Peptide transporter 2(PepT2)transports short peptides from the blood into bovine mammary epithelial cells(BMEC)to stimulate milk protein *** the fact that the effect of PepT2 is acknowledged in BMEC,little is known about its *** study was completed to investigate the role of mammalian target of the rapamycin(mTOR)signaling in regulating the expression and function of PepT2 in *** regulation of PepT2 by mTOR in BMEC was studied in vitro using peptide transport assay,gene silencing,Western *** membrane expression of PepT2 and the uptake of b-Ala-Lys-N-7-amino-4-methylcoumarin-3-acetic acid(b-Ala-Lys-AMCA),a model dipeptide,in BMEC were reduced by rapamycin(a mTOR inhibitor)and silencing of either mTOR complex 1(mTORC1)or mTOR complex 2(mTORC2),stimulated by DEP domain-containing mTOR-interacting protein(DEPTOR,endogenous inhibitor of mTORC1 and mTORC2)*** trafficking of PepT2 to the membrane and the uptake of b-Ala-Lys-AMCA was promoted by neuronal precursor cell-expressed developmentally down-regulated 4 isoform 2(Nedd4-2)*** effects of knockdown of mTORC1,but not mTORC2,on cell membrane expression and transport activity of PepT2 was abolished by Nedd4-2 *** immunofluorescence staining,PepT2 was identified to be interacting with *** Nedd4-2 expression and the interaction between PepT2 and Nedd4-2 was increased through mTORC1 knockdown,indicating an increased ubiquitination of *** results revealed that mTORC1 can regulate the expression and function of PepT2 through Nedd4-2 in BMEC.

Coal seam pyrolysis occurs during coal seam fires and during underground coal gasification. This is an important source of polycyclic aromatic hydrocarbon （PAH） emission in China. Pyrolysis in a coal seam was simulated in a tubular furnace. The 16 US Environmental Protection Agency priority controlled PAHs were analyzed by HPLC. The effects of temperature, heating rate, pyrolysis atmosphere, and coal size were investigated. The results indicate that the 3-ring PAHs AcP and AcPy are the main species in the pyrolysis gas. The 2-ring NaP and the 4-ring Pyr are also of concern. Increasing temperature caused the total PAH yield to go through a minimum. The lowest value was obtained at the temperature of 600℃. Higher heating rates promote PAH formation, especially formation of the lower molecular weight PAHs. The typical heating rate in a coal seam, 5 ℃/min, results in intermediate yields of PAHs. The total PAHs yield in an atmosphere of N2 is about 1.81 times that seen without added N2, which indicates that an air flow through the coal seam accelerates the formation of PAHs. An increase in coal particle size reduces the total PAHs emission but promotes the formation of 5- and f-ring PAHs.

Jervine,a novel steroidal alkaloid from Veratrum nigrum L.,exhibits both antitumor effect and potential *** aim of study was to characterize the pharmacokinetic behaviors and enterohepatic circulation of jervine in rats.A rapid and simple ultra-high performance liquid chromatography-tandem mass spectrometric method was developed and validated for quantification of jervine and alpinetin (internal standard) in rat *** extraction from rat plasma by a simple protein-precipitation method,the analyte was separated on a C18 column (2.1 mm×50 mm,1.7μm) using water with 0.1%formic acid and acetonitrile as the mobile phase delivered at a flow rate of 0.4mL/*** and alpinetin were determined in the positive mode with multiple reaction monitoring (MRM) of the ion transitions at m/z426.3→108.8 and m/z 271.0 166.9,*** docking method was used to investigate the binding of jervine to p-glycoprotein and dehydroepiandrosterone *** method was well validated within acceptance limits including specificity,matrix effect,recovery,precision,accuracy,and stability,and was successfully applied to the pharmacokinetic study of jervine after oral and intravenous administration to *** presented a small volume of distribution,fast absorption,high oral bioavailability,and enterohepatic *** enterohepatic circulation was first observed in veratrum alkaloids,and was further investigated by molecular docking studies,which was related to the binding of jervine to p-glycoprotein and dehydroepiandrosterone *** pharmacokinetic properties and enterohepatic circulation of jervine in rats provided a significant basis for the drug-drug interaction and toxicity study in the future.

Application of response surface methodology to the chemical cleaning process of ultrafiltration membrane

A numerical model was established to predict and optimise the chemical cleaning process of Polyvinylidene Fluo- ride （PVDF） Ultrafiltration （UF） membranes with the results from the experiment that applied the Response Sur- face Method （RSM） and Central Composite Design （CCD）. The factors considered in the experimental design were sodium hydroxide （NaOH） concentration, sodium bypochlorite concentration （NaCIO）, citric acid concentration and cleaning duration, The interactions between the factors were investigated with the numerical model. Humic acid （20 mg· L-1） was used as the model foulant, and chemical enhanced backflush （CEB） was employed to sim- ulate the chemical cleaning process. The concentrations of sodium hydroxide, sodium hypochlorite, citric acid and cleaning duration tested during the experiments were in the range of 0.1%-0.3% 100-300 mg· L-1 1%-3% and 0.5-1.5 h, respectively. Among the variables, the sodium hypochlorite concentration and the cleaning dura- tion showed a positive relationship involving the increased efficiency of the chemical cleaning. The chemical cleaning efficiency was hardly improved with increasing concentrations of sodium hydroxide. However, the data was sharply decreased when at a low level of sodium hydroxide concentration. In total, 54 sets of cleaning schemes with 80% to 100K cleaning efficiency were observed with the R＆M model after calibration.

Herein,a flexible ZIF-67/PAN hybrid membrane was successfully prepared by the incorporation of ZIF-67 nanoparticles and PAN nanofibers through electrospinning *** hybrid membrane presented tomatoes on sticks structures with one single PAN fiber stringing series of ZIF-67 *** morphology,electrolyte wettability,heat resistance,flexibility,and electrochemical properties of the electrospun ZIF-67/PAN membranes were *** the membranes prepared with different percentage of ZIF-67,the 30% ZIF-67/PAN membrane exhibited outstanding heat shrinkage resistance(remained intact at 200℃ for 1 h),excellent electrolyte uptake(556.39%),wide electrochemical window(~5.25 V)and high ionic conductivity(2.98 mS cm^(−1)).When used as lithium-ion batteries(LIBs)separators,the cells assembled by 30% ZIF-67/PAN membrane presented excellent rate capacity and high capacity retention of 86.9% after 300 cycles at *** importantly,the cells assembled with ZIF-67/PAN membranes repeated bent for 1000 times also exhibited high rate performance and maintained capacity retention of 92% after 100 cycles at 1 *** characterization and the electrochemical testing suggest the electrospinning prepared ZIF-67/PAN flexible membranes can be expected to be used as potential separator for advanced batteries with high safety and high performance.
