NOX4 exacerbates Parkinson's disease pathology by promoting neuronal ferroptosis and neuroinflammation

Parkinson's disease is primarily caused by the loss of dopaminergic neurons in the substantia nigra ***,a novel form of regulated cell death characterized by iron accumulation and lipid peroxidation,plays a vital role in the death of dopaminergic ***,the molecular mechanisms underlying ferroptosis in dopaminergic neurons have not yet been completely *** oxidase 4 is related to oxidative stress,however,whether it regulates dopaminergic neuronal ferroptosis remains *** aim of this study was to determine whether NADPH oxidase 4 is involved in dopaminergic neuronal ferroptosis,and if so,by what *** found that the transcriptional regulator activating transcription factor 3 increased NADPH oxidase 4 expression in dopaminergic neurons and astrocytes in an 1-methyl-4-phenyl-1,2,3,6 tetrahydropyridine-induced Parkinson's disease *** oxidase 4 inhibition improved the behavioral impairments observed in the Parkinson's disease model animals and reduced the death of dopaminergic ***,NADPH oxidase 4 inhibition reduced lipid peroxidation and iron accumulation in the substantia nigra of the Parkinson's disease model ***,we found that NADPH oxidase 4 interacted with activated protein kinase Cαto prevent ferroptosis of dopaminergic ***,by lowering the astrocytic lipocalin-2 expression,NADPH oxidase 4 inhibition reduced 1-methyl-4-phenyl-1,2,3,6 tetrahydropyridine-induced *** findings demonstrate that NADPH oxidase 4 promotes ferroptosis of dopaminergic neurons and neuroinflammation,which contribute to dopaminergic neuron death,suggesting that NADPH oxidase 4 is a possible therapeutic target for Parkinson's disease.

Dynamic modeling of a three-dimensional braided composite thin plate considering braiding directions

Currently, there are a limited number of dynamic models available for braided composite plates with large overall motions, despite the incorporation of three-dimensional(3D) braided composites into rotating blade components. In this paper, a dynamic model of 3D 4-directional braided composite thin plates considering braiding directions is established. Based on Kirchhoff's plate assumptions, the displacement variables of the plate are expressed. By incorporating the braiding directions into the constitutive equation of the braided composites, the dynamic model of the plate considering braiding directions is obtained. the effects of the speeds, braiding directions, and braided angles on the responses of the plate with fixed-axis rotation and translational motion, respectively, are investigated. this paper presents a dynamic theory for calculating the deformation of 3D braided composite structures undergoing both translational and rotational motions. It also provides a simulation method for investigating the dynamic behavior of non-isotropic material plates in various applications.

Nanoindentation in elastoplastic materials:insights from numerical simulations

Finite element simulations of nanoindentation were performed on an elastoplastic material using Berkovich and conical indenters to investigate the effects of geometry on the load-displacement response of the *** Berkovich indenter,widely used in nanoindentation experiments,is typically simplified to a theoretically equivalent 70.3°conical indenter for numerical simulations,which allows for a less computationally intensive two-dimensional(2D)axisymmetric *** studies into the validity of this equivalence assumption for indentations in elastoplastic materials have varying *** 2D and 3D finite element simulations,the present study investigates the response of elastoplastic materials,obeying a combined isotropic and kinematic hardening,to indentation with conical and Berkovich *** show that there is a clear difference in the load-displacement response of the selected material to the two *** Berkovich geometry is found to produce a more localized pattern of contact stresses and plastic strains,leading to a smaller mobilized force for the same magnitude of *** further validate the numerical simulations,experimental results of nanoindentation into an aluminium specimen were compared to elastoplastic finite element simulation *** suggest that machining-induced residual stresses have likely affected the experimental results.

Deciphering the role of mtH2O2 in hepatic ischemia-reperfusion injury mechanisms with low-background super-resolution fluorogenic probe

Hepatic ischemia-reperfusion injury（HIRI） is characterized by mitochondrial dysfunction and oxidative stress. Monitoring mitochondrial hydrogen peroxide（mtH2O2） levels in real-time through super-resolution imaging is crucial for elucidating its distribution in live cells and its mechanism of action during HIRI. However, low-background fluorogenic probes have been overlooked in the context of super-resolution imaging. In this study, we developed a low-background fluorogenic probe（MitowG） with the potential for super-resolution morphology-correlated mitochondrial identification to track the fluctuates of mtH2O2in HIRI. Activation of the desirable fluorescence properties of the probe by mtH2O2was confirmed using structural illumination microscopy（SIM）, enabling high-quality mitochondrial imaging with exceptional specificity and *** in mtH2O2levels were successfully observed in both cellular and rat models of HIRI. Furthermore, we associated the decline in mitochondrial redox homeostasis with accelerated mtH2O2production during HIRI, which triggered mitophagy deficiency and led to cell death. In conclusion, Mito-wG possesses excellent photophysical and low-background properties for SIM imaging, making it a promising tool for mtH2O2tracking in HIRI research and clinical diagnosis.

Noise-induced hearing loss is the primary non-genetic factor contributing to auditory ***,there are currently no effective pharmacological interventions for patients with noise-induced hearing ***,we present evidence suggesting that the lysine-specific demethylase 1 inhibitor–tranylcypromine is an otoprotective agent that could be used to treat noise-induced hearing loss,and elucidate its underlying regulatory *** established a mouse model of permanent threshold shift hearing loss by exposing the mice to white broadband noise at a sound pressure level of 120 d B for 4 *** found that tranylcypromine treatment led to the upregulation of Sestrin2(SESN2)and activation of the autophagy markers light chain 3B and lysosome-associated membrane glycoprotein 1 in the cochleae of mice treated with *** noise exposure group treated with tranylcypromine showed significantly lower average auditory brainstem response hearing thresholds at click,4,8,and 16 k Hz frequencies compared with the noise exposure group treated with *** findings indicate that tranylcypromine treatment resulted in increased SESN2,light chain 3B,and lysosome-associated membrane glycoprotein 1 expression after noise exposure,leading to a reduction in levels of 4-hydroxynonenal and cleaved caspase-3,thereby reducing noise-induced hair cell ***,immunoblot analysis demonstrated that treatment with tranylcypromine upregulated SESN2 expression via the autophagy *** treatment also reduced the production of NOD-like receptor family pyrin domaincontaining 3(NLRP3)*** conclusion,our results showed that tranylcypromine treatment ameliorated cochlear inflammation by promoting the expression of SESN2,which induced autophagy,thereby restricting NLRP3-related inflammasome signaling,alleviating cochlear hair cell loss,and protecting hearing *** findings suggest that inhibiting lysine-specific demethylase 1 is a

Photoluminescence of ZnS Clusters in Zeolite-Y

two obvious emissions are observed from the ZnS clusters encapsulated in zeolite-Y. the emission around 355 nm is sharp and weak, locating at the onset of the absorption edge. the band around 535 nm is broad, strong and Stokes-shifted. Both the two emissions shift to blue and their intensities firstly increase then decrease as the loading of ZnS in zeolite-Y or clusters size decreases. through investigation, the former is attributed to the excitonic fluorescence, and the latter to the trapped luminescence from surface states. the cluster size-dependence of the luminescence may be explained qualitatively by considering both the carrier recombination and the nonradiative recombination rates. Four peaks appearing in the excitation spectra are assigned to the transitions of 1S-1S, 1S-1P, 1S-1D and surface state, respectively. the excitation spectra of the clusters do not coincide with their absorption spectra. the states splitted by quantum-size confinement are detected in the excitation spectra, but could not be differentiated in the optical absorption spectra due to inhomogeneous broadening. the size-dependence of the excitation spectra is similar to that of the absorption spectra. Both the excitation spectra of excitonic and of trapped emissions are similar, but change in relative intensity and shift in position are observed.
