Downregulation of Hes1 expression in experimental biliary atresia and its effects on bile duct structure

AIM To analyze the expression and function of the notchsignaling target gene Hes1 in a rhesus rotavirusinduced mouse biliary atresia model. METHODS The morphologies of biliary epithelial cells in biliary atresia patients and in a mouse model were examined by immunohistochemical staining. Then, the differential expression of Notch signaling pathway-related molecules was investigated. Further, the effects of the si RNAmediated inhibition of Hes1 expression were examined using a biliary epithelial cell 3 D culture *** Both immature(Ep CAM+) and mature(CK19+) biliary epithelial cells were detected in the livers of biliary atresia patients without a ductile structure and in the mouse model with a distorted bile duct structure. The hepatic expression of transcripts for most Notch signaling molecules were significantly reduced on day 7 but recovered to normal levels by day 14, except for the target molecule Hes1, which still exhibited lower m RNA and protein levels. Expression of the Hes1 transcriptional co-regulator, RBP-Jκ was also reduced. A 3 D gel culture system promoted the maturation of immature biliary epithelial cells, with increased expression of CK19+ cells and the formation of a duct-like structure. The administration of Hes1 si RNA blocked this process. As a result, the cells remained in an immature state, and no duct-like structure was *** Our data indicated that Hes1 might contribute to the maturation and the cellular structure organization of biliary epithelial cells, which provides new insight into understanding the pathology of biliary atresia.

Modeling the Interaction between a Thermal Flow and a Liquid: Review and Future Eulerian-Lagrangian Approaches

Suspension Plasma Spraying is a complex process in which several physical mechanisms play a part. So the modeling and understanding of the interaction between a high-velocity and thermal flow and a liquid precursor phase is of major importance concerning the control and characterization of the process. The liquid droplet size distribution has a high influence on the kinetic properties of the as-sprayed nanometer particles before impacting on a target substrate. An overview of existing models is provided dealing with the penetration of the liquid phase into the thermal flame and the resulting fragmentation and vaporization of this phase before impact. The physical characteristics of the flow as well as existing Lagrangian and Eulerian modeling strategies are briefly discussed while paying attention to the physical parameters characterized and measured by numerical simulation. The potential of the various models and also their limits are intended to be highlighted. Future coupled Eulerian-Lagrangian modeling strategies are also proposed for a global and more exhaustive representation of the injection, fragmentation and dispersion part of the two-phase gas-liquid flow before particle impact on the substrate.

Role of CD56-expressing immature biliary epithelial cells in biliary atresia

AIM: To analyze the clinical and pathological parameters and expression of the neural cell adhesion molecule(CD56) in patients with biliary atresia(BA).METHODS: Established clinical laboratory markers of hepatic function, including enzyme activity, protein synthesis, and bilirubin metabolism, were evaluated in patients with BA and compared with those in patients with choledochal cysts and neonatal hepatitis. Pathological changes in tissue morphology and fibrosis were examined by histological and tissue collagen staining. Immunohistochemical staining for the biliary epithelial cell markers CD56 and CK19 together with the Notch signaling related molecules Notch1 and Notch2 was performed in the context of alterations in the structure of intrahepatic biliary ***: Differences in some clinical laboratoryparameters among the three diseases examined were observed, but they did not correlate with the pathological classification of fibrosis in BA. Immunohistochemical staining showed the presence of CD56-positive immature bile ducts in most patients(74.5%) with BA but not in patients with choledochal cysts or neonatal hepatitis. The number of CD56-expressing cells correlated with disease severity, with more positive cells present in the later stages of liver damage(81.8% vs 18.2%). Furthermore, bile plugs were mainly found in CD56-positive immature biliary ducts. Notch signaling was a key regulatory pathway in biliary duct formation and played a role in tissue fibrosis. Notch1 was co-expressed in CD56-positive cells, whereas Notch2 was found exclusively in blood vessels in the portal area of patients with BA. CONCLUSION: The maturation of biliary epithelial cells and the expression of Notch may play a role in the pathogenesis of BA.

In this paper, we present hands-on experience related to on-going implementation in aircraft of power supply for a wireless sensor network deployed for aerodynamic flight tests. This autonomous battery-free power supply is capturing, managing and storing primary energy from the environment, using solar light and PV （photovoltaic） cells. For practical purposes, it is also equipped with an auxiliary power input. The specifications are detailed, the general architecture is presented and justified, and test results are discussed.

The present work aims to study the influence of some physicochemical parameters (light, temperature, ethanol, bile salts, potassium hydroxide, hydrogen peroxide) on the content of Metronidazole (MTZ) contained in the reference substance (SR) and in a pharmaceutical specialty Flagyl® 250 mg tablet (FLG). The method developed was linear and accurate in accordance with USP 38 and the MTZ contents were obtained by UV-visible spectrophotometry at 278 nm. These contents ranged from 225 mg to 275 mg and were thus consistent with the concentration present in the proprietary medicines (250 mg). The study of the influence of physicochemical parameters on the MTZ content in SR and FLG showed that MTZ contents are unstable in the presence of Ethanol at 96˚, KOH at 0.1N and bile salts but also when the temperature is higher than 25˚C. However, they remain stable in the presence of light and H2O2 and undergo degradation in an acidic environment.

Influence of Some Physico-Chemical Exposure Factors on the Carbocysteine Content of an Opened Pharmaceutical Product

The aim of this study was to investigate the stability of Carbocysteine (CBC) contained in a reference substance (RS) and in a sample of opened RHINATHIOL® 5% syrup (RNTL 5%) under the effect of some physicochemical parameters (temperature, light, time, pH, bile salts). The developed method was linear, precise and accurate according to USP 38. The co-efficient of determination R2 for linearity was 0.9993. The respective RSD of intra-day and inter-day between days (1st, 2nd and 3rd day) were respectively 0.338% and the interval from 0.05% to 0.387%. The average recovery rate ranged from 98.490% to 100.450%. The detection and quantification limits were 0.0001 mg/mL and 0.001 mg/mL respectively. The method was applied to four samples of opened syrup containing CBC and the CBC content in these samples was found to be in accordance with USP 38. The CBC content in the opened sample of RNTL 5% was obtained by UV-visible spectrophotometry at 217 nm and was 4.887 g/100mL. The study of the influence of physico-chemical factors on the content of CBC in RS and RNTL 5% showed that the evolution of CBC contents in each drug matrix remained dependent on pH and temperature. However, these levels remained stable in the presence of light.

Effect of Drought Stress on Yield Performance of Parental Chickpea Genotypes in Semi-arid Tropics

Chickpea （Cicer arietinum L.） is an important cool season food legumes with indeterminate growth habit. The crop is valued for its nutritive seeds and used as animal feed in many developing countries. The productivity of the crop is constrained by several abiotic stresses, among which drought stress is one of the key determinants of crop performance aaccounting for 40-50% yield reduction globally. The present study was conducted to screen, evaluate and select chickpea genotypes possessing high yield potential under drought stress condition at ASALs （arid and semi-arid lands） of Kenya. The experiment was conducted at Chemeron dry land and Eco-tourism Research station, Egerton University and Kenya Agricultural and Livestock Research Institute （KALRO）, Pekerra, Marigat, Baringo County. The genotypes were planted in RCBD （randomized complete block design） in three replicates at a spacing of 30 cm × 10 cm, giving a plant density of approximately 25 plants/m2. Combined analysis of variance revealed existence of highly significant differences among the tested genotypes for most of the agronomic traits. Overall, the highest grain yield was obtained from ICCV 92944 （1,173 kg/ha）, ICCV 92318 （1,103 kg/ha） and CAVIR （975 kg/ha）, ICCV 92318 （967 kg/ha）, ICCV 00108 （956 kg/ha） and ICC 4958 （921 kg/ha）： possibly due to its comparatively higher drought （and heat） tolerance, and hence could be used as sources of drought tolerance in further breeding programs. This study was carried out in few drought tolerant sites and further more sites need to be evaluated in addition to other drought and heat screening and optimization of protocols, facilities and analytical approaches to identify better genotypes that respond appropriately to climate change.

Factors Associated with Rising Trend in Teenage Pregnancy within the West Gonja Municipality of the Savannah Region of Ghana

Background: Teenage pregnancy is a major public health problem worldwide, also in northern Ghana. The aim of this study was to identify factors contributing to the rising trend in teenage pregnancy and the resultant effects. Materials and Methods: This was a cross-sectional study using a quasi-structured questionnaire on 60 pregnant teenagers. Demographic data and factors associated with teenage pregnancy were collected/analyzed. Results: The mean age of respondents was 17.25 ± 1.93 years. Junior high school students or graduates accounted for 55%. About 35% of participants dropped out of school due to pregnancy. The following factors were identified to be associated with teenage pregnancy: low socio-economic status, no/less sexual- and reproductive-communication with parents, low level of education, no sexual and reproductive health knowledge, and unsafe sexual behaviours. The consequences of teenage pregnancy identified were: truncation of the girls’ education, psychological trauma, and worsening family relationships. The following health consequences were identified;anaemia in pregnancy, severe malaria, hypertension in pregnancy, and prolonged labour. Conclusion: The following factors may account for the rising trend in teenage pregnancy in this area: family-related problem, sociocultural factors, low education level, and the lack of adequate knowledge on reproductive health. Teenage pregnancy caused several problems in themselves and their family members.

Controlling the gelation temperature of biomimetic polyisocyanides

Thermosensitive polymers show an entropy-driven transition from a well-solvated to a poorly solvated polymer chain, resulting in a more compact globular conformation. The transition at the lower critical solution temperature（LCST） is often sharp, which allows for a wide range of smart material *** the LCST, oligo（ethylene glycol）-substituted polyisocyanides（PICs） form soft hydrogels, composed of polymer bundles similar to biological gels, such as actin, fibrin and intermediate filaments. Here, we show that the LCST of PICs strongly depends linearly on the length of the ethylene glycol（EG） tails; every EG group increases the LCSTand thus the gelation temperature by nearly 30 ℃. Using a copolymerisation approach, we demonstrate that we can precisely tailor the gelation temperature between 10 ℃ and 60 ℃and, consequently, tune the mechanical properties of the PIC gels.

Small fishes crossed a large mountain range:Quaternary stream capture events and freshwater fishes on both sides of the Taebaek Mountains

The Taebaek Mountains in Korea serve as the most apparent biogeographic barrier for Korean freshwater fishes,resulting in 2 distinct ichthyofaunal assemblages on the eastern(East/Japan Sea slope)and western(Yellow Sea and Korea Strait slopes)sides of the mountain *** nearly 100 species of native primary freshwater fishes in Korea,only 18 species occur naturally on both sides of the mountain ***,there are 5 rheophilic species(Phoxinus phoxinus,Coreoleuciscus splendidus,Ladislavia taczanowskii,Iksookimia koreensis and Koreocobitis rotundicaudata)found on both sides of the Taebaek Mountains that are geographically restricted to the Osip River(and several neighboring rivers,for *** and ***)on the eastern side of the mountain *** Osip River and its neighboring rivers also shared a rheophilic freshwater fish,Liobagrus mediadiposalis,with the Nakdong River on the western side of the mountain *** assessed historical biogeographic hypotheses on the presence of these rheophilic fishes,utilizing DNA sequence data from the mitochondrial cytochrome b *** of our divergence time estimation indicate that ichthyofaunal transfers into the Osip River(and several neighboring rivers in East Sea slope)have occurred from the Han(Yellow Sea slope)and Nakdong(Korea Strait slope)Rivers since the Late *** inferred divergence times for the ichthyofaunal transfer across the Taebaek Mountains were consistent with the timing of hypothesized multiple reactivations of the Osip River Fault(Late Pleistocene),suggesting that the Osip River Fault reactivations may have caused stream capture events,followed by ichthyofaunal transfer,not only between the Osip and Nakdong Rivers,but also between the Osip and Han Rivers.
