Metagenomic approaches are recently used for searching novel open reading frames （ORFs） coding enzymes employed in pharmaceutical, rood industries, etc. In this study, a metagenornic library was constructed from Chumathang hotspring sediment DNA. The library consisted of approximately 9,000 clones and was screened for protease activity. A clone exhibiting protease activity was identified and named CHprol. Sequencing of CHprol revealed that the ORF encoded a functional protein of 363 amino acids belonging to peptidase S8-S53 superfamily. CHprol shared 41% sequence similarity with a reported protease （subtilase family） and 35 % structural similarity with the crystal structure of Pro-Tk sps. of Thermococcus kodarkaenasis. In silico modeling the 3D structure of CHprol showed that it has two beta sheets, 10 alpha helices and 11 strands. Catalytic triad prediction implied CHprol to be a serine protease. The optimum temperature and pH of the purified protease were found to be 80 ℃ and 11.0, respectively. The enzyme was active at 5 % concentration of hydrogen peroxide and retained 60 % of activity at 10 % concentration. The thermotolerant, alkalophilic and oxidant stable properties of the protease make it a potential can- didate for biotechnological applications.

Effect of baffles on performance of fluid catalytic cracking riser

Increasing demand of automobile fuel and a need to process heavier crude oil makes it imperative to find improvements to the design of existing fluid catalytic cracking （FCC） units. Several modifications to the design of the riser section of FCC units have been suggested in previous studies including： improved feed nozzle designs, multiple nozzle configurations, internal baffles, and novel two-stage-riser systems. In this study, we investigate the effects of baffles on the performance of FCC risers using computational fluid dynamics simulations. In this study, predictions from a basis model （without baffles） are compared with those from four different configurations including：（i） 5-cm baffles at 5-m spacing, （ii） 7.5-cm baffles at 5- m spacing, （iii） 10-cm baffles with 5-m spacing, （iv） lO-cm baffles at 2.5-m spacing, and （v） lO-cm baffles at 1-m spacing. The baffles force the catalyst away from wails toward the center of the riser, enhancing the radial dispersion of the catalyst and the heat transfer inside the riser. The use of longer baffles and smaller spacings further increases the dispersion, yielding more homogeneous radial profiles. The changes in the radial dispersion result in variations in the conversion, yields, and pressure drops. The baffles increase conversion of vacuum gas oil （VGO） and the yield of gasoline. However, the simulations showed that longer baffles and a larger number of baffles did not always give a higher yield or higher conversion. Among the simulated configurations, the 5-cm baffles at 5-m spacing gave the highest conversion of VGO, whereas the 10-cm baffles at 1 -m spacing resulted in the highest yield of the gasoline. Thus, rational optimization of baffle configurations is required to achieve optimal performance.

Candida endocarditis is extremely rare in term neonates,and gall bladder involvement due to candidemia has never been reported amongst neonates and infants.A term,appropriate for gestational age neonate developed Candida tropicalis blood stream infection in second week of *** was started on conventional amphotericin ***,he failed to show any clinical improvement,and candidemia keep on *** sanctuary sites screening revealed multiple echogenic masses in heart(vegetations) and gall *** changing the treatment to liposomal amphotericin B and fluconazole,he recovered clinically,echogenic masses in gall bladder disappeared,and intracardiac vegetations decreased in size.

Cells are crowded microenvironments filled with macromolecules undergoing constant phys- ical and chemical interactions. The physicochemical makeup of the cells aff）cts various cellular responses, determines cell-cell interactions and influences cell decisions. Chemical and physical properties diff）r between cells and within cells. Moreover, these properties are subject to dynamic changes in response to environmental signals, which often demand adjustments in the chemical or physical states of intracellular molecules. Indeed, cellular responses such as gene expression rely on the faithful relay of information from the outside to the inside of the cell, a process terrned signal transduction. The signal often traverses a complex path across subcellular spaces with variable physical chemistry, sometimes even influencing it. Understanding the molecular states of such signaling molecules and their intracellular environments is vital to our understanding of the cell. Exploring such intricate spaces is possible today largely because of experimental and theoretical tools. Here, we focus on one tool that is commonly used in chemical physics studies light. We summarize recent work which uses light to both visualize the cellular environment and also control intracel- lular processes along the axis of signal transduction. We highlight recent accomplishments in optical microscopy and optogenetics, an emerging experimental strategy which utilizes light to control the molecular processes in live cells. We believe that optogenetics lends un- precedented spatiotemporal precision to the manipulation of physicochemical properties in biological contexts. We hope to use this work to demonstrate new opportunities for chemical physicists who are interested in pursuing biological and biomedical questions.

A series of 2-（arylmethylidene）-2,3-dihydro-1-benzofuran-3-one derivatives（aurones, 1–20） were synthesized and screened for their inhibitory activity against h MAO. Seventeen compounds（1–5, 7–17,19） were found to be selective towards h MAO-B, while two were non-selective（6 and 20） and one（18）selective towards h MAO-A. Compound 17（Ki = 0.10 0.01 mmol/L） was found to be equally potent and selective towards h MAO-B, when compared with the standard drug Selegiline（Ki = 0.12 0.01 mmol/L）.Nature and position of substitution in aryl ring at 2nd position of benzofuranone influences h MAO-B inhibitory potency, while their structural bulkiness influences selectivity between h MAO-A and h *** docking simulation was also carried out to understand the interaction of inhibitor with the enzyme at molecular level, and we found the docking results were in good agreement with the experimental values. Comparison of the activity profile of the aurones with their corresponding flavones reported earlier by our group revealed that there exists no difference in potency as well as selectivity.

Empirical assessment of adaptation to climate change impacts of mountain households:development and application of an Adaptation Capability Index

The present study proposes an index to assess the potential for adaptation to climate change for households in the mountainous regions. The index provides a realistic approach to recognize social and natural factors which contribute to successful adaptation and addresses several household functions, such as social networking, livelihood strategy, adjustment strategies, resource availability and accessibility. The proposed Adaptation Capability Index （ACI） is analytically defined, mathematically formulated and field tested on mountainous households in urban and semi-urban regions of the Uttarakhand Himalaya in India. To gather data on the topic relevant to the ACI, a household scale questionnaire was developed and administered to 12o heads of households through face-to-face interviews. The results highlight higher adaptive capability of urban households and low adaptation capacity of rural households due to poor farm productivity, low accessibility and availability of resources and technological input. Future programs and policies must include and implement regulations to remedy attributive factors responsible for higher adaptation. This paper may be applicable to other mountainous regions and may provide insights for effective adaptation strategies to climate change.
