Endometriosis （ EM ） as a common and intractable gynecological disease is characterized by unknown etiology and complex pathologic changes. Many factors of the disease are uncertain at the molecular level and it is difficult to study clinically. In this study, we attempted to establish a nude mice model of EM for dynamical observation of the genesis and development of the disease, morphological changes in tissue, and biological behaviors.

目的:探讨芳香化酶在子宫内膜异位症(EMs)在位内膜及其裸鼠模型异位病灶中的表达意义。方法:征集EMs组及非异位症(NEMs)组各24例,应用其分泌晚期子宫内膜,建立EMs裸鼠模型,取建模后5d、15d、30d裸鼠异位病灶,RT-PCR及免疫组化方法检测两组在位内膜和异位病灶芳香化酶mRNA及蛋白表达。结果:芳香化酶mRNA表达,EMs组异位病灶(0.894±0.23)表达强于在位内膜(0.685±0.15),P<0.05;阳性表达率(87.50%,83.33%)无差异。NEMs组异位病灶表达强度和阳性表达率(0.920±0.24,95.83%)均高于在位内膜(0.612±0.15,62.50%),P<0.05。两组在位内膜比较,EMs组阳性表达率高于NEMs组,P<0.05。两组异位病灶及各组不同时期异位病灶(EMs组,5d:0.889±0.23;15d:0.990±0.19;30d:0.880±0.29;NEMs组,5d:0.889±0.23;15d:0.905±0.23;30d:0.918±0.22)比较,芳香化酶mRNA表达无差异。芳香化酶蛋白阳性表达率,EMs组异位病灶(95.83%)与在位内膜(91.67%)比较,差异不显著。NEMs组异位病灶(95.83%)高于在位内膜(70.83%),P<0.05。EMs组在位内膜高于NEMs组,P<0.05;两组异位病灶比较,差异不显著。结论:局部雌激素合成增强在EMs发病机理中起重要作用。芳香化酶是EMs等雌激素依赖性疾病在子宫内膜特异性表达标记物。

Intracellular pH plays a critical role in biological functions,and abnormal pH values are related to various ***,we report on an intracellular pH sensor AgInS_(2)(AIS)/ZnS quantum dots(QDs)that show long fluorescence lifetimes of hundreds of nanoseconds and low *** lifetime imaging microscopy(FLIM)combined with AIS/ZnS QDs is used for the imaging of live cells in different pH buffers and different cell *** FLIM images of AIS/ZnS QDs in live cells demonstrate different intracellular pH values in different regions,such as in lysosomes or *** method can also distinguish cancer cells from normal cells,and the fluorescence lifetime difference of the AIS/ZnS QDs between the two types of cells is 100±7 *** importantly,the exfoliated cervical cells from 20 patients are investigated using FLIM combined with AIS/ZnS *** lifetime difference value between the normal and cervical cancer(CC)groups is 115±9 ns,and the difference between the normal and the precancerous lesion group is 64±9 *** the first time,the noninvasive method has been used for cervical cancer screening,and it has shown great improvement in sensitivity compared with a clinical conventional cytology examination.

Objectives:To develop a new Chinese medicine(CM)-based drug and to evaluate its safety and effect for suppressing acute respiratory distress syndrome(ARDS)in COVID-19 ***:A putative ARDS-suppressing drug Keguan-1 was first developed and then evaluated by a randomized,controlled two-arm *** two arms of the trial consist of a control therapy(alpha interferon inhalation,50(x g twice daily;and lopinavir/ritonavir,400 and 100 mg twice daily,respectively)and a testing therapy(control therapy plus Keguan-119.4 g twice daily)by random number table at 1:1 ratio with 24 cases each *** 2-week treatment,adverse events,time to fever resolution,ARDS development,and lung injury on newly diagnosed COVID-19 patients were ***:An analysis of the data from the first 30 participants showed that the control arm and the testing arm did not exhibit any significant differences in terms of adverse *** on this result,the study was expanded to include a total of 48 participants(24 cases each arm).The results show that compared with the control arm,the testing arm exhibited a significant improvement in time to fever resolution(P=0.035),and a significant reduction in the development of ARDS(P=0.048).Conclusions:Keguan-1-based integrative therapy was safe and superior to the standard therapy in suppressing the development of ARDS in COVID-19 patients.(Trial registration *** 04251871 at ***).

Hyperhomocysteinemia is an important risk factor for preeclampsia-eclampsia. This study established a pregnant rat model of hyperhomocysteinemia, in which blood plasma homocysteine concentrations were twice or three times greater than that of normal pregnant rats. TUNEL revealed an increase in the number of apoptotic cells in the frontal cortex of pregnant rats with hyperhomocysteinemia. In addition, immunohistochemical staining detected activated nuclear factor-KB-positve cells in the frontal cortex. Reverse transcription-PCR detected that mRNA expression of the anti-apoptotic gene bcl-2 diminished in the frontal cortex. In situ hybridization and western blotting revealed that N-methyi-D- aspartate receptor 1 mRNA and protein expression was upregulated in the frontal cortex and hippocampus. These results indicate that hyperhomocysteinemia can induce brain cell apoptosis, increase nerve excitability, and promote the occurrence of preeclampsia in pregnant rats.

Background Foetal echocardiography has become a diagnostic method to detect foetal congenital heart disease with high probability. However, it is not only time consuming and but also difficult to visualize outflow tract of foetus early in the second trimester of pregnancy, even for an experienced obstetric uhrasonographer. Recently, many methods for screening foetal cardiac anomalies were explored, but much more work is needed to develop an effective and suitable screening method. The aim of this study was to investigate the clinical significance of utilising the ductus venosus （DV） Doppler examination and the four-chamber view of heart to screen for foetal cardiac malformation in early second trimester of pregnancy. Methods Heart and DV of 401 consecutive foetuses in early second trimester （12^＋1- 17^ ＋6 weeks） in high risk pregnancies were examined with Acuson 128 xp/10 or Sequoia 512 ultrasound diagnostic systems. Absent or reversed flow during atrial contraction （A-wave） in the DV was defined as sufficiently abnormal to screen for foetal cardiac malformations. The foetal echocardiographic diagnosis was confirmed by postnatal echocardiography （or postmortem）. The sensitivities of screening tests were compared among the three methods： DV Doppler examination, four-chamber view alone, and the combination of both *** Satisfactory examinations were obtained in 383/401 foetuses （95%）. Thirty foetuses with cardiac abnormalities were confirmed by neonatal echocardiography （ or postmortem ）. The sensitivity of DV Doppler examination or four-chamber view alone is 63 % （19/30） and 60 % （ 18/30）, respectively. The sensitivity of combining information, DV Doppler flow waveform and four-chamber view, to screen for foetal cardiac malformation is 83% （25/30） and significantly better than that of either DV Doppler flow waveform or four chamber view alone （ P 〈 0. 05 ）.
