Addressing the Role of microRNAs in Reprogramming Leaf Growth during Drought Stress in Brachypodium distachyon

Plant responses to drought are regulated by complex genetic and epigenetic networks leading to rapid reprogramming of plant growth, miRNAs have been widely indicated as key players in the regulation of growth and development. The role of miRNAs in drought response was investigated in young leaves of Brachypodium dis- tachyon, a drought-tolerant monocot model species. Adopting an in vivo drought assay, shown to cause a dramatic reduction in leaf size, mostly due to reduced cell expansion, small RNA libraries were produced from proliferating and expanding leaf cells. Next-generation sequencing data were analyzed using an in-house bioinformatics pipeline allowing the identification of 66 annotated miRNA genes and 122 new high confidence predictions greatly expand- ing the number of known Brachypodium miRNAs. In addition, we identified four TAS3 loci and a large number of siRNA-producing loci that show characteristics suggesting that they may represent young miRNA genes. Most miR- NAs showed a high expression level, consistent with their involvement in early leaf development and cell identity. Proliferating and expanding leaf cells respond differently to drought treatment and differential expression analyses suggest novel evidence for an miRNA regulatory network controlling cell division in both normal and stressed condi-tions and demonstrate that drought trigger a genetic reprogramming of leaf growth in which miRNAs are deeply involved.

Light fl ux and quality are crucial factor for setting endogenous plant circadian *** the daily rhythmicity of leaf chlorophyll content is an eff ective method to monitor the plant physiological endogenous clock in response to environmental signals such as light availability/***,we used a leaf-clip sensor to monitor diurnal rhythms in the content of chlorophyll and fl avonoids such as fl avonols and anthocyanins in three green-(Ailanthus altissima,Tilia platyphyllos and Platanus×acerifolia)and two red-leafed(Acer platanoides *** King and Prunus cerasifera ***)tree species,adapted to sun(L)or shade(S).Signifi cant diff erences in chlorophyll content(Chl)and its variations during the day were observed among treatments in all the analyzed species.S-plants had more Chl than L-plants irrespective of leaf color,and Chl variations were more distinct during the day than in *** particular,contents were lowest in the morning(9:00)and in the middle of the day(at 12:00 and 15:00),and the highest at dusk(21:00).The less evident trends in Chl variation in L-plants were attributed to a decrease in Chl content in high light,which likely masked any increases in the shaded counterparts during the *** fl avonol levels did not vary no notably during the *** sun-exposed red leaves,anthocyanins partially screened mesophyll cells from incident light,and its levels were similar to the Chl dynamics in the shaded *** study provides new bases for further work on endogenous rhythms of plant pigments and improves our understanding of plant physiology in the context of day/night rhythmicity.

New Primers for Denaturing Gradient Gel Electrophoresis Analysis of Nitrate-Reducing Bacterial Community in Soil

The narG gene is frequently used as a molecular marker for bacterial nitrate-reducing community analysis. In this study, a new set of primers targeting the narG gene was designed and applied to semi-nested polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) assay. The potential of the new primers was verified on DNA directly extracted from soils from five different experimental sites distributed in Central and Southern Italy. Specificity of the primers was determined by excision, amplification, and sequencing of bands resolved by DGGE. A phylogenetic analysis showed the correlation between the sequences retrieved from the soils studied and the narG sequences from β and γ-Proteobacteria. These primers expanded the existing molecular tools for ecological study on the size and diversity of nitrate-reducing bacterial community in soil.

Incidental gallbladder cancer during laparoscopic cholecystectomy:Managing an unexpected finding

ISWI ATP-dependent remodeling of nucleoplasmic u-speckles in the brain of Drosophila melanogaster

Heterogeneous nuclear ribonucleoproteins（hn RNPs） belong to the RNA-binding proteins family. They are involved in processing heterogeneous nuclear RNAs（hn RNAs） into mature m RNAs. These proteins participate in every step of m RNA cycle, such as m RNA export, localization, translation, stability and alternative splicing. At least 14 major hn RNPs, which have structural and functional homologues in mammals, are expressed in Drosophila melanogaster. Until now, six of these hn RNPs are known to be nucleus-localized and associated with the long non-coding RNA（lnc RNA） heat shock responsive u（hsru）in the omega speckle compartments（u-speckles）. The chromatin remodeler ISWI is the catalytic subunit of several ATP-dependent chromatin-remodeling complexes, and it is an essential factor for organization of u-speckles. Indeed, in ISWI null mutant, severe defects in u-speckles structure are detectable. Here, we clarify the role of ISWI in the hn RNPs-hsru interaction. Moreover, we describe how ISWI by its remodeling activity, controls hsru and hn RNPs engagement in u-speckles. Finally, we demonstrate that the sequestration of hn RNPs in u-speckles nuclear compartment is a fundamental event in gene expression control and represents a key step in the regulation of several pathways.

采用热裂解-气相-质谱(Pyr-TMAH-GC/MS)技术对长期不同施肥处理下太湖地区黄泥土本体土壤及水稳性团聚体中颗粒有机碳(POC)的化学组分进行了研究。结果表明,POC的热裂解产物主要来源于脂肪族化合物和木质素类化合物。木质素类化合物主要以愈疮木基丙烷(G)和p-羟苯基(P)两种木质素单体为主,从而揭示黄泥土POC积累中草本植物(水稻、油菜的残茬和秸秆)的输入贡献。脂肪族化合物主要为C8-C30脂肪酸甲基酯(FAMEs),且大多为偶数碳结构。施肥改变了各裂解产物在本体土壤和水稳性团聚体中的分布,从而使得不同施肥处理下各类化合物对POC稳定性的贡献出现了差异。化肥配施猪粪下木质素类化合物以及微生物源的脂肪族化合物均贡献于本体土壤POC的积累与稳定,而化肥配施秸秆下本体土壤POC的化学稳定性主要归因于高等植物源化合物的贡献,其次为微生物源脂肪族化合物。化肥配施有机肥下水稳性团聚体中较高的POC主要归因于植物源有机物质的分解贡献。本研究揭示了合理施肥下水稻土中有机碳的化学抗性组分在土壤有机碳积累中的促进作用,各组分的相对分布差异可指示有机碳积累的不同来源。

采用固体交叉极化魔角自旋^(13)C核磁共振(CPMAS^(13)C-NMR)波谱技术对长期不同施肥处理下红壤性水稻土和太湖地区黄泥土本体土壤以及水稳性团聚体中颗粒有机质(POM)的化学结构特征进行了研究。结果表明,不同施肥处理下本体土壤和不同粒径水稳性团聚体中POM的结构组成相似,主要由烷氧C、烷基C和芳香C组成,其中以烷氧C含量最高。施肥改变了本体土壤POM中各类C原子的相对百分含量,有机肥以及化肥配施有机肥条件下烷氧C含量明显降低,芳香C和酚基C含量有不同程度的增加,表明POM的稳定性增强;单施化肥下烷氧C含量最高,而烷基C、芳香C和芳香度均最低,POM的稳定性减弱,不利于POM的积累。施肥改变了黄泥土不同粒径水稳性团聚体中POM各类C原子的相对百分含量,从而使得不同粒径中POM对其团聚体的稳定性作用发生变化;而红壤性水稻土不同粒径水稳性团聚体POM各类C原子的相对百分含量并未明显受到施肥措施的影响。
