Isolation and Characterization of SARS-CoV-2 in Kenya

The discovery of Severe Acute Respiratory Syndrome-Coronavirus-2 (SARS-CoV-2) in Wuhan, Hubei province, China, in December 2019 raised global health warnings. Quickly, in 2020, the virus crossed borders and infected individuals across the world, evolving into the COVID-19 pandemic. Notably, early signs of the virus’s existence were observed in various countries before the initial outbreak in Wuhan. As of 12th of April, the respiratory disease had infected over 762 million people worldwide, with over 6.8 million deaths recorded. This has led scientists to focus their efforts on understanding the virus to develop effective means to diagnose, treat, prevent, and control this pandemic. One of the areas of focus is the isolation of this virus, which plays a crucial role in understanding the viral dynamics in the laboratory. In this study, we report the isolation and detection of locally circulating SARS-CoV-2 in Kenya. The isolates were cultured on Vero Cercopithecus cell line (CCL-81) cells, RNA extraction was conducted from the supernatants, and reverse transcriptase-polymerase chain reaction (RT-PCR). Genome sequencing was done to profile the strains phylogenetically and identify novel and previously reported mutations. Vero CCL-81 cells were able to support the growth of SARS-CoV-2 in vitro, and mutations were detected from the two isolates sequenced (001 and 002). Genome sequencing revealed the circulation of two isolates that share a close relationship with the Benin isolate with the D614G common mutation identified along the S protein. These virus isolates will be expanded and made available to the Kenya Ministry of Health and other research institutions to advance SARS-CoV-2 research in Kenya and the region.

目的探究铜绿假单胞菌(PA)感染对肺血管内皮细胞功能的影响,并探讨其加重小鼠肺部炎症反应的可能机制。方法PAO1菌株感染人肺微血管内皮细胞HULEC-5a 2 h后,采用逆转录实时荧光定量PCR(RT-qPCR)检测自噬相关基因5(ATG5)、6-磷酸果糖-2-激酶/果糖-2,6-二磷酸酶3(PFKFB3)和钙黏蛋白5(CDH5)的mRNA表达,免疫荧光检测ATG5、PFKFB3和血管内皮钙黏蛋白(VE-cadherin)的蛋白表达。采用小干扰RNA(siRNA)分别敲减ATG5、PFKFB3后,RT-qPCR检测ATG5、PFKFB3和CDH5的mRNA表达,免疫荧光检测PFKFB3和VE-cadherin的蛋白表达,并采用乳酸测定试剂盒检测细胞内乳酸的水平。采用PAO1菌株感染小鼠后,肺组织切片病理染色观察小鼠肺部炎症,激光共聚焦显微镜观察并分析荧光标记的内皮细胞PFKFB3和VE-cadherin的表达量。结果与对照组比较,PAO1感染HULEC-5a细胞后,PFKFB3 mRNA和蛋白表达均增加(P均<0.05),CDH5 mRNA表达减少(P=0.023),VE-cadherin蛋白表达减少(P<0.001),且连续性被破坏,乳酸含量升高(P=0.017)。与PAO1感染HULEC-5a细胞比较,PAO1感染敲减PFKFB3的HULEC-5a细胞后,CDH5 mRNA表达增加(P=0.043),VE-cadherin荧光连续性得到恢复,乳酸含量降低(P=0.047);PAO1感染敲减ATG5的HULEC-5a细胞后,PFKFB3 mRNA(P=0.013)和蛋白表达(P=0.003)均增加,CDH5 mRNA(P=0.020)和VE-cadherin蛋白表达(P=0.001)均减少,乳酸含量升高(P=0.015)。PAO1感染小鼠后,病理HE染色结果显示,肺泡内明显的红细胞渗漏,炎症细胞浸润,肺泡间隔增宽和部分血管内皮细胞脱落。免疫荧光染色结果显示,与正常小鼠比较,PAO1感染小鼠内皮细胞PFKFB3表达增加,VE-cadherin荧光减弱,且不连续。结论PA可能通过AGT5调控PFKFB3通路,破坏肺血管内皮细胞功能,进而加重小鼠肺部炎症反应。

Provably secure authentication protocol for traffic exchanges in unmanned aerial vehicles

Unmanned aerial vehicles offer services such as military reconnaissance in potentially adversarial controlled *** addition,they have been deployed in civilian critical infrastructure *** this environment,real-time and massive data is exchanged between the aerial vehicles and the ground control *** on the mission of these aerial vehicles,some of the collected and transmitted data is sensitive and ***,many security protocols have been presented to offer privacy and security ***,majority of these schemes fail to consider attack vectors such as side-channeling,de-synchronization and known secret session temporary information *** last attack can be launched upon adversarial physical capture of these *** addition,some of these protocols deploy computationally intensive asymmetric cryptographic primitives that result in high *** this paper,an authentication protocol based on lightweight quadratic residues and hash functions is *** formal security analysis is executed using the widely deployed random oracle *** addition,informal security analysis is carried out to show its robustness under the Dolev–Yao(DY)and Canetti–Krawczyk(CK)threat *** terms of operational efficiency,it is shown to have relatively lower execution time,communication costs,and incurs the least storage costs among other related ***,the proposed protocol provides a 25%improvement in supported security and privacy features and a 6.52%reduction in storage *** overall,the proposed methodology offers strong security and privacy protection at lower execution time,storage and communication overheads.

A modeling framework for the integration of electrical and thermal energy systems in greenhouses

Greenhouse horticulture is associated to a significant energy consumption in temperate countries,mainly for lighting and for ***,the potential for energy optimization and energy savings is high but requires detailed models capable of considering various system configurations and control *** paper provides an open-source modeling framework capable of simulating and optimizing the design and the control of both the greenhouse and the generation systems covering all energy *** proposed model is composed of sub-models from different scientific fields:a greenhouse climate model,a crop yield model,a large number of energy generation and storage units models and different rule-based control *** association of such state-of-the-art models in a single framework provides a powerful tool for optimization purposes and allows the definition of completely customized systems by means of an object-oriented *** this work,various control strategies are defined and simulated,thus demonstrating the capabilities of the proposed *** indicate that,by performing minor changes to the control of the thermal screen,heating consumption can be reduced by 3%without any loss in crop *** control of heat-generation units also has a significant impact on the operational costs,which vary by up to 17%when self-consumption levels are accounted for in the control strategy.

Myeloproliferative neoplasms(MPNs)occur due to the abnormal proliferation of one or more terminal myeloid cell lines in peripheral *** suffering from MPNs display a high burden of cardiovascular risk factors,and thrombotic events are often the cause of death in this population of ***,we provide a brief overview of dyslipidemia and metabolic syndrome and their epidemiology in MPNs and examine the common molecular mechanisms between dyslipidemia,metabolic syndrome,and MPNs,with a special focus on cardio-vascular risk,atherosclerosis,and thrombotic ***,we investigate the impact of dyslipidemia and metabolic syndrome on the occurrence and survival of thrombosis in MPN patients,as well as the management of dyslipidemia in MPNs,and the impact of MPN treatment on serum lipid concentrations,particularly as side/adverse effects reported in the context of clinical trials.

Gradient descent decomposition of force-field motor primitives optogenetically elicited for motor mapping of the murine lumbosacral spinal cord

Generating diverse motor behaviors critical for survival is a challenge that confronts the central nervous system(CNS)of all *** movement execution,the CNS performs complex calculations to control a large number of neuromusculoskeletal *** theory of modular motor control proposes that spinal interneurons are organized in discrete modules that can be linearly combined to generate a variety of behavioral *** modules have been previously represented as stimulus-evoked force fields(FFs)comprising isometric limb-endpoint forces across workspace ***,we ask whether FFs elicited by different stimulations indeed represent the most elementary units of motor control or are themselves the combination of a limited number of even more fundamental motor *** probe for potentially more elementary modules,we optogenetically stimulated the lumbosacral spinal cord of intact and spinalized Thy1-ChR2 transgenic mice(n=21),eliciting FFs from as many single stimulation loci as possible(20-70 loci per mouse)at minimally necessary *** found that the resulting varieties of FFs defied simple categorization with just a few *** used gradient descent to further decompose the FFs into their underlying basic force fields(BFFs),whose linear combination explained FF *** mice,we identified 4-5 BFFs with partially localizable but overlapping representations along the spinal *** BFFs were structured and topographically distributed in such a way that a rostral-to-caudal traveling wave of activity across the lumbosacral spinal cord may generate a swing-to-stance gait *** BFFs may represent more rudimentary submodules that can be flexibly merged to produce a library of motor modules for building different motor behaviors.
