Silybin counteracts lipid excess and oxidative stress in cultured steatotic hepatic cells

AIM: To investigate in vitro the therapeutic effect and mechanisms of silybin in a cellular model of hepatic steatosis.METHODS: Rat hepatoma Fa O cells were loaded with lipids by exposure to 0.75 mmol/L oleate/palmitate for 3 h to mimic liver steatosis. Then, the steatotic cells were incubated for 24 h with different concentrations(25 to 100 μmol/L) of silybin as phytosome complex with vitamin E. The effects of silybin on lipid accumulation and metabolism, and on indices of oxidative stress were evaluated by absorption and fluorescence microscopy, quantitative real-time PCR, western blot, spectrophotometric and fluorimetric assays.RESULTS: Lipid-loading resulted in intracellular triglyceride(TG) accumulation inside lipid droplets, whose number and size increased. TG accumulation was mediated by increased levels of peroxisome proliferator-activated receptors(PPARs) and sterol regulatory element-binding protein-1c(SREBP-1c). The lipid imbalance was associated with higher production of reactive oxygen species(ROS) resulting in increased lipid peroxidation, stimulation of catalase activity and activation of nuclear factor kappa-B(NF-κB). Incubation of steatotic cells with silybin 50 μmol/L significantly reduced TG accumulation likely by promoting lipid catabolism and by inhibiting lipogenic pathways, as suggested by the changes in carnitine palmitoyltransferase 1(CPT-1), PPAR and SREBP-1c levels. The reduction in fat accumulation exerted by silybin in the steatotic cells was associated with the improvement of the oxidative imbalance caused by lipid excess as demonstrated by the reduction in ROS content, lipid peroxidation, catalase activity and NF-κB activation.CONCLUSION: we demonstrated the direct antisteatotic and anti-oxidant effects of silybin in steatotic cells, thus elucidating at a cellular level the encouraging results demonstrated in clinical and animal studies.

In a full-arch implant rehabilitation ad modum Branemark, the distribution of stress and strain in mandibular bone is influenced by the type, number and position of implants used. In particular, the biomechanical behaviour of the bone structure after complete osseointegration depends on the load transferred to the bone by each fixture. In this study, a finite-element analysis of two models was performed. Models of an all-on-four configuration and a six-implant configuration were compared in a worst-case scenario. A new V parameter is presented to aid the quantitative and comparative analysis of the all-on-four and six-implant configurations. The influence of orthotropy was also investigated, and a geometric change in the all-on-four configuration is presented.

Aim:Anti-CD20 monoclonal antibody is a cornerstone therapy for follicular lymphoma.Following anti-CD20 therapy,a potential decrease in CD20 antigen,and therefore a loss of the tumor target might be expected.However,the incidence and clinical significance of CD20 loss on tumor cells in patients with relapsed or refractory follicular lymphoma are unknown.This study aims to investigate the incidence and outcome of patients with relapsed or refractory follicular lymphoma patients harboring the loss of the tumor target,CD20.Methods:All consecutive adult patients with relapsed or refractory follicular lymphoma referred to the Early Drug Department at Gustave Roussy were included.The main objectives were to assess the incidence and prognosis of the loss in expression of CD20 antigen on the surface of tumor cells on patient outcome.Results:Over the study period 2013-2018,131 patients were screened for clinical trials with B-cell malignancies in the early drug department of Gustave Roussy in France.Forty-four patients presented with relapsed or refractory follicular lymphoma and 32 had tumor biopsies at the time of relapse that were retained for analysis.The median(range)age was 67.5 years(55.3-75.3)and the median number of prior anti-cancer systemic therapies was 3(2-4).At the time of relapse,CD20 expression was positive in 84%of tumors(n=27)and negative in 16%of tumors(n=5).At a median follow-up of 18.3(0.6-83.3)months,CD20 negativity was associated with a poorer prognosis with a median overall survival of 8.9 months(95%CI:2.4-19.1)in comparison to CD20 positive patients(28.3 months,95%CI:25.1-75.3 months,P=0.019).Conclusion:The loss of the tumor target antigen,CD20,occurred in 16%of patients with relapse or refractory follicular lymphoma.Due to confounding factors in patients who received anti-CD20 immunotherapy,it was not possible to formally establish the prognostic significance of CD20 negativity.However,we suggest that a check for CD20 antigen positivity nevertheless be performed to adapt subsequent therapies for p

Discrimination of background events in the PolarLight X-ray polarimeter

Despite its efficiency, the general approach of hardcoding protocol format descriptions in packet processing applications suffers from many limitations. Among the others, the lack of flexibility when needing to extend the software for supporting new protocols, and the proliferation of modules with similar functionality between different applications, resulting in decreased maintainability. The NetPDL language was defined for overcoming such limitations, allowing decoupling applications from the knowledge of the format of protocol headers. The main criticism to NetPDL relates to its supposed performance penalties;this paper demonstrates that this language can be effectively used for the dynamic generation of optimized, i.e. efficient and fast, packet-processing code, and presents the architecture of a compiler implemented for such purpose.

PolarLight is a space-borne X-ray polarimeter that measures the X-ray polarization via electron tracking in an ionization chamber.It is a collimated instrument and thus suffers from the background on the whole detector plane.The majority of background events are induced by high energy charged particles and show ionization morphologies distinct from those produced by X-rays of interest.Comparing on-source and off-source observations,we find that the two datasets display different distributions on image properties.The boundaries between the source and background distributions are obtained and can be used for background discrimination.Such a means can remove over 70%of the background events measured with PolarLight.This approaches the theoretical upper limit of the background fraction that is removable and justifies its effectiveness.For observations with the Crab nebula,the background contamination decreases from 25%to 8%after discrimination,indicative of a polarimetric sensitivity of around 0.2 Crab for PolarLight.This work also provides insights into future X-ray polarimetric telescopes.

冠向复位瓣是一种有效地治疗多个相邻牙龈退缩的手术方法．5年的长期随访可以获得稳定的效果。本研究的目的是报告10例采用冠向复位瓣联合结缔组织移植术治疗下颔后牙区至少两颗相邻牙有牙龈退缩的病例。记录基线和术后1年术区牙齿的牙周指标。入选了26颗牙龈退缩牙齿，没有出现明显的手术并发症。没有患者失访。牙龈退缩从基线时340±083mm减少到术后1年时0284±032mm。探诊深度改变未见显著差异。角化龈的增加从057±0．46mm至305±0．71mm。基线时牙龈退缩越严重．牙龈退缩改善越明显。平均根面覆盖率为91．2％±4．1％。冠向复位瓣联合结缔组织移植术是一种有效地治疗下颔后牙区多发牙龈退缩的方法。
