Background： This study evaluated the impact of hormonal modulation at the onset of proestrus on ovarian response and uterine gene expression of beef ***： A total of 172 anestrous beef cows were assigned to one of four groups according to the treatment with estradiol cypionate（ECP） and/or equine chorionic gonadotropin（eCG） [CON（n = 43）, ECP（n = 43）, eCG（n = 44）and ECP ＋ eCG（n = 42）].Results： ECP-treated cows（ECP and ECP ＋ eCG groups） presented greater occurrence of estrus（44.6% vs. 65.4%; P = 0.01） and pregnancy per AI [47.1% vs. 33.3%; P = 0.07], but similar progesterone（P4） concentration at subsequent diestrus than cows not treated with ECP（CON and eCG groups）. Nonetheless, eCG-treated cows（eCG and ECP ＋ eCG groups） presented larger follicle at timed AI（12.6 ± 0.3 vs. 13.5 ± 0.3 mm; P = 0.03）, greater ovulation rate（96.5%vs. 82.6%; P = 0.008） and greater P4 concentration at d 6（3.9 ± 0.2 vs. 4.8 ± 0.2 ng/mL; P = 0.001） than cows not treated with eCG（CON and ECP groups）. Next, cows with a new corpus luteum 6 d after TAI were submitted to uterine biopsy procedure. Uterine fragments [CON（n = 6）, ECP（n = 6）] were analyzed by RNA-Seq and a total of 135 transcripts were differentially expressed between groups（73 genes up-regulated by ECP treatment）. Subsequently,uterine samples were analyzed by q PCR（genes associated with cell proliferation）. ECP treatment induced greater abundance of PTCH2（P = 0.07） and COL4 A1（P = 0.02）, whereas suppressed EGFR（P = 0.09） expression. Conversely, eCG treatment increased abundance of HB-EGF（P = 0.06）, ESR2（P = 0.09）, and ITGB3（P = 0.05）, whereas it reduced transcription of ESR1（P = 0.05）. Collectively, supplementation with ECP or eCG at the onset of proestrous of anestrous beef cows influenced ovarian responses, global and specific endometrial gene ***： Proestrus estradiol regulate the endometrial transcriptome, particularly stimulating proliferative activity in the endometrium.

BACKGROUNd Endoscopic drainage remains the treatment of choice for unresectable or inoperable malignant distal biliary obstruction(MdBO).AIM To compare the safety and efficacy of plastic stent(PS)vs self-expanding metal stent(SEMS)placement for treatment of *** This meta-analysis was developed according to Preferred Reporting Items for Systematic Reviews and Meta-Analysis guidelines.A comprehensive search was performed in MEdLINE,Cochrane,Embase,Latin American and Caribbean Health Sciences Literature,and grey literature to identify randomized clinical trials(RCTs)comparing clinical success,adverse events,stent dysfunction rate,reintervention rate,duration of stent patency,and mean *** difference(Rd)and mean difference(Md)were calculated and heterogeneity was assessed with I2 *** analyses were performed by SEMS *** Twelve RCTs were included in this study,totaling 1005 *** was no difference in clinical success(Rd=-0.03,95%confidence interval[CI]:-0.01,0.07;I 2=0%),rate of adverse events(Rd=-0.03,95%CI:-0.10,0.03;I2=57%),and mean patient survival(Md=-0.63,95%CI:-18.07,19.33;I2=54%)between SEMS vs PS ***,SEMS placement was associated with a lower rate of reintervention(Rd=-0.34,95%CI:-0.46,-0.22;I2=57%)and longer duration of stent patency(Md=125.77 d,95%CI:77.5,174.01).Subgroup analyses revealed both covered and uncovered SEMS improved stent patency compared to PS(Rd=152.25,95%CI:37.42,267.07;I2=98%and Rd=101.5,95%CI:38.91,164.09;I2=98%;respectively).Stent dysfunction was higher in the covered SEMS group(Rd=-0.21,95%CI:-0.32,-0.1;I²=205%),with no difference in the uncovered SEMS group(Rd=-0.08,95%CI:-0.56,0.39;I²=87%).CONCLUSION While both stent types possessed a similar clinical success rate,complication rate,and patient-associated mean survival for treatment of MdBO,SEMS were associated with a longer duration of stent patency compared to PS.

The authors describe long- term follow- up (mean, 5 years) in patients with anterior (AN) (n = 6) or centromedian (n = 2) thalamic deep brain stimulation (dBS) for epilepsy. Five patients (all AN) had ≥ 50% seizure reduction, although benefit was delayed in two until years 5 to 6, after changes in antiepileptic drugs. dBS electrode implantation in AN patients was followed by seizure reduction 1 to 3 months before active stimulation, raising the possibility of a beneficial microthalamotomy effect.

Biocatalytic production of chitosan polymers from shrimp shells,using a recombinant enzyme produced by Pichia pastoris

Chitosan has a unique chemical structure with high charge density, reactive hydroxyl and amino groups, and extensive hydrogen bonding. Chitin deacetylase (EC 3.***.1.41) catalyzes the hydrolysis of the N-acetamido groups of N-acetyl-d-glucosamine residues in chitin, converting it to chitosan and releasing acetate. The entire ORF of the CdA2 gene encoding one of the two isoforms of chitin deacetylase from Saccharomyces cerevisiae was cloned in Pichia pastoris. The Tg (Cda2-6xHis)p was expressed at high levels as a soluble intracellular protein after induction of the recombinant yeast culture with methanol, and purified using nickel-nitrilotriacetic acid chelate affinity chromatography, resulting in a protein preparation with a purity of >98% and an overall yield of 79%. Chitin deacetylase activity was measured by a colorimetric method based on the O-phthalaldehyde reagent, which detects primary amines remaining in chitinous substrate after acetate release. The recombinant enzyme could deacetylate chitin, chitobiose, chitotriose and chitotetraose, with an optimum temperature of 50°C and pH 8.0, determined using oligochitosaccharides as the substrates. The recombinant protein was also able to deacetylate its solid natural substrate, shrimp chitin, to a limited extent, producing chitosan with a degree of acetylation (dA) of 89% as determined by Fourier transform infrared spectroscopy. The degree of deacetylation was increased by pre-hydrolysis of crystalline shrimp chitin by chitinases, which increased the deacetylation ratio triggered by chitin deacetylase, producing chito-oligosaccharides with a degree of acetylation of 33%. The results described here open the possibility to use the rCda2p, combined with chitinases, for biocatalytic conversion of chitin to chitosan with controlled degrees of deacetylation. We show herein that the crystalline chitin form can be cleanly produced in virtually quantitative yield if a combined and sequential enzyme treatment is performed.

Brazilian Buffalo Genetic Variability by Cross-Specific Microsatellite Set

Buffaloes （Bubalus bubalis） are widely distributed and were introduced to Brazil in 1895. Most of the molecular genetic characterization of buffaloes has been done with cross-specific （cattle） markers, but few of them include Brazilian populations. Nineteen commonly used cattle microsatellites were tested to develop a multiplexed set of microsatellites and characterize Brazilian buffalo. Three PCR mixes were finally developed with the 11 markers that succeed in amplify and were polymorphic （58%）. The average number of alleles was 5.42, with an average observed and expected heterocigozity of 0.441 and 0.695, respectively. As it was expected, Brazilian buffalo variability was lower than the previously reported from the domestication centres （China and India）, but higher than the seriously selected European populations. The exclusion power calculated for the eleven markers in Brazilian buffalo was 0.9999999996, this allows its use in dNA based traceability.

Neisseria meningitidis is a gram-negative diplococcus which causes invasive disease. The most frequent clinical manifestations caused by infection with this pathogen, are meningitis and fulminant septic shock. More benign forms with a subacute or chronic pattern are rare but still can evolve to meningitis and have a fatal outcome. This entity is difficult to recognize, as febrile erythema with otherwise good general condition may simulate other diseases, namely viral infections. The authors reported the cases of two healthy young adults who were misdiagnosed with a viral infection. What they really had was a meningococcemia without meningitis presenting with mild and unspecific symptoms, such as, intermittent fever and a discrete macular rash. We pretend to highlight this rare, unspecific and heterogeneous symptoms which can provoke negligence of a potentially life threatening condition.

Occurrence of Antimicrobials in River Water Samples from Rural Region of the State of Rio de Janeiro, Brazil

The occurrence of antimicrobials in the aquatic environment and drinking water has raised the question of their impact on the environment and public health. Animal production is one of the most expressive activities of Brazilian agribusiness. In order to ensure the productivity and competitiveness of the sector, the use of drugs for therapeutic and prophylactic purposes is a common practice. due to the continuous release of antimicrobials into the environment, the aim of this study was to compare the frequency of detection of tetracyclines and sulfonamides in surface water collected from rural areas in Lidice district of Rio Claro, in the State of Rio de Janeiro, Brazil. An investigative study was conducted with 24 river water samples analyzed by high-performance liquid chromatography coupled to tandem mass spectrometry and the aim of this study was to determine residues of sulfonamides and tetracyclines based on the USEPA method 1694. The results indicated the presence of sulfamethoxazole and oxytetracycline concentrations at the ng·L-1 level. The applied method showed overall good performance with recoveries above 57%, method detection limits ≤ 7.17 ng·L-1, method quantification limits ≤ 23.90 ng·L-1 and good linearity.

Characterization and Lifetime Estimation of High density Polyethylene Containing a Prodegradant Agent

High density polyethylene (HdPE) samples, containing different concentrations of prodegradant additive d2w?, were prepared. The properties of the samples were evaluated through differential scanning calorimetry (dSC), Fourier transform infrared spectroscopy (FTIR), rheometry, and scanning electron microscopy (SEM). The work contributes to decreasing the products made of non-biodegradable polymeric materials derived from fossil sources which are have become a problem due to their increasingly inappropriate disposal and long degradation time in the environment. The obtained results indicated that there was no degradation of the samples due to processing. No significant changes in melting temperature, crystallinity, viscoelastic behavior, molecular weight and chemical composition were observed. Images from SEM analysis showed particles on HdPE surface, attributed to prodegradant additive d2w?. Oxidation Onset Temperature (OOT) results showed that the additive d2w? accelerated the degradation of HdPE. The activation energy (Ea) was determined by Ozawa-Wall-Flynn method. The obtained values were used for lifetime estimation of the samples. At 25°C, HdPE with d2w? showed a lifetime 50% higher than that of HdPE without this additive. This fact is attributed to the presence of stabilizers in masterbatch d2w? and the absence of oxygen in thermogravimetric analysis.
