Autophagy plays a pro-apoptotic role in arsenic trioxide-induced cell death of liver cancer

Objective: The effects of arsenic trioxide(As2O3) on hepatocellular carcinoma have been documented widely. Autophagy plays dual roles in the survival and death of cancer cells. Therefore, we investigated the exact role of autophagy in As2O3-induced apoptosis in liver cancer ***: The viability of hepatoma cells was determined using the MTT assay with or without fetal bovine serum. The rate of apoptosis in liver cancer cells treated with As2O3was evaluated using flow cytometry, Hoechst 33258 staining, and tuNEL assays. The rate of autophagy among liver cancer cells treated with As2O3was detected using immunofluorescence, Western blot assay and transmission electron ***: Upon treatment with As2O3, the viability of HepG2 and SMMC-7721 cells was decreased in a time-and dose-dependent manner. The apoptosis rates of both liver cancer cell lines increased with the concentration of As2O3, as shown by flow cytometry. Apoptosis in liver cancer cells treated with As2O3was also shown by the activation of the caspase cascade and the regulation of Bcl-2/Bax ***, As2O3treatment induced autophagy in liver cancer cells;this finding was supported by Western blot, immunofluorescence of LC3-II and beclin 1, and transmission electron microscopy. In liver cancer cells, As2O3inhibited the phosphoinositide 3-kinase/protein kinase B/mammalian target of rapamycin(PI3K/AKT/mTOR) signal pathway that plays a vital role in both apoptosis and autophagy. The PI3K activator SC-79 partially reversed As2O3-induced autophagy and apoptosis. Furthermore, inhibiting autophagy with 3-methyladenine partially reversed the negative effects of As2O3on cell *** starvation increased autophagy and amplified the effect of As2O3on cell ***: As2O3induces apoptosis and autophagy in liver cancer cells. Active self-training for weakly supervised 3D scene semantic segmentation

Since the preparation of labeled datafor training semantic segmentation networks of pointclouds is a time-consuming process, weakly supervisedapproaches have been introduced to learn fromonly a small fraction of data. These methods aretypically based on learning with contrastive losses whileautomatically deriving per-point pseudo-labels from asparse set of user-annotated labels. In this paper, ourkey observation is that the selection of which samplesto annotate is as important as how these samplesare used for training. Thus, we introduce a methodfor weakly supervised segmentation of 3D scenes thatcombines self-training with active learning. Activelearning selects points for annotation that are likelyto result in improvements to the trained model, whileself-training makes efficient use of the user-providedlabels for learning the model. We demonstrate thatour approach leads to an effective method that providesimprovements in scene segmentation over previouswork and baselines, while requiring only a few userannotations.

Genetic parameter evaluation for growth and cold resistance traits of the giant freshwater prawn Macrobrachium rosenbergii

The giant freshwater prawn Macrobrachium rosenbergii distributed from tropical to subtropical regions,is a warm-water species,and its survival temperature is 14-35°C,which greatly limits its culture cycle and culture area in ***,it is urgent to cultivate a new high quality,high yield variety with improved cold-resistance,but the genetic parameters for cold-resistance traits are unknown in *** this study,the cold-resistance of adult *** populations was tested using the indoor artificial cooling *** were selected from 139 families of Shufeng G3 generation and cultured for 200 days.A linear mixed model was constructed by ASReml-R to evaluate the genetic parameters of the cold-resistance trait(cooling degree hours,CDH)and growth traits(body weight,BW,and body length,BL)based on the restricted maximum likelihood(REML)*** results show that the heritability of CDH was low(0.12±0.05),while the growth traits(BW and BL)had low to moderate heritability,with 0.20±0.06 for BW and 0.06±0.04 for *** phenotypic and genetic correlation between BW and BL was significantly positive,but significantly negative phenotypic and genetic correlations were detected between CDH and BW and between CDH and ***,the analysis of the differences between cold-resistance and phenotypic traits showed that the female reproductive status,exoskeleton hardness and claw number of adult prawns had a great influence on the cold-resistance of ***(P<0.05),indicating that adults with claws and hard exoskeletons are preferred as parents in subsequent breeding *** present results can be attributed to the selection and breeding of a new cold-resistant variety of ***.

Epilepsy is a neurological disorder characterized by high morbidity,high recurrence,and drug *** signaling through the excitatory neurotransmitter glutamate is intricately associated with *** glutamate receptors(mGluRs)are G protein-coupled receptors activated by glutamate and are key regulators of neuronal and synaptic *** mGluR signaling has been associated with various neurological disorders,and numerous studies have shown a close relationship between mGluRs expression/activity and the development of *** this review,we first introduce the three groups of mGluRs and their associated signaling ***,we detail how these receptors influence epilepsy by describing the signaling cascades triggered by their activation and their neuroprotective or detrimental roles in *** addition,strategies for pharmacological manipulation of these receptors during the treatment of epilepsy in experimental studies is also *** hope that this review will provide a foundation for future studies on the development of mGluR-targeted antiepileptic drugs.

Comparative analysis of four edible mushrooms based on HPLC fingerprint and pattein recognition analysis

Isolation,identification,and virulence gene analysis of pathogenic Aeromonas dhakensis in Macrobrachium rosenbergii and histopathological observation

To identify the cause of mass mortality of adult Macrobrachium rosenbergii in a farm in Gaoyou City,Jiangsu Province,China,a dominant strain named DKQ-1 was isolated from the hepatopancreas of dying *** and identified as Aeromonas dhakensis by purification culture,biochemical characterization,and 16S rRNA and gyrB gene sequence *** results of the challenge test revealed that the strain was highly pathogenic and the 50%lethal dose(LD_(50))in 72 h to *** was 1.54×10^(5)CFU/*** amplification results of virulence genes show that strain DKQ-1 carried 9 virulence genes,including ascV,aexT,aer,act,lip,ompAI,gcaT,acg,and exu,supporting the strong virulence of strain DKQ-1 to *** observation of the hepatopancreas,gills,and intestines indicated that DKQ-1 injection into *** could cause serious tissue damage,which further supported the strong virulence of this *** addition,a drug susceptibility test revealed that strain DKQ-1 was sensitive to 16 kinds of antibiotics,resistant to 9 kinds of antibiotics,and had intermediate resistance to spectinomycin and *** study is the first report of *** isolated from *** and provided a reference for the pathogen identification of bacterial diseases in ***,and for the prevention and treatment caused by ***.

Adhesion performance of alkali-activated material for 3-dimensional printing of tunnel linings at different temperatures

As one of the most widely used personal protective equipment(PPE),body armors play an important role in protecting the human body from the high-velocity impact of bullets or *** body torso and critical organs of the wear may suffer severe behind-armor blunt trauma(BABT)even though the impactor is stopped by the body armor.A type of novel composite material through incorporating shear stiffening gel(STG)into ethylene-vinyl acetate(EVA)foam is developed and used as buffer layers to reduce *** this paper,the protective performance of body armors composed of fabric bulletproof layers and a buffer layer made of foam material is investigated both experimentally and *** effectiveness of STG-modified EVA in damage relief is verified by ballistic *** parallel with the experimental study,numerical simulations are conducted by LS-DYNA®to investigate the dynamic response of each component and capture the key mechanical parameters,which are hardly obtained from field *** fully describe the material behavior under the transient impact,the selected constitutive models take the failure and strain rate effect into consideration.A good agreement between the experimental observations and numerical results is achieved to prove the validity of the modelling *** tests and simulations show that the impact-induced deformation on the human body is significantly reduced by using STG-modified EVA as the buffering *** improvement of protective performance is attributed to better dynamic properties and more outstanding energy absorption capability of the composite foam.
