activation in vivo. After injection of drug, the amount of immunocytes was the lowest from d 1 to d 3 and returned to normal level on the 10th day. Transfection with plasmid pCH510 alone could inhibit tumor induce

PD L PD 1是参与肿瘤免疫逃避的一条抑制性信号途径。为了用可溶性的PD 1受体阻断PD L PD 1的相互作用 ,将小鼠PD 1胞外段 (aa1 aa16 7)作为独立可溶性分子进行了真核表达 ,并对其功能进行鉴定。构建了编码小鼠PD 1胞外段cDNA(sPD 1)的真核质粒表达载体pPD 1A和编码sPD 1 GFP重组蛋白的真核质粒表达载体pPD 1B ;细胞转染实验表明其表达产物主要是分泌到细胞外的可溶性产物 (sPD 1) ,流式细胞仪检测表明sPD 1可有效结合PD 1配体 ;肿瘤细胞杀伤实验表明 ,sPD 1作用于肿瘤细胞或在脾淋巴细胞激活过程中作用于淋巴细胞 ,均可增强Hsp70 H2 2抗原肽复合物激活的脾淋巴细胞杀伤肿瘤细胞的作用。sPD 1真核表达载体的构建为在肿瘤局部表达抑制性共刺激分子的可溶性受体 ,拮抗肿瘤微环境中负调节因素对T细胞的抑制作用 ,增强机体的抗肿瘤能力。

多巴胺能正电子发射断层扫描(PET)脑显像是目前诊断帕金森病的重要依据,但其对帕金森综合征的鉴别能力有限。葡萄糖代谢PET显像中不同的脑代谢模式为进一步鉴别诊断提供可能。文中在前期PET脑显像相关技术操作和临床应用专家共识的基础上,对开展多巴胺能联合葡萄糖代谢显像进行系统梳理,征求专家意见后形成本专家共识。旨在规范帕金森综合征患者的多巴胺能显像及葡萄糖代谢显像的技术操作,为医生提供报告解读建议,进一步指导多巴胺能与葡萄糖代谢PET脑显像联合应用在帕金森相关疾病中的临床实践流程。

Immune cells play key roles in cancer and chronic inflammatory disease. A better understanding of the mechanisms and risks will help develop novel target therapies. At the 2017 International Workshop of the Chinese Academy of medical sciences Initiative for Innovative Medicine on Tumor Immunology held in Beijing, china, on May 12, 2017, a number of speakers reported new findings and ongoing studies on immune-related diseases such as cancer, fibrotic disease, diabetes, and others. A considerably insightful overview was provided on cancer immunity, tumor microenvironments,and new immunotherapy for cancer. In addition, chronic inflammatory diseases were discussed. These findings may offer new insights into targeted immunotherapy.

背景:骨形态发生蛋白2(bone morphogenetic protein2,BMP-2)是已知的所有生长因子中对骨的形成作用最强的生长因子,被认为是最具有前途的骨诱导物质。目的:构建人骨形成蛋白2真核表达载体并观察其体外表达情况。设计、时间及地点:自身对照实验,于2005-07/2006-05在华中科技大学同济医学院分子生物中心实验室完成。材料:pcDNA3.1(+)载体由华中科技大学同济医学院左石博士惠赠;成骨肉瘤组织由华中科技大学同济医学院附属协和医院骨科提供。方法:从人成骨肉瘤细胞中提取细胞总RNA,利用反转录-聚合酶链反应方法扩增获得人BMP-2基因cDNA,将基因片断重组到pGEM-T质粒中构建pGEM-T-hBMP-2重组质粒载体,转化到大肠杆菌DH5α后筛选阳性克隆,利用限制性酶切和DNA序列分析鉴定重组质粒。分别用RcoRI和NotI双酶切pGEM-T-hBMP-2质粒和pcDNA3.1真核表达载体,将克隆载体中人骨形成蛋白2基因重组到pcDNA3.1真核表达载体,提取质粒作酶切电泳、聚合酶链反应鉴定及DNA测序后,用脂质体体外转染小鼠骨髓基质细胞,反转录-聚合酶链反应检测BMP-2的表达。主要观察指标:①人骨肉瘤细胞总RNA反转录-聚合酶链反应结果。②重组质粒pGEM-T-hBMP-2和pcDNA3.1-hBMP-2的构建和酶切鉴定。③BMP-2在小鼠骨髓基质细胞内的表达。结果:人骨肉瘤细胞总RNA经反转录-聚合酶链反应扩增后,获得1.2kb条带。经酶切电泳、聚合酶链反应鉴定及DNA测序证实实验成功克隆BMP-2基因,重组质粒pcDNA3.1-hBMP-2构建正确;该重组质粒能在体外培养的小鼠骨髓基质细胞中有效表达BMP-2。结论:实验成功克隆人骨形成蛋白2基因并构建了此基因的真核表达载体。

To assess the peripheral refractive changes that occur when eye growth decelerates in *** We measured peripheral refraction and on-axis vitreous chamber depth on a total of 53 marmosets;36 treated monocularly for 12 weeks 9 hrs/day(26 with-5 D contact lenses,10 with+5 D)and 17 untreated *** the 26 marmosets treated with-5 D,10 wore contact lenses 9 hrs/day without interruptions and were mea-sured at the end of 10 weeks of treatment(T10)and after 4 weeks of recovery(R14).Eight had both contact lenses removed for 30 mins twice/day(mid-morning and mid-afternoon)during the first four weeks of treatment(early interruption group)and eight had the lenses removed during the second four weeks of treatment(late interruption group).The two interruption groups were measured after 4,8 and 12 weeks of treatment only(T4,T8,T12).
