Dissolution method for delapril and manidipine combination tablets based on an absorption profile of manidipine

The present study describes the development and validation of a dissolution method for delapril （DEL） and manidipine （MAN） combination tablets, using a simulated absorption profile based on in vivo data for MAN. The suitable in vitro dissolution profile for this formulation was obtained using 900 mL of citrate buffer pH 3.2 at 37 ℃ ± 0.5 ℃ as dissolution medium and USP apparatus 2 （paddle） at 75 rpm. All samples were analyzed by a liquid chromatography （LC） method. Under these conditions, a significant linear relationship between the absorbed （calculated by deconvolution approach） and dissolved fractions of MAN was obtained （R=0.997） and an in vivo-in vitro （IVIV） correlation for this particular formulation containing MAN can be established. Validation parameters for dissolution methodology such as the specificity, linearity, accuracy and precision were also evaluated according to the international guidelines, giving results within the acceptable range. Therefore, the proposed dissolution conditions can be applied for the simultaneous release analysis of DEL and MAN from the solid dosage form, contributing to the improvement of the quality control of pharmaceutics and minimizing the number of bioavailability studies.

Hydralazine Hydrochloride:An Alternative Complexometric Reagent for Total Iron Spotometric Determination

An alternative spectrophotometric method has been developed for total iron determination using flow injection analysis (FIA). The procedure is based on the coordination reaction between hydralazine and Fe2+ ions, which results in the formation of a purple complex monitored at 538 nm. For determination of total iron, Fe3+ ions were reduced using ascorbic acid. Under optimized conditions, a linear calibration graph (0.1 - 6.0 ?g?ml–1;n = 6) was obtained. The method allows LOD (3? of blank/slope = 0.06 ?g?ml–1) and LOQ (10? of blank/slope = 0.22 ?g?ml–1). The RSD ((s/ ) × 100) for a mixed standard containing 0.60 ?g?ml–1 Fe2+ and Fe3+ was 0.10% (n = 10). Recoveries of spiked samples were 94.3% - 106.0%. The analytical frequency was 60 h–1. The effect of possible interferences has been studied. The procedure was successfully applied for analysis of environmental samples. The real samples results were comparable with those obtained by the official method considering a paired t-test and 95% of confidence level.

The aim of this study was to assess the stress patterns on the peri-implant zone and residual alveolar ridge in different overdenture attachment system designs by the photoelasticity method. Four attachments systems were tested: O-ring, ERA, Bar-clip, and Bar-clip/O-ring association. The prostheses were loaded with 100 N in five pre-determined points and the photoelastic model was evaluated by a circular polariscope. The anterior load, O-ring, and ERA showed better stress distribution in relation to the bars systems, which presented stress levels surrounding implants. The molar load, Bar-clip/O-ring association, presented the biggest stress concentration on the peri-implant region in relation to the others. When the second molar was loaded, there was a concentration of stress in the alveolar ridge in all situations analyzed. Within the limitation of this “in vitro” study, it could be concluded that there were biomechanical differences among the attachments systems analyzed, principally between isolated and the bars systems. The O-ring showed better stress distribution and the Bar-clip/O-ring showed higher stress concentration.

COVID-19 and Gestational Diabetes: A Systematic Review

In March 2020, the World Health Organization declared a pandemic state in response to the number of infected and killed by COVID-19. Gestational diabetes (GD) is characterized by physiological changes making pregnant women even more vulnerable to viral respiratory infections, such as SARS-CoV-2. For these reasons and due to the restriction of data related to DMG and the involvement by COVID-19, the importance of raising information about these two conditions is evident. Therefore, this study aimed to perform a literature review about the complications of COVID-19 in patients with GD. MEDLINE-PubMed, EMBASE, and Scielo databases were searched and followed the PRISMA guidelines. Only four studies were included in the systematic review. The complications and mortality are more significant in pregnant women with diabetes since hyperglycemia promotes immune changes in the inflammatory state and worsens insulin resistance. The imbalance in the immune response favors the development of infections, the most likely being viral respiratory infections. On the other hand, other studies show insufficient recent evidence to indicate that pregnant women are more susceptible to infection with severe acute coronavirus respiratory syndrome. There are divergences between studies concerning complications related to COVID-19 in patients with GD. In view of this scenario and from the information obtained, it is noted that the literature is scarce regarding the worsening of COVID-19 in patients with GD. Therefore, it is necessary to continue research on these mechanisms that may justify their correlation.
