Virtual colonoscopy-induced perforation in a patient with Crohn's disease

We report a case of sigmoid colon perforation in a patient with Crohn's disease undergoing computed-tomographic (CT) colonography. A 70-year-old patient with Crohn's disease with terminal ileitis and sigmoid stricture underwent CT colonography after incomplete conventional colonoscopy. During the procedure, the colon wa. inflated by air insufflation and the patient developed abdominal pain with radiological evidence of retroperitoneal and intraperitoneal free gas. Hartmann's operation wa. performed. This case highlights that CT colonography is not risk-free. The risk of perforation may be higher in patients with inflammatory bowel disease.

Prostate magnetic resonance imaging and the value of experience:An intrareader variability study

Atrophy of the left dorsolateral prefrontal cortex is associated with poor performance in verbal fluency in elderly poststroke women

This study aimed to investigate the association between atrophy in the prefrontal cortex with executive function and verbal fluency in elderly male and female patients poststroke. Thirty elderly female patients with non-aphasic ischemic stroke aged -〉 60 years and 30 age-matched non-aphasic male patients with ischemic stroke were recruited. Automatic magnetic resonance imaging segmentation wa. used to assess the volume of the whole prefrontal cortex, along with its subdivisions： anterior cingulate cortex, orbitofrontal cortex and dorsolateral prefrontal cortex. The Semantic Verbal Fluency Test wa. administered at 3 and 15 months poststroke. At 3 months poststroke, left dorsolateral prefrontal cortex volume wa. significantly correlated with Verbal Fluency Test score in female patients only （partial coefficient = 0.453, P = 0.045）, after controlling for age, education, diabetes, neurological deficit, white matter lesions volume, as well as the location and volume of infarcts. At 15 months poststroke, there remained a significant association between the left dorsolateral prefrontal cortex volume and Verbal Fluency Test （partial coefficient = 0.661, P = 0.001） and between the left prefrontal cortex volume and Verbal Fluency Test （partial coefficient = 0.573, P = 0.004） in female patients after the same adjustments. These findings indicate that atrophy of the left dorsolateral prefrontal cortex contributes to the impairment of verbal fluency in elderly female patients with stroke. Sex differences may be present in the neuropsychological mechanisms of verbal fluency impairment in patients with stroke.

Glaciers are highly sensitive to climate change and are undergoing significant changes in *** this study,we analyzed the spatiotemporal changes of typical glaciers and their responses to climate change in the period of 1990-2015 in 4 different mountainous sub-regions in Xinjiang Uygur Autonomous Region of Northwest China:the Bogda Peak and Karlik Mountain sub-regions in the Tianshan Mountains;the Yinsugaiti Glacier sub-region in the Karakorum Mountains;and the Youyi Peak sub-region in the Altay *** standardized snow cover index(NDSI)and correlation analysis were used to reveal the glacier area changes in the 4 sub-regions from 1990 to *** areas in the Bogda Peak,Karlik Mountain,Yinsugaiti Glacier,and Youyi Peak sub-regions in the period of 1990-2015 decreased by 57.7,369.1,369.1,and 170.4 km^(2),*** of glacier area center of gravity showed that quadrant changes of glacier areas in the 4 sub-regions moved towa.ds the *** area on the south aspect of the Karlik Mountain sub-region wa. larger than that on the north aspect,while glacier areas on the north aspect of the other 3 sub-regions were larger than those on the south *** precipitation in the Karlik Mountain sub-region inhibited the retreat of glaciers to a certain ***,glacier area changes in the Bogda Peak and Youyi Peak sub-regions were not sensitive to the increased *** a seasonal time scale,glacier area changes in the Bogda Peak,Karlik Mountain,Yinsugaiti Glacier,and Youyi Peak sub-regions were mainly caused by accumulated temperature in the wet season;on an annual time scale,the correlation coefficient between glacier area and annual average temperature wa.-0.72 and passed the significance test at Pwa.er resources management in the arid and semi-arid regions of Northwest China in the context of global wa.min

In an effort to understand the molecular events contributing to the cytotoxicity activity of resveratrol(RSV), we investigated its effects on human lung adenocarcinoma epithelial cell line A549 at different concentrations. Cellular nucleoside metabolic profiling wa. determined by an established liquid chromatography-mass spectrometry method in A549 cells. RSV resulted in significant decreases and imbalances of deoxyribonucleoside triphosphates(d NTPs) pools suppressing subsequent DNA synthesis. Meanwhile, RSV at high concentration caused significant cell cycle arrest at S phase, in which cells required the highest dNTPs supply than other phases for DNA replication. The inhibition of DNA synthesis thus blocked subsequent progression through S phase in A549 cells,which may partly contribute to the cytotoxicity effect of RSV. However, hydroxyurea(HU), an inhibitor of RNR activity, caused similar dNTPs perturbation but no S phase arrest, finally no cytotoxicity effect. Therefore, we believed that the dual effect of high concentration RSV, including S phase arrest and DNA synthesis inhibition, wa. required for its cytotoxicity effect on A549 cells. In summary,our results provided important clues to the molecular basis for the anticancer effect of RSV on epithelial cells.

We report in this study the identification of a natural product-like antagonist(1a) of Vps34 as a potent autophagy modulator via structure-based virtual screening. Aurone derivative 1a strongly inhibited Vps34 activity in cell-free and cell-based assays. Significantly, 1a prevents autophagy in human cells induced either by starvation or by an mT OR inhibitor. In silico modeling and kinetic data revealed that 1a could function as an ATP-competitive inhibitor of Vps34. Moreover, it suppressed autophagy in vivo and without inducing heart or liver damage in mice. 1a could be utilized as a new motif for more selective and efficacious antagonists of Vps34 for the potential treatment of autophagy-related human diseases.

Polo-like kinase(PLK1) has been identified as a potential target for cancer *** a number of small molecules have been investigated as PLK1 inhibitors, many of which showed limited selectivity. PLK1 harbors a regulatory domain, the Polo box domain(PBD), which has a key regulatory function for kinase activity and substrate recognition. We report on 3-bromomethylbenzofuran-2-carboxylic acid ethyl ester(designated: MCC1019) as selective PLK1 inhibitor targeting PLK1 PBD. Cytotoxicity and fluorescence polarization-based screening were applied to a library of 1162 drug-like compounds to identify potential inhibitors of PLK1 PBD. The activity of compound MC1019 against the PLK1 PBD wa. confirmed using fluorescence polarization and microscale *** compound exerted specificity towa.ds PLK1 over PLK2 and PLK3. MCC1019 showed cytotoxic activity in a panel of different cancer cell lines. Mechanistic investigations in A549 lung adenocarcinoma cells revealed that MCC1019 induced cell growth inhibition through inactivation of AKT signaling pathwa., it also induced prolonged mitotic arrest—a phenomenon known as mitotic catastrophe, which is followed by immediate cell death via apoptosis and necroptosis. MCC1019 significantly inhibited tumor growth in vivo in a murine lung cancer model without affecting body weight or vital organ size, and reduced the growth of metastatic lesions in the lung. We propose MCC1019 as promising anti-cancer drug candidate.

Background:With the rising global prevalence of fatty liver disease related to metabolic dysfunction,the association of this common liver condition with chronic kidney disease(CKD)has become increasingly *** 2020,the more inclusive term metabolic dysfunction-associated fatty liver disease(MAFLD)wa. proposed to replace the term non-alcoholic fatty liver disease(NAFLD).The observed association between MAFLD and CKD and our understanding that CKD can be a consequence of underlying metabolic dysfunction support the notion that individuals with MAFLD are at higher risk of having and developing CKD compared with those without ***,to date,there is no appropriate guidance on CKD in individuals with ***,there has been little attention paid to the link between MAFLD and CKD in the Nephrology *** and Results:Using a Delphi-based approach,a multidisciplinary panel of 50 international experts from 26 countries reached a consensus on some of the open research questions regarding the link between MAFLD and ***:This Delphi-based consensus statement provided guidance on the epidemiology,mechanisms,management and treatment of MAFLD and CKD,as well as the relationship between the severity of MAFLD and risk of CKD,which establish a framework for the early prevention and management of these two common and interconnected diseases.
