OsPRMT6a-mediated arginine methylation of OsJAZ1 regulates jasmonate signaling and spikelet development in rice

Although both protein arginine methylation(PRMT)and jasmonate(JA)signaling are crucial for regulating plant development,the relationship between these processes in the control of spikelet development remains *** this study,we used the CRISPR/Cas9 technology to generate two OsPRMT6a loss-of-function mutants that exhibit various abnormal spikelet ***,we found that OsPRMT6a can methylate arginine residues in JA signal repressors OsJAZ1 and *** showed that arginine methylation of OsJAZ1 enhances the binding affinity of OsJAZ1 with the JA receptors OsCOI1a and OsCOI1b in the presence of JAs,thereby promoting the ubiquitination of OsJAZ1 by the SCF^(OsCOI1a/OsCOI1b) complex and degradation via the 26S *** process ultimately releases OsMYC2,a core transcriptional regulator in the JA signaling pathway,to activate or repress JA-responsive genes,thereby maintaining normal plant(spikelet)***,in the osprmt6a-1 mutant,reduced arginine methylation of OsJAZ1 impaires the interaction between OsJAZ1 and OsCOI1a/OsCOI1b in the presence of *** a result,OsJAZ1 proteins become more stable,repressing JA responses,thus causing the formation of abnormal spikelet ***,we discovered that JA signaling reduces the OsPRMT6a mRNA level in an OsMYC2-dependent manner,thereby establishing a negative feedback loop to balance JA *** further found that OsPRMT6a-mediated arginine methylation of OsJAZ1 likely serves as a switch to tune JA signaling to maintain normal spikelet development under harsh environmental conditions such as high ***,our study establishes a direct molecular link between arginine methylation and JA signaling in rice.

Heading date(or flowering time)is one of the most important agronomic traits in rice,influencing its regional adaptability and crop *** major-effect genes for rice heading date have been identified,but in practice they are difficult to be used for rice molecular breeding because of their dramatic effects on heading *** with minor effects on heading date,which are more desirable for fine-tuning flowering time without significant yield penalty,were seldom *** this study,we identified a new minor-effect heading date repressor,Delayed Heading Date 4(DHD4).The dhd4 mutant shows a slightly earlier flowering phenotype without a notable yield penalty compared with wild-type plants under natural long-day ***4 encodes a CONSTANS-like transcription factor localized in the ***,biochemical,and genetic assays show that DHD4 can compete with 14-3-3 to interact with OsFD1,thus affecting the formation of the Hd3a-14-3-3-OsFD1 triprotein FAC complex,resulting in reduced expression of OsMADS14 and OsMADS15,and ultimately delaying *** together,these results shed new light on the regulation of flowering time in rice and provide a promising target for fine-tuning flowering time to improve the regional adaptability of rice.

作物生产常遭受各类虫害威胁。揭示昆虫与寄主之间的互作机制,对害虫的绿色防治具有重要意义。武汉大学何光存团队鉴定了褐飞虱(BPH)唾液蛋白BISP。在易感品种中, BISP靶向OsRLCK185并抑制其介导的基础防御。在携带褐飞虱抗性基因Bph14的水稻(Oryza sativa)品种中,BPH14直接结合BISP并激活寄主的免疫反应,但会抑制水稻的生长。BISP-BPH14与自噬装载受体OsNBR1结合,通过自噬途径降解BISP,下调水稻对BPH的抗性,恢复植株正常生长。该研究鉴定到首个被植物免疫受体感知的昆虫唾液蛋白,揭示了寄主通过感知并调节昆虫效应蛋白水平来平衡水稻抗性与生长发育的分子机制,为培育高产水稻抗虫品种提供了新思路。

Heading date and plant height are important determinants of yield in rice（Oryza sativa L.）.In this study,we characterized a late heading,dwarf mutant known as lhdd10 selected following ethyl methane sulfonate（EMS）-treatment of *** cultivar ***10 showed late heading,dwarfness and slightly darker green leaves than wild type 93-11 under long day and short day *** isolated LHDD10 by map-based cloning;it encoded a putative FAD-linked oxidoreductase protein（a brassinosteroid biosynthetic gene） that localized to the ***10 was constitutively expressed in various tissues,but more so in shoot apices and *** data showed that LHDD10 influences heading date by controlling the expression of heading date regulators,such as OsMADS50 in both LD and SD ***10 also negatively regulated expression of chlorophyll biosynthetic genes to reduce the chlorophyll *** data indicated that BRs play important roles in regulating heading date and chlorophyll *** work provides material that will allow study of how BRs regulate heading date in rice.

Rice(Oryza sativa) is a staple food cror the world’s ***(Chl) biosynthesis plays a vital role to rice grain forming and final ***-deficient mutants generally affect leaf colors,and are ideal genetic materials to investigate molecular mechanisms that regulate Chl biosynthesis and chloroplast *** than ninety rice mutants were identified associated with leaf colors in a rice science database,for example albino,chlorina, stripe,virescent and zebra according to their phenotypes(http://***/rice/ oryzabaseV4/).Although some of genes underlying the mutations have been cloned,information is still limited to elucidate the molecular mechanism controlling Chl biosynthesis in rice. Here we reported a rice mutant,designated yellow-leave 3(t),which showed yellow leave phenotype during seedling growth stages in the field.A F population by crossing the mutant to ’N22’(a normal leaf color variety) was *** segregate analysis with 10 recessive individuals as a DNA mixture pool was conducted initially to screen linked molecular markers with mutant phenotype,and then more recombinants and markers were used to narrow target *** present yellow-leave 3(t) was mapped in long arm of chromosome 4 within 10 centimorgan (cM).To our knowledge,yellow-leave 3(t) is a new locus related with leaf color, which provides a basis to decipher molecular mechanism in rice Chl biosynthesis.

在全球化语境下,如何对中小学生进行"地方认同"教育,研学旅行课程提供了极好的教育载体。以全国中小学研学实践教育基地滕王阁为例,从"地方认同"的角度探讨了研学实践课程开发的一般过程,即以促进地方认同为目标,首先对研学基地进行地方性分析,提炼地方文化符号,构建课程内容;其次按照具身认知和生成认知规律,设计身体和环境互动的课程形式;最后对学生过程参与、学习结果呈现和目标达成度进行综合评价。
