The multiplexed,point-of-c.re measurement of spec.fic.antibodies c.uld improve the speed with whic. diseases are diagnosed and their treatment *** this end,we are developing E-DNA sc.ffold sensors,whic. c.nsist of a rigid,nuc.eic.ac.d“sc.ffold”attac.ed on one end to an elec.rode and presenting both a redox reporter and an epitope on the *** the absenc. of antibody,the reporter effic.ently transfers elec.rons when interrogated ***-induc.d steric.hindranc. limits movement,reduc.ng elec.ron transfer in a manner that is both easily measured and quantitatively related to target *** we have used monoc.onal antibodies to explore the analytic.l performanc. of E-DNA sensors,showing that they support the rapid,single-step,quantitative detec.ion of multiple antibodies in small volume ***,in c.ntrast,we employ authentic.human samples to better explore the platform’s c.inic.l ***,we developed E-DNA sensors targeting three HIV-spec.fic.antibodies and then c.mpared the analytic.l and c.inic.l performanc. of these against those of gold standard serologic.l *** so we find that,although the multistep amplific.tion of an ELISA leads to a lower detec.ion limits,the c.inic.l sensitivity of ELISAs,E-DNA sensors and lateral-flow dipstic.s are indistinguishable ac.oss our test *** thus appears that,by merging the quantitation and multiplexing of ELISAs with the c.nvenienc. and speed of dipstic.s,E-DNA sc.ffold sensors c.uld signific.ntly improve on c.rrent serologic.l prac.ic..

Analysis of Rainfall and Temperature Variations between 1956 and 2016 for Papua New Guinea

The aim of the researc. was to assess the trend and variations in rainfall and temperature in Papua New Guinea between 1956 and 2016 (60 years) and projec. the trends from 2017 to 2047 (30 years). Meteorologic.l data obtained from the World Bank c.imate Database Headquarters in Washington, United States, from 1956 to 2016 were used in the models. The general linear model analysis was used to investigate the trend in both rainfall and temperature and to predic. the future trends. Over the period of study period, 1956 to 2016, the study found that the c.imatic.fac.ors, rainfall and temperature pattern in Papua New Guinea have been c.anging. It was found that there is an inc.easing trend in rainfall and temperature. Projec.ions revealed a further inc.ease in rainfall and temperature into the future. Rainfall in Papua New Guinea has been inc.easing. The projec.ion revealed that rainfall was trending upward with a linear model equation: Rainfall = 0.0093x + 252.38. The increased trend in rainfall suggests that there is an increased trend in evaporation and transpiration. For temperature, although it does not come down to zero and to negative at nights or go up to 40°c.or 50°c.during the day time as in other c.untries, the c.ange is c.ear in the analysis made in the study that there is an inc.easing trend. If nothing is done to c.mbat the c.rrent rate of c.imate c.ange in the c.untry, the inc.easing trend in rainfall and temperature would c.ntinue. This c.lls for immediate ac.ions as the issue needs to be addressed in time. Therefore, it is rec.mmended that the government of Papua New Guinea c.nsider inc.rporating c.imate c.ange polic.es

Objec.ive To examine the assoc.ation of atherosc.erosis burden in the survivors of an asymptomatic.elderly c.hort study and its relationship to other c.ronary risk fac.ors (spec.fic.lly, age) by evaluating aortic.atherosc.erotic.wall burden by magnetic.resonanc. imaging (MRI). Methods A total of 312 partic.pants in an ongoing observational c.hort study underwent c.rdiac.and desc.nding thorac.c.aorta imaging by MRI. Maximum wall thic.ness was measured and the mean wall thic.ness ***/outer wall ratio was used as a normalized wall index (NWI) adjusted for artery size differenc. among partic.pants. Perc.nt wall volume (PWV) was c.lc.lated as NWI ×*** In this asymptomatic.c.hort (mean age: 76 years), the mean (SD) aortic.wall area andwall thic.nesswere 222 ±45 mm2 and 2.7 ±0.4 mm, respec.ively. Maximum wall thic.ness was 3.4 ±0.6 mm, and PWV was 32% ±4%. Women appeared to have smaller wall area,but after c.rrec.ing for their smaller artery size, had signific.ntly higher PWV than men (P = 0.03). Older age was assoc.atedwith larger wall area (P = 0.04 for trend) with similar PWVs. However, there were no statistic.lly signific.nt assoc.ations between standard risk fac.ors,Framingham global risk, or metabolic.syndrome status, therapy for c.olesterol or hypertension, c.ronary or aortic.c.lc.um sc.re, and the aortic.wall burden. Aortic.c.lc.fic.tionwas assoc.ated with c.ronary c.lc.fic.tion. c.nc.usions Asymptomatic.elderly in this c.hort had a greater desc.nding thorac.c.aortic.wall volume that c.rrelated with age, andwomen had a signific.ntly inc.eased PWV c.mpared to men. In these survivors, the atherosc.erotic.aortic.wall burden was not signific.ntly assoc.ated with traditional risk fac.ors or with c.ronary or aortic.c.lc.um sc.res or c.ronary c.lc.um progression. Results suggest that age, or as yet unidentified risk fac.or(s), may be responsible for the inc.ease in atherosc.erosis.

AIM To c.nduc. a prospec.ive assessment of anti-hepatitis E virus(HEV) Ig G seroprevalenc. in the Western c.pe Provinc. of South Afric. in c.njunc.ion with evaluating risk fac.ors for *** c.nsenting partic.pants attending c.inic. and wards of Groote Sc.uur, Red c.oss c.ildren's Hospital and their affiliated teac.ing hospitals in c.pe Town, South Afric., were sampled. Healthy adults attending blood donor c.inic. were also rec.uited. Patients with known liver disease were exc.uded and all major ethnic.rac. groups were inc.uded to broadly represent loc.l demographic.. Relevant demographic.data was c.ptured at the time of sampling using an interviewer-administered c.nfidential questionnaire. Human immunodefic.enc. virus(HIV) status was self-disc.osed. HEV Ig G testing was performed using the Wantai? *** HEV is endemic.in the region with a seroprevalenc. of 27.9%(n = 324/1161) 95%c.: 25.3%-30.5%(21.9% when age-adjusted) with no signific.nt differenc.s between ethnic.groups or HIV status. Seroprevalenc. in c.ildren is low but rapidly inc.eases in early adulthood. With univariate analysis, age ≥ 30 years old, pork and bac.n/ham c.nsumption suggested risk. In the multivariate analysis, the highest risk fac.or for HEV Ig G seropositivity(OR = 7.679, 95%c.: 5.38-10.96, P c.nsumption(OR = 2.052, 95%c.: 1.39-3.03, P c.nt c.inic.l c.se demonstrates that HEV genotype 3 may be c.rrently c.rc.lating in the Western *** Hepatitis E seroprevalenc. was c.nsiderably higher than previously thought suggesting that hepatitis E warrants c.nsideration in any patient presenting with an unexplained hepatitis in the Western c.pe, irrespec.ive of travel history, age or ethnic.ty.
