Culture and Molecular Identification of <i>Helicobacter pylori</i>in Drinking Water from Areas of High and Low Incidence of Gastric Cancer in Costa Rica

Background: Most patients infected with Helicobacter pylori show no clinical symptoms. Nonetheless, approximately 10% to 20% of these patients will develop peptic ulcers and 1% will develop gastric cancer. The International Agency for Cancer Research has classified Helicobacter pylori as a Group 1 carcinogen. Materials and Methods: Drinking water samples from 20 cantons situated in areas of low and high incidence of gastric cancer in Costa Rica were analyzed. Water samples were concentrated and the nitrocellulose filter was cultivated for later molecular identification of Helicobacter pylori using the glmM marker. The tumor necrosis factor-alpha inducing protein (Tipalpha TNF-α) was used as a pathogenicity marker of the strains found followed by the sequencing of two products. Information on water management by aqueduct operators was collected in order to establish statistical relationships. Results: A total of 112 water samples were analyzed. Successful cultivation and molecular identification using the glmM marker was achieved in 39% of samples from areas of high incidence and in 7.5% of samples from areas of low incidence. Two glmM gene PCR products were sequenced and confirmed as true positives. Conclusions: The highest prevalence rate of Helicobacter pylori was found in water from areas with a high incidence of gastric cancer. Additionally, significant statistical correlations were established among environmental conditions, drinking water management and incidence of gastric cancer.

The market for amino acids: understanding supply and demand of substrate for more efficient milk protein synthesis

For dairy production systems, nitrogen is an expensive nutrient and potentially harmful waste product. With three quarters of fed nitrogen ending up in the manure, significant research efforts have focused on understanding and mitigating lactating dairy cows' nitrogen losses. Recent changes proposed to the Nutrient Requirement System for Dairy Cattle in the US include variable efficiencies of absorbed essential AA for milk protein production. This first separation from a purely substrate-based system, standing on the old limiting AA theory, recognizes the ability of the cow to alter the metabolism of AA. In this review we summarize a compelling amount of evidence suggesting that AA requirements for milk protein synthesis are based on a demand-driven system. Milk protein synthesis is governed at mammary level by a set of transduction pathways, including the mechanistic target of rapamycin complex 1(mTORC1), the integrated stress response(ISR), and the unfolded protein response(UPR). In tight coordination, these pathways not only control the rate of milk protein synthesis, setting the demand for AA, but also manipulate cellular AA transport and even blood flow to the mammary glands, securing the supply of those needed nutrients. These transduction pathways, specifically mTORC1, sense specific AA, as well as other physiological signals, including insulin, the canonical indicator of energy status. Insulin plays a key role on mTORC1 signaling, controlling its activation, once AA have determined mTORC1 localization to the lysosomal *** on this molecular model, AA and insulin signals need to be tightly coordinated to maximize milk protein synthesis rate. The evidence in lactating dairy cows supports this model, in which insulin and glucogenic energy potentiate the effect of AA on milk protein synthesis. In addition to their value as cereal grains, wheat (Triticum aestivum L.) and triticale (× Triticosecale Wittmack) are important cool-season annual forages and cover crops. Yearling steer (Bos taurus) performance was compared in the spring following autumn establishment as for age cover crops after soybean [Glycine max (L.) Merr.] grain harvest. Replicated pastures (0.4 ha) were no-till seeded in three consecutive years into soybean stubble in autumn, fertilized, and grazed the following spring near Ithaca, NE, USA. Each pasture (n = 3) was continuously stocked in spring with four yearling steers (380 ± 38 kg) for 17, 32, and 28 d in 2005, 2006, and 2007, respectively. In 2005, average daily gain (ADG) for steers grazing triticale exceeded the ADG for wheat by 0.31 kghd-1d-1. In 2006, wheat ADG exceeded that for triticale by 0.12 kghd-1d-1. In 2007, steers grazing wheat lost weight, while steers grazing triticale gained 0.20 kghd-1d-1. Based on the 3-year average animal gains valued at $1.32 kg-1, mean net return ($ ha-1 yr-1) was $62.15 for triticale and $22.55 for wheat. Since these grazed cover crops provide ecosystem services in addition to forage, grazing could be viewed as a mechanism for recovering costs and adds additional value to the system. Based on this 3-year grazing trial, triticale was superior to wheat and likely will provide the most stable beef yearling performance across years with variable weather for the western Cornbelt USA.

Dear Editor, The maize Mutator （MuDRIMu） transposon family is the most active DNA transposon in plants （Lisch, 2002）. The Mu family contains diverse elements, all sharing similar -215-bp terminal inverted repeats （TIRs）. MuDR is the autonomous element, and a transcriptionally active MuDR is required for transposition of the non-autonomous Mu elements （Chomet et al., 1991）. MuDR contains two genes, mudrA and mudrB （Figure 1A）; mudrA encodes the MURA transposase, and mudrB encodes a protein with unknown function. To avoid the deleterious effects of transposons, plants have acquired mechanisms to epigenetically silence them. Silenced Mu ele- ments become heavily methylated, and their reactivation is very rare. To date, only UV-B radiation treatments have reac- tivated silenced Mu （Walbot, 1999）.

Lead-free piezoceramics based on the(Ba,Ca)(Zr,Ti)O3(BCZT) system exhibit excellent electromechanical properties for low-temperature actuation applications, but suffer from relatively high processing temperatures. Here we demonstrate an approach for the reduction of the sintering temperature and simultaneous increase of the electromechanical strain response of(Ba,Ca)(Zr,Ti)O3 piezoceramics by aliovalent doping with Ce. The samples were prepared by solid state synthesis and their crystallographic structure, dielectric, ferroelectric, and electromechanical properties were investigated. The highest d33* value of 1189 pm/V was obtained for the sample with 0.05 mol% Ce, substituted on the A-site of the perovskite lattice. The results indicate a large potential of these materials for off-resonance piezoelectric actuators.

Simulation and animation model to boost mining efficiency and enviro-friendly in multi-pit operations

A discrete-event system simulation and animation program was developed to enhance the efficiency of a truck-excavator operation and reduce the environmental impact of haulage in an open-cut coal mine with multiple-pit operations. In any mine, a key objective is to have sufficient equipment for production and not to have excess to where it becomes counterproductive. Due to the advent of responsible mining,environmental regulations, and eco-friendly practices, these factors must also be considered in the analysis. Simulation studies can be financially advantageous for both the optimization of existing mine operations and new development phases in a mine. This study is a new approach to use discrete-event system simulation for mine systems, in order to investigate and possibly reduce environmental impact considering mining haulage performance and production target. A hypothetical layout of a surface coal mine with two pit operations was used for the simulation and animation model. The simulation model includes the animation of the operation. Animation is helpful to enhance the benefit of a mine simulation model. GPSS/Hòand Proof Professionalòwere the software used for the investigation.
