Bulking sludge for PHA production:Energy saving and comparative storage capacity with well-settled sludge

Two acetate-fed sequencing batch reactors （SBR） were operated under an aerobic dynamic feeding （ADF） model （SBR#2） and with anaerobic phase before aerobic phase （SBR#1） to select mixed cultures with a high polyhydroxyalkanoates （PHA） storage response. Although kinetic selection based on storage response should bring about a predominance of floc-formers, a bulking sludge with storage response comparable to well-settled sludge was steadily established. An anaerobic phase was introduced before the aerobic phase in the ADF model to improve the sludge settleability （SBR #1）, however, due to the consequent increased feast/famine ratio, the performance of SBR #1, in terms of both the maximum PHB （polyhydroxybutyrate） cell content and APHB, was lower than that of SBR #2. SBR #2 gradually reached a steady state while SBR #1 failed suddenly after 50 days of operation. The maximum specific substrate uptake rate and storage rate for the selected bulking sludge were 0.4 Cmol Ae/（Cmol ***） and 0.18 Cmol Ac/（Cmol ***）, respectively, resulting a yield of 0.45 Cmol PHB/（Cmol Ae） in SBR #2 in the culture enrichment phase. A maximum PHB content of 53% of total suspended solids and PHB storage rate of 1.36 Cmol Ac/（Cmol ***） was achieved at 10.2 hr in batch accumulation tests under nitrogen starvation. The results indicated that it was feasible to utilize filamentous bacteria to accumulate PHA with a rate comparable to well-settled sludge, Furthermore, the lower dissolved oxygen demand of filamentous bacteria would save energy required for aeration in the culture enrichment stage.

Chronic electrical stimulation induces functional network changes in cortical neuron cultures

Recent studies demonstrated that a functional brain network could be regarded as a complex *** the help of network theory,neuroscientists can identify common organizational principles of the functional brain *** a consequence,some non-random organizational features,such as"small world"(most of the nodes are not connected directly but can communicate with few intermediate relay steps)and"rich club"(nodes that are rich in connections tend to form strongly interconnected clubs),have been found in functional brain ***,the"small world"organizational feature of neuronal functional networks in vitro was found to be influenced by external ***,little is known about the influence of chronic electrical stimulation on functional networks of dissociated cortical cultures during network *** the present study,cortical cultures were electrically stimulated at a frequency of 0,0.02,and 0.2 Hz,between 7 and 26 days in vitro(DIV).The spontaneous activity of the cortical cultures was recorded using ***,a cross-covariance method and graph theory were applied to investigate organizational feature of functional *** results showed that over 3 weeks of stimulation,the network density significantly increased with maturation in the control and 0.02 Hz stimulation groups,but not in 0.2 Hz stimulation ***,all the cultures had a small-world topology at 14,18,22,and 26 DIV,free from the effect of chronic electrical ***,we found an asymmetry effect that partial electrical stimulation inhibited the formation of node connections in stimulated *** effect was more pronounced at 0.2 Hz than at 0.02 Hz *** results suggest that electrical stimulation does not affect the small-world properties of neural ***,electrical stimulation modulates connectivity patterns,and neurons within the stimulated area are less connected than neurons outside the stimulated area.

Neural activity extraction and neural decoding from neural signals are an important part of critical components of brain-computer interface *** the development of brain-computer interface technology,the demand for precise external control and nervous activities in macaque monkey during unilateral hand grasp has increased the complexity of control and neural decoding,which puts forward higher requirements for the accuracy and stability of feature extraction and neural *** this study,a micro Capsnet network architecture that consists of a few network layers,a vector feature structure,and optimization network parameters,is proposed to decrease the computing time and complexity,decrease artificial debugging,and improve the decoding *** with KNN,SVM,XGBOOST,CNN,Simple RNN,and LSTM,the algorithm in this study improves the decoding accuracy by 98.03%,and achieves state-of-the-art accuracy and stronger ***,the proposed algorithm can further enhance the control accuracy in the brain-computer interface.

Embryoid bodies formation and differentiation from mouse embryonic stem cells in collagen/Matrigel scaffolds

Embryonic stem （ES） cells have the potential to develop into any type of tissue and are considered as a promising source of seeding cells for tissue engineering and transplantation *** main catalyst for ES cells differentiation is the growth into embryoid bodies （EBs）,which are utilized widely as the trigger of in vitro *** this study,a novel method for generating EBs from mouse ES cells through culture in collagen/Matrigel scaffolds was successfully *** single ES cells were seeded in three dimensional collagen/Matrigel scaffolds,they grew into aggregates gradually and formed simple EBs with circular *** 7 days＇ culture,they formed into cystic EBs that would eventually differentiate into the three embryonic germ *** of the EBs in terms of morphology and potential to differentiate indicated that they were typical in structure and could generate various cell types;they were also able to form into tissue-like ***,with introduction of ascorbic acid,ES cells differentiated into cardiomyocytes efficiently and started contracting synchronously at day *** results demonstrated that collagen/Matrigel scaffolds supported EBs formation and their subsequent differentiation in a single three dimensional environment.

The cell-derived extracellular matrix(ECM)-modified scaffolds have advantages of mimic tissue specificity and been thought to better mimic the native cellular microenvironment in *** derived from cardiac fibroblasts(CFs)are considered as key elements that provide a natural cell growth microenvironment and change the fate of cardiomyocytes(CMs).Here,a new hybrid scaffold is designed based on silk fibroin(SF)scaffold and CFs-derived *** were seeded on the SF scaffold for 10days culturing and decellularized to produce CFs-derived ECM-coated SF *** results showed that the cell-derived ECM-modified silk fibroin scaffold material contained collagen,laminin,fibronectin and other ECM components with myocardial-like *** to explore its effects on brown adipose stem cells(BASCs)differentiation into *** found that the CFderived ECM-coated scaffold also increased the expression of CM-specific proteins(*** troponin T and α-actinin)of ***,the b1-integrin-dependent transforming growth factor-β1 signaling pathway was also involved in the regulation of CF-derived ECM by promoting the differentiation of BASCs into ***,these findings provide insights into the bionic manufacturing of engineered cardiac tissues(ECTs)and establish a theoretical basis for the construction of ECTs.

Distribution and characteristics of telocytes as nurse cells in the architectural organization of engineered heart tissues

Interstitial Cajal-like cells are a distinct type of interstitial cell with a wide distribution in mammalian organs and tissues,and have been given the name"telocytes".Recent studies have demonstrated the potential roles of telocytes in heart development,renewal,and ***,further research on the functions of telocytes is limited by the complicated in vivo *** study was designed to construct engineered heart tissue(EHT)as a three-dimensional model in vitro to better understand the role of telocytes in the architectural organization of the *** were constructed by seeding neonatal cardiomyocytes in collagen/Matrigel scaffolds followed by culture under persistent static *** in EHTs were identified by histology,toluidine blue staining,immunofluorescence,and transmission electron *** results from histology and toluidine blue staining demonstrated widespread putative telocytes with compact toluidine blue-stained nuclei,which were located around *** from the cell bodies showed a characteristic dichotomous branching pattern and formed networks in *** revealed positive staining of telocytes for CD34 and vimentin with typical moniliform prolongations.A series of electron microscopy images further showed that typical telocytes embraced the cardiomyocytes with their long prolongations and exhibited a marked appearance of nursing cardiomyocytes during the construction of *** finding highlights the great importance of telocytes in the architectural organization of *** also suggests that EHT is an appropriate physical and pathological model system in vitro to study the roles of telocytes during heart development and regeneration.

Single-cell transcriptomics reveals neural stem cell trans-differentiation and cell subpopulations in whole heart decellularized extracellular matrix

The whole heart decellularized extracellular matrix(ECM) has become a promising scaffold material for cardiac tissue engineering. Our previous research has shown that the whole heart acellular matrix possesses the memory function regulating neural stem cells(NSCs) trans-differentiating to cardiac lineage cells. However, the cell subpopulations and phenotypes in the trans-differentiation of NSCs have not been clearly identified. Here, we performed single-cell RNA sequencing and identified 2,765 cells in the recellularized heart with NSCs revealing the cellular diversity of cardiac and neural lineage, confirming NSCs were capable of trans-differentiating into the cardiac lineage while maintaining the original ability to differentiate into the neural lineage. Notably, the trans-differentiated heart-like cells have dual signatures of neuroectoderm and cardiac mesoderm. This study unveils an in-depth mechanism underlying the trans-differentiation of NSCs and provides a new opportunity and theoretical basis for cardiac regeneration.

Modulation of Beta Oscillations for Implicit Motor Timing in Primate Sensorimotor Cortex during Movement Preparation

Motor timing is an important part of sensorimotor control. Previous studies have shown that beta oscillations embody the process of temporal perception in explicit timing tasks. In contrast, studies focusing on beta oscillations in implicit timing tasks are lacking. In this study, we set up an implicit motor timing task and found a modulation pattern of beta oscillations with temporal perception during movement preparation. We trained two macaques in a repetitive visually-guided reach-to-grasp task with different holding intervals. Spikes and local field potentials were recorded from microelectrode arrays in the primary motor cortex, primary somatosensory cortex, and posterior parietal cortex. We analyzed the association between beta oscillations and temporal interval in fixedduration experiments(500 ms as the Short Group and1500 ms as the Long Group) and random-duration experiments(500 ms to 1500 ms). The results showed that the peak beta frequencies in both experiments ranged from15 Hz to 25 Hz. The beta power was higher during the hold period than the movement(reach and grasp) ***, in the fixed-duration experiments, the mean poweras well as the maximum rate of change of beta power in the first 300 ms were higher in the Short Group than in the Long Group when aligned with the Center Hit event. In contrast, in the random-duration experiments, the corresponding values showed no statistical differences among groups. The peak latency of beta power was shorter in the Short Group than in the Long Group in the fixed-duration experiments, while no consistent modulation pattern was found in the random-duration experiments. These results indicate that beta oscillations can modulate with temporal interval in their power mode. The synchronization period of beta power could reflect the cognitive set maintaining working memory of the temporal structure and attention.
