Evaluation and clinical application of a new method for detecting ADAMTS13 activity

Background A severe deficiency of ADAMTS13 activity contributes to the pathogenesis of thrombotic thrombocytopenic purpura （TTP）. Measuring the activity of ADAMTS13 is helpful for the diagnosis of TTP and the prognostic monitor in TTP patients. Most available assays are cumbersome and costly, so not easily adapted to routine laboratories. ADAMTS13 cleaves von Willebrand factor （VVVF） within the domain A2, located between domains A1 and A3. Therefore, specific assays for ADAMTS13 activity could be based on the different structures of VWF before and after the cleavage. Using this hypothesis we try to establish a new and simple method to determine ADAMTS13 activity. Methods First, plasma samples were exposed in denaturing condition to allow cleavage of VWF by ADAMTS13. Then, the ADAMTS13 activity was measured with two novel monoclonal antibodies, SZ-129 and SZ-125, which specifically recognize the VWF A1 and A3 domains by using a two-site sandwich ELISA. Compared with a residual-collagen binding assay （R-CBA）, plasma ADAMTS13 activities in 161 samples were assessed, and the inhibitory activities of ADAMTS13 autoantibody in 24 TTP patients were determined. The relationship of these two assays was analyzed by linear correlation, and the sensitivity and specificity of the new assay was also evaluated. Results Plasma ADAMTS13 activities in normal people and TTP, acute myocardial infarction （AMI）, and idiopathic thrombocytopenic purpura （ITP） patients determined by the new assay were （89.75±7.93）%, （17.63±18.71）%, （68.55±18.08）%, （85.83±9.84）%, respectively. Results were consistent with those of R-CBA, the squared correlation factor was 0.9183 of the two assays. The new assay can easily discriminate a ＂TTP plasma sample from a non-TTP plasma sample （P 〈0.01）, and the coefficient of variation for the new assay was 6.17%. The Moon provides a unique environment for investigating nearby astrophysical events such as *** samples retain valuable information from these events,via detectable long-lived“fingerprint”radionuclides such as^(60)*** this work,we stepped up the development of an accelerator mass spectrometry(AMS)method for detecting^(60)Fe using the HI-13tandem accelerator at the China Institute of Atomic Energy(CIAE).Since interferences could not be sufficiently removed solely with the existing magnetic systems of the tandem accelerator and the following Q3D magnetic spectrograph,a Wien filter with a maximum voltage of±60 kV and a maximum magnetic field of 0.3 T was installed after the accelerator magnetic systems to lower the detection background for the low abundance nuclide^(60)Fe.A 1μm thick Si_(3)N_(4) foil was installed in front of the Q3D as an energy *** particle detection,a multi-anode gas ionization chamber was mounted at the center of the focal plane of the ***,an^(60)Fe sample with an abundance of 1.125×10^(-10)was used to test the new AMS *** results indicate that^(60)Fe can be clearly distinguished from the isobar^(60)*** sensitivity was assessed to be better than 4.3×10^(-14)based on blank sample measurements lasting 5.8 h,and the sensitivity could,in principle,be expected to be approximately 2.5×10^(-15)when the data were accumulated for 100 h,which is feasible for future lunar sample measurements because the main contaminants were sufficiently separated.

目的在具有正常血管性血友病因子裂解酶（ADAMTS13）活性且不存在抗ADAMTS13自身抗体的特发性血栓性血小板减少性紫癜（1vrP）患者血浆中检测是否存在抗血管性血友病因子（VwF）自身抗体，并通过体外实验验证鼠源性抗人VwF单克隆抗体（单抗）是否可以降低ADAMTS13对VwF的裂解活性。方法本研究为临床实验诊断研究。应用酶联免疫吸附试验对53例成人特发性1TrP患者检测抗VWF自身抗体，采用十二烷基硫酸钠-琼脂糖凝胶电泳进行VwF多聚体分析，检测是否存在超大分子量VwF（uL—VWF）多聚体。并且，在高剪切力和变性条件下分别观察8种鼠源性抗VwF单抗对重组ADAMTS13（rADAMTS13）裂解血浆源纯化VwF的活性的影响。结果在2例具有正常血浆ADAMTS13活性且不存在抗ADAMTS13自身抗体的特发性TrP患者血浆中检测到抗VwF自身抗体和UL—VwF多聚体。另外，一种鼠源性抗人VwF单抗SZ34在高剪切力条件下能够降低rADAMTS13对VwF的水解活性，但在静态／盐酸胍变性条件下并无此影响。结论抗VwF抗体有可能是具有正常ADAMTS13活性且不存在抗ADAMTS13自身抗体的特发性TTP患者的发病原因之一。

We investigate high time resolution data obtained by the Gravitational wave high-energy Electromagnetic Counterpart All-sky Monitor(GECAM)during the flare event on 2022 April 21 at 01:52 *** subpeaks with durations of 4-6 s have been detected in the hard X-ray precursor phase,and the key feature is that they appear in pairs and seem like double-peak *** subpeaks are rarely observed in hard X-ray band and confirmed by the microwave obtained by Nobeyama Radio Polarimeters(NoRP)and Radio Solar Telescope Network(RSTN).While an exponential function can describe the continuum component of the time profile from the precursor to part of the impulsive *** periods of quasi-periodic pulsations(QPPs)are detected to be about 7.3 and 12.8 s for the precursor and impulsive phase,respectively,with at least 95%confidence *** paired QPPs are assumed to be double-peak QPPs and then the scenario of current loop coalescence model is found to be in good agreement with our *** precursor phase can be interpreted as the oscillating coalescence of two islands,while the impulsive phase can be interpreted as more islands to coalesce one by one to form larger islands.

mammalian target of rapamycin(m TOR) inhibitor as an attractive drug target with promising antitumor effects has been widely investigated. High quality clinical trial has been conducted in liver transplant(LT) recipients in Western countries. However, the pertinent studies in Eastern world are paucity. Therefore, we designed a clinical trial to test whether sirolimus can improve recurrence-free survival(RFS) in hepatocellular carcinoma(HCC) patients beyond the Milan criteria after LT. This is an open-labeled, single-arm, prospective, multicenter, and real-world study aiming to evaluate the clinical outcomes of early switch to sirolimus-based regimens in HCC patients after LT. Patients with a histologically proven HCC and beyond the Milan criteria will be enrolled. The initial immunosuppressant regimens are center-specifc for the frst 4-6 weeks. The following regimens integrated sirolimus into the regimens as a combination therapy with reduced calcineurin inhibitors based on the condition of patients and centers. The study is planned for 4 years in total with a 2-year enrollment period and a 2-year follow-up. We predict that sirolimus conversion regimen will provide survival benefts for patients particular in the key indicator RFS as well as better quality of life. If the trial is conducted successfully, we will have a continued monitoring over a longer follow-up time to estimate indicator of overall survival. We hope that the outcome will provide better evidence for clinical decision-making and revising treatment guidelines based on Chinese population data.

The Large sky Area Multi-Object Fiber Spectroscopic Telescope（LAMOST） general survey is a spectroscopic survey that will eventually cover approximately half of the celestial sphere and collect 10 million spectra of stars, galaxies and QSOs. Objects in both the pilot survey and the first year regular survey are included in the LAMOST DR1. The pilot survey started in October 2011 and ended in June 2012, and the data have been released to the public as the LAMOST Pilot Data Release in August 2012. The regular survey started in September 2012, and completed its first year of operation in June 2013. The LAMOST DR1 includes a total of 1202 plates containing 2 955 336 spectra, of which 1 790 879 spectra have observed signalto-noise ratio（SNR） ≥ 10. All data with SNR ≥ 2 are formally released as LAMOST DR1 under the LAMOST data policy. This data release contains a total of 2 204 696 spectra, of which 1 944 329 are stellar spectra, 12 082 are galaxy spectra and 5017 are quasars. The DR1 not only includes spectra, but also three stellar catalogs with measured parameters： late A,FGK-type stars with high quality spectra（1 061 918 entries）, A-type stars（100 073 entries）, and M-type stars（121 522 entries）. This paper introduces the survey design, the observational and instrumental limitations, data reduction and analysis, and some caveats. A description of the FITS structure of spectral files and parameter catalogs is also provided.

We operated a p-type point contact high purity germanium(PPCGe)detector(CDEX-1B,1.008 kg)in the China Jinping Underground Laboratory(CJPL)for 500.3 days to search for neutrinoless double beta(0νββ)decay of ^(76)Ge.A total of 504.3 kg⋅day effective exposure data was *** anti-coincidence and the multi/single-site event(MSE/SSE)discrimination methods were used to suppress the background in the energy region of interest(ROI,1989–2089 keV for this work)with a factor of 23.A background level of 0.33 counts/(keV⋅kg⋅yr)was *** lower limit on the half life of^(76)Ge 0νββdecay was constrained as T_(1/2)^(0ν)>1.0×10^(23)yr(90%C.L.),corresponding to the upper limits on the effective majorana neutrino mass:<3.2–7.5 eV.

北京部分地区肠道病原菌的分布及耐药状况

目的监测北京部分地区肠道致病菌的组成及耐药状况，为本地区的流行病学研究及临床合理用药提供依据。方法480份粪便标本按常规培养，筛选出致病菌后经生化及血清学进一步鉴定到种、群或血清型，并进行药敏试验。结果肠道病原菌感染，以男性为主，儿童和青年发病为多，6～8月为腹泻发病高峰。病原以致泻性大肠杆菌居首位，其次是志贺菌。本次检测出肠道致病菌对氨苄西林（67．8％）、甲氧苄氨嘧啶（67．8％）等均有严重的抗药性；对哌拉西林（42．5％）、氨苄西林-舒巴坦（33．0％）、呋喃妥因（30．5％）等也均产生了不同程度的抗药性：只有对关洛培南（0％）、磷霉素（1．1％）、头孢塞肟-克拉维酸（3．4％）、头孢他啶（4．6％）、氨曲南（5．2％）均比较敏感。结论北京地区感染性腹泻的病原种类繁多，有性别、年龄、季节的分布特点，耐药性不同，应重视监测。

加速钢渣水化过程、胶凝活性激发对钢渣综合利用率的提高有重要意义。以矿渣和脱硫石膏为复合激发剂,基于交叉试验的设计方法,对复合胶凝材料的组成进行了优化,分析了复合胶凝材料的综合性能,采用X射线衍射(X-ray diffraction,XRD)、扫描电子显微镜(scanning electronic microscopy,SEM)、傅里叶变换红外光谱(Flourier transform-infrared spectroscopy,FT-IR)等测试方法表征了复合胶凝材料的组成及结构,并以此揭示复合胶凝材料协同优化的机理。复合胶凝材料配合比的交叉试验结果表明:钢渣30%、矿渣60%、脱硫石膏10%,水灰比为0.5,28 d抗压强度为40.4 MPa,其他各项性能指标满足GB/T1346—2011要求。通过多项测试手段可以判断出,矿渣在脱硫石膏的激发作用下不断水解,缩短了水化诱导前期和延长了诱导期,提升早期水化反应程度,释放大量热量,生成了generates ettringite(AFt)和C—S—H凝胶。而浆体的pH值上升,促进钢渣水化反应,加速了浆体由液相反应转为固相反应,证明了各原料之间存在协同反应。
