Microbial phenolic metabolites are associated with better frontal lobe cognition

With increasing life expectancy， neurodegenerative diseases have become one of the leading causes of ill-health in the elderly. Preventive strategies include following healthy diets， such as the Mediterranean diet， which is particularly rich in polyphenols， bioactive compounds with neuroprotective properties. The aim of this study was to assess the association of microbial phenolic metabolites (MPM) with cognition. This cross-sectional analysis was performed with 200 participants of the PREDIMED trial (Barcelona-Clinic recruitment center). A novel method based on liquid chromatography coupled to mass spectrometry was used to identify urinary MPM (protocatechuic acid， enterodiol glucuronide， enterolactone glucuronide， urolithin B glucuronide， and vanillic acid glucuronide)， and cognitive function was evaluated with neuropsychological tests. Multivariable-adjusted ordinary least squares regression was used to assess the associations between cognitive function and MPM， and a score was calculated as the weighted sum of MPM. A higher MPM score was associated with better frontal lobe function. Among individual metabolites， vanillic acid glucuronide was correlated with frontal cognitive performance. Participants with higher concentrations of vanillic acid glucuronide and urolithin B glucuronide obtained better scores in the Color Trail Test part 2. A higher score for urinary multi-MPM was associated with better frontal cognitive performance in an older Mediterranean population.

The study aimed to explore the efficacy and potential mechanisms of a naturally aromatic cyanogenic compound-amygdalin (AMY) in treating glucocorticoid (GC)-associated necrosis of the femoral head (GANFH). We demonstrated that GC exacerbates the oxidative stress (OS) microenvironment via promoting nicotinamide adenine dinucleotide phosphate oxidase 4 (NOX4) expression in human， rat， and mesenchymal stem cells (MSCs) samples， thus generating excessive ros， leading to increased apoptosis and unbalanced osteolipogenic differentiation. Furthermore， computational docking results revealed that AMY could bind specifically to the predicted binding sites of NOX4. Additionally， AMY ameliorated the OS microenvironment of MSCs via decreasing NOX4 expression and inhibiting NOX4/ros/p38MAPK signaling， thereby reversing the GC-induced apoptosis and imbalanced osteolipogenic differentiation， and ultimately alleviating GANFH. In summary， we demonstrated for the first time that AMY attenuated apoptosis and maintained osteolipogenic differentiation balance in MSCs via specifically targeting NOX4， inhibiting NOX4/ros/p38MAPK signaling， thereby treating GANFH.

【目的】探究不同类型砧木应对重金属镉的机制,为砧木育种及在镉污染土壤中瓜类嫁接栽培安全生产提供参考。【方法】以野生西瓜类型砧木YZ1、中国南瓜类型砧木SZ111、印度与中国南瓜杂交类型砧木SZ7、葫芦类型砧木LZS为材料,利用0.6 mg·L^(-1)CdCl_(2)溶液进行水培处理,以不加镉培养的实生苗为对照,一段时间后取样测定地上部鲜质量、主根长、丙二醛(MDA)含量、超氧化物歧化酶(SOD)和过氧化物酶(POD)活性以及叶绿素荧光参数并进行分析。【结果】与对照相比,YZ1、SZ111和SZ7地上部鲜质量的影响程度差异极显著;YZ1的MDA含量极显著升高;YZ1叶片和LZS整体的SOD活性显著降低,YZ1根部SOD活性显著升高;SZ111和LZS根部及SZ7整体POD活性均显著降低,YZ1叶片POD活性极显著升高;SZ111和SZ7的Y(Ⅱ)和ETR值极显著降低,YZ1和SZ7的F_(v)/F_(m)和F_(v)/F_(o)值显著降低。【结论】SZ111和LZS这两种类型砧木抗镉能力较强,可以作为镉污染地区的瓜类砧木使用。

Ligustrum vicaryi L. is a hybrid of Ligustrum ovalifolium Hassk. var. aureo-marginatum and Ligustrum vulgale L., and displays a chlorophyll-less phenotype. Therefore it is widely used as a horticultural shrub because of its golden-color leaves. Its putative mechanism, light responses, chlorophyll synthesis and plastid development were studied. L. vicaryi has a higher level of *** acid （ALA）, but lower levels of chlorophylls compared with L. quihoui. The yellowish phenotype of L. vicaryi upper leaves could be attributed to their hampered conversion from chlorophyllide into chlorophyll a. Despite the enhanced ALA level and the decreased thylakoid stacking in plastids, L. vicaryi golden leaves contain normal levels of Lhcb transcripts and photosystem apoproteins. Furthermore, reactive oxygen species （ros） accumulation is almost the same in L. vicaryi and L. quihoui. The golden leaves often turn green and the contents of chlorophylls increase with decreasing light intensity. Dynamic changes of chlorophyll- synthesis-system under the light transition were also analyzed.

目的：探讨体外葡萄籽原花青素对氯化钴致视网膜神经节细胞RGC－5缺氧损伤模型的保护作用。方法：采用氯化钴（400μmol／L）诱导RGC－5细胞化学缺氧损伤，观察葡萄籽原花青素（40μg／mL ）对缺氧的抗损伤作用。以MTT法检测细胞存活率；Hoechst 33342荧光染色法观察细胞凋亡情况；以H2 DCFDA荧光法检测细胞内活性氧的变化；实时定量PCR检测细胞Bcl－2、半胱酰胺天冬氨酸蛋白酶（ caspase ）9和caspase 3 mRNA水平。结果：与缺氧对照组比较，葡萄籽原花青素预处理可提高细胞存活率（ P＜0．001）、减少细胞凋亡（ P＜0．001），并减少细胞内的活性氧（ P ＜0．001）；葡萄籽原花青素还显著升高 Bcl－2基因的表达（ P ＜0．001），显著降低caspase 9和caspase 3基因的表达（ P＜0．001）。结论：葡萄籽原花青素对氯化钴诱导的视网膜神经节RGC－5细胞缺氧损伤起保护作用，其保护作用可能与细胞内活性氧减少、Bcl－2基因表达上调及casepase 9和casepase 3基因表达下调有关。

目的探究重组α-Klotho蛋白对化疗药物环磷酰胺(CTX)诱导的卵巢早衰(POF)小鼠卵巢功能的保护作用。方法将55只雌性C57BL/6小鼠随机分为对照组(Control组,n=10)、CTX诱导的卵巢早衰组(POF组,n=15)、重组α-Klotho蛋白治疗组(POF+α-Klotho组,n=15)和雌二醇治疗组(POF+Estradiol组,n=15),其中POF+Estradiol组为阳性对照组。采用酶联免疫吸附试验(ELISA)法检测各组小鼠血清中卵巢激素[卵泡刺激素(FSH)、黄体生成素(LH)、雌二醇(E2)和孕酮(P)]浓度;取小鼠新鲜卵巢组织,采用试剂盒检测卵巢组织中氧化应激因子活性氧(ros)、超氧化物歧化酶(SOD)、丙二醛(MDA)和过氧化氢酶(CAT)的表达水平;采用Western blot法检测卵巢组织中凋亡相关蛋白Bcl-2、Bax、caspase-3与氧化应激信号通路Nrf2/HO-1中Keap1、Nrf2和HO-1的蛋白表达水平。结果与Control组相比,POF组、POF+α-Klotho组和POF+Estradiol组卵巢组织中ros、MDA水平和Bax、caspase-3、Keap1蛋白表达水平升高(Pros、MDA水平和Bax、caspase-3、Keap1蛋白表达水平降低(P<0.05);SOD、CAT水平和Bcl-2、Nrf2和HO-1蛋白表达水平升高(P<0.05);血清中FSH、LH的表达水平降低(P<0.05),E2、P的表达水平升高(P<0.05)。结论重组α-Klotho蛋白可能通过抑制氧化应激水平和颗粒细胞凋亡来保护CTX诱导的POF小鼠的卵巢功能,这可能与Nrf2/HO-1信号通路的激活有关。

莱菔硫烷对高糖条件下大鼠晶状体上皮细胞活性氧产生与线粒体膜电位的影响及机制研究

目的观察莱菔硫烷(sulforaphane,SFN)对高糖条件下大鼠晶状体上皮细胞(lense pithelial cell,LEC)活性氧(reactive oxygen species,ros)和线粒体膜电位(mitochondrial membran epotential,MMP)的影响,并探讨其可能的分子机制。方法人LEC系SRA01/04细胞分别用含5.5mmol·L-1葡萄糖(正常对照组)和30.5mmol·L-1葡萄糖(高糖处理组)的培养基培养。SFN干预组在高糖处理组培养基基础上加入不同浓度SFN。处理24h后,荧光探针法检测ros产生量,罗丹明123染色检测MMP的改变,Western blot观察血红素氧合酶-1(heme oxygenase-1,HO-1)的表达。为了评价SFN对LEC的保护作用与HO-1的关系,SFN干预组(30μmol·L-1)中分别加入不同浓度的HO-1激动剂钴原卟啉和抑制剂锡原卟啉,24h后观察ros产生量及MMP变化。结果与正常对照组相比,高糖处理组LEC中ros的产生量增加了29.0%,差异有统计学意义(Pros的产生量随着SFN浓度的增高而逐渐降低,当SFN浓度为30μmol·L-1时,ros产生量接近正常对照组,差异无统计学意义(P>0.05)。高糖处理组MMP水平与正常对照组相比下降了61.0%,差异有统计学意义(Pros的抑制作用并降低MMP水平,而HO-1激动剂钴原卟啉则能进一步协同SFN降低高糖诱导的ros产生,并提高MMP水平。结论 SFN能在一定程度上抑制高糖诱导的人LEC内ros产生及MMP改变,机制可能与其诱导HO-1的表达有关。
