In the two past decades, a number of communications, case-control studies, and retrospective reports have appeared in the literature with concerns about the development of a complex set of clinical, laboratory and histological characteristics of a liver graft dysfunction that is compatible with autoimmune hepatitis. The de novo prefix was added to distinguish this entity from a pre-transplant primary autoimmune hepatitis, but the globally accepted criteria for the diagnosis of autoimmune hepatitis have been adopted in the diagnostic algorithm. Indeed, de novo autoimmune hepatitis is characterized by the typical liver necroinflammation that is rich in plasma cells, the presence of interface hepatitis and the consequent laboratory findings of elevations in liver enzymes, increases in serum gamma globulin and the appearance of nonorgan specific auto-antibodies. Still, the overall features of de novo autoimmune hepatitis appear not to be attributable to a univocal patho-physiological pathway because they can develop in the patients who have undergone liver transplantation due to different etiologies. Specifically, in subjects with hepatitis C virus recurrence, an interferon-containing antiviral treatment has been indicated as a potential inception of immune system derangement. Herein, we attempt to review the currently available knowledge about de novo liver autoimmunity and its clinical management.

The correct folding and assembly of newly synthesized secretory proteins are monitored by the protein quality control system of the endoplasmic reticulum （ER）. Through interactions with chaperones such as the binding protein （BiP） and other folding helpers, quality control favors productive folding and sorts for degradation defective proteins. A major route for quality control degradation identified in yeast, plants, and animals is constituted by retrotranslocation from the ER to the cytosol and subsequent disposal by the ubiquitin/proteasome system, but alternative routes involving the vacuole have been identified in yeast. In this study, we have studied the destiny of sGFP418, a fusion between a secretory form of GFP and a domain of the vacuolar protein phaseolin that is involved in the correct assembly of phaseolin and in BiP recognition of unassembled subunits. We show that sGFP418, despite lacking the phaseolin vacuolar sorting signal, is delivered to the vacuole and fragmented, in a process that is inhibited by the secretory traffic inhibitor brefeldin A. Moreover, a fusion between GFP and a domain of the maize storage protein γ-zein involved in zein polymerization also undergoes post-translational fragmentation similar to that of sGFP418. These results show that defective secretory proteins with permanently exposed sequences normally involved in oligomerization can be delivered to the vacuole by secretory traffic. This strongly suggests the existence of a plant vacuolar sorting mechanism devoted to the disposal of defective secretory proteins.

Molecular Mechanism of the Specificity of Protein Import into Chloroplasts and Mitochondria in Plant Cells

Plants possess both types of endosymbiotic organelles, chloroplasts and mitochondria. Transit peptides and presequences function as signal sequences for specific import into chloroplasts and mitochondria, respectively. However, how these highly similar signal sequences confer the protein import specificity remains elusive. Here, we show that mitochondrial- or chloroplast-specific import involves two distinct steps, specificity determination and translocation across envelopes, which are mediated by the N-terminal regions and functionally interchangeable C-terminal regions, respectively, of transit peptides and presequences. A domain harboring multiple-arginine and hydrophobic sequence motifs in the N-terminal regions of presequences was identified as the mitochondrial specificity factor. The presence of this domain and the absence of arginine residues in the N-terminal regions of otherwise common targeting signals confers specificity of protein import into mitochondria and chloroplasts, respectively. AtToc159, a chloroplast import receptor, also contributes to determining chloroplast import specificity. We propose that common ancestral sequences were functionalized into mitochondrial- and chloroplast-specific signal sequences by the presence and absence, respectively, of multiple-arginine and hydrophobic sequence motifs in the N-terminal region.

In large fusion experiments, such as tokamak devices, there is a common trendfor slow control systems. Because of complexity of the plants, the so-called 'Standard Model' (SM)in slow control has been adopted on several tokamak machines. This model is based on a three-levelhierarchical control: 1) High-Level Control (HLC) with a supervisory function; 2) Medium-LevelControl (MLC) to interface and concentrate I/O field equipments; 3) Low-Level Control (LLC) withhard real-time I/O function, often managed by PLCs. FTU control system designed with SM concepts hasunderwent several stages of developments in its fifteen years duration of runs. The latestevolution was inevitable, due to the obsolescence of the MLC CPUs, based on VME-MOTOROLA 68030 withOS9 operating system. A large amount of C code was developed for that platform to route the dataflow from LLC, which is constituted by 24 Westinghouse Numalogic PC-700 PLCs with about 8000field-points, to HLC, based on a commercial Object-Oriented Real-Time database on Alpha/CompaqTru64platform. Therefore, we have to look for cost-effective solutions and finally a CompactPCI—Intelx86 platform with Linux operating system was chosen. A software porting has been done, taking intoaccount the differences between OS9 and Linux operating system in terms of Inter/Network ProcessesCommunications and I/O multi-ports serial driver. This paper describes the hardware/softwarearchitecture of the new MLC system, emphasizing the reliability and the low costs of the open sourcesolutions. Moreover, a huge amount of software packages available in open source environment willassure a less painful maintenance, and will open the way to further improvements of the systemitself.

A group of 19th century inmates dead in the prison of Parma are the protagonist of an incredible scientific collection. Lorenzo Tenchini started the creation of this collection and dedicated his work and his studies to its completeness. Anatomist and academic, Lorenzo Tenchini (1852–1906) dedicated his scientific studies to macroscopic anatomy, particularly about central nervous system and its correlation with psychic function. In 1881 he became ordinary professor in Normal Human Anatomy at the University of Parma dedicating himself to the study of the anatomical organization of the brain and psychic and social disturbs. During the study of the skulls and brains of psychotic patients and the deformations of skulls belonging to patients admitted in the Hospital of Brescia, he started a collaboration with Alessandro Cugini (1829–1913), founder of the Institute of Legal Medicine at the University of Parma. Tenchini realized an anatomical collection, preserved today in the Museum of Biomedicine of the University of Parma. This collection represents the master-piece of his research carried out during his academic activity and still a unicum in the west-ern world, as there are no similar collection assembling such a multidisciplinary information. The peculiarity of this collection is due not only to the scientific interest of the anatomic samples and their full clinical documentation, but also to the methods employed in order to realize it. At the end of the 19th century, as a student of Cesare Lombroso (1835–1909), Tenchini based his work on the study of the face, the skull and brain of each dead inmate of Parma's prison or Colorno's mental hospital. These individuals as protagonists of Tenchini's collection, leave a legacy identifiable as scientific heritage. Their skulls and brains, the reproduction of their faces through ceroplastic and other anatomical samples treated with other techniques, are accompanied by an autoptic and psychiatric full documentation, allowing t
