Erectile dysfunction （ED） is a major complication of diabetes mellitus （DM）. This study investigates the relationship between ED and the downregulation of constitutive nitric oxide synthase （cNOS） in the corpus cavernosum （CC） of diabetic rats. It also examines the effects of udenafil, a phosphodiesterase type 5 （PDE5） inhibitor, on ED and cNOS expression levels. After 16 weeks of daily oral treatment with udenafil in diabetic rats, the intracavernous pressure/mean arterial pressure （ICP/MAP） ratio was recorded to measure erectile function, and cNOS expression was measured using reverse transcriptase （RT）-PCR and immunoblots. Although the ICP/ MAP ratio and the expression levels of endothelial NOS （eNOS） and neuronal NOS （nNOS） in the CC were markedly decreased in diabetic rats, long-term udenafil treatment improved the erectile function and increased cNOS expression compared with diabetic controls. These findings suggest that ED in DM is closely related to decreased cNOS expression in the CC and that udenafil has the ability to compensate for this pathological change by modulating cNOS expression. Udenafil also has an inhibitory role in cyclic guanosine monophosphate （cGMP） degradation.

Histone methyltransferase enzyme SET7 and mixed-lineage leukemia protein(MLL)complex are crucial co-activators of androgen receptor(AR)and have recently emerged as potential therapeutic targets for advanced castration-resistant prostate cancer(CRPC).In this study,we described the identification of a rhodium-based hybrid complex(SM_1)as a potent blocker of AR activity via simultaneously inhibiting SET7 and MLL complex activity,which makes it a potential lead scaffold for CPRC drug development.

T Objective： To examine the Eastem-Westem difference in the interpretation of Integrative Medicine Attitude Questionnaire （IMAQ） by assessing the psychometric properties of a revised chinese medicine （CM）- specific version of IMAQ （CM-IMAQ）. Methods： Factor and Rasch analysis were performed with data collected from a mail survey of 165 Hong Kong Western medical doctors （WMD） randomly sampled from the official registry. The structural validity, unidimensionality, item fit, and differential item functioning （DIF） of the Hong Kong CM-IMAQ were evaluated. Results： Confirmatory factor analysis （CFA） demonstrated that the original IMAQ factor structure was not concordant with our data on chinese WMD, and subsequent explanatory factor analysis （EFA） validated a new three-factor model for CM-IMAQ：（1） attitude towards tonification , （2） attitude towards the effectiveness of CM, and （3） attitude towards CM knowledge. The original IMAQ factor on holism and doctor-patient relationship disappeared. Rasch analysis confirmed the unidimensionality of tonification and the effectiveness domains, but further refinement of the knowledge domain is needed. Coaclusions： Cultural adaptation of the IMAQ has demonstrated differences between Eastern and Westem doctors trained in allopathic medicine in their interpretations of hoiism in healthcare. For chinese WMD, the emphasis of holistic care is placed on tonifying＇ the body rather than on nurturing the mind and spidt. Confucian and Taoist conceptualizations of mental health as well as the persistent stigma towards mental illness within modern chinese culture may explain why chinese WMD do not regard mental health promotion as part of routine healthcare.

We report in this study the identification of a natural product-like antagonist(1a) of Vps34 as a potent autophagy modulator via structure-based virtual screening. Aurone derivative 1a strongly inhibited Vps34 activity in cell-free and cell-based assays. Significantly, 1a prevents autophagy in human cells induced either by starvation or by an mT OR inhibitor. In silico modeling and kinetic data revealed that 1a could function as an ATP-competitive inhibitor of Vps34. Moreover, it suppressed autophagy in vivo and without inducing heart or liver damage in mice. 1a could be utilized as a new motif for more selective and efficacious antagonists of Vps34 for the potential treatment of autophagy-related human diseases.

Temozolomide(TMZ)is an anticancer agent used to treat glioblastoma,typically following radiation therapy and/or surgical ***,despite its effectiveness,at least 50%of patients do not respond to TMZ,which is associated with repair and/or tolerance of TMZ-induced DNA *** have demonstrated that alkyladenine DNA glycosylase(AAG),an enzyme that triggers the base excision repair(BER)pathway by excising TMZ-induced N3-methyladenine(3meA)and N7-methylguanine lesions,is overexpressed in glioblastoma tissues compared to normal ***,it is essential to develop a rapid and efficient screening method for AAG inhibitors to overcome TMZ resistance in ***,we report a robust time-resolved photoluminescence platform for identifying AAG inhibitors with improved sensitivity compared to conventional steady-state spectroscopic *** a proof-of-concept,this assay was used to screen 1440 food and drug administration-approved drugs against AAG,resulting in the repurposing of sunitinib as a potential AAG *** restored glioblastoma(GBM)cancer cell sensitivity to TMZ,inhibited GBM cell proliferation and stem cell characteristics,and induced GBM cell cycle ***,this strategy offers a new method for the rapid identification of small-molecule inhibitors of BER enzyme activities that can prevent false negatives due to a fluorescent background.

Dear Editor,Triple-negative breast cancer(TNBC)is an aggressive breast cancer subtype enriched in cancer stem cells(CSCs),which is characterized by high malignancy and drug resistance.1 The PI3K/Akt signaling cascade is frequently hyperactivated in *** and non-redundant functions of the three Akt isoforms in tumorigenesis,however,have not been studied *** is noteworthy that despite a few isoform-specific substrates have been identified for Akt1 and Akt2,an Akt3-specific substrate is yet to be *** have previously shown that overexpression of Akt3,but not Akt1 or Akt2,plays an essential role in TNBC growth and therapeutic resistance of breast cancer.2 However,the role of Akt3 in TNBC CSCs remains unclear.

The pandemic H1N12009 inﬂuenza virus has caused theﬁrst inﬂuenza pandemic of the 21st century,leading to disproportionate fatalities in the low-risk population despite the generally mild nature of the *** in science and technology have allowed very detailed study on the pathogenesis of this novel virus,and many have already been published in less than a year after the start of the *** generated from cell lines,animal models,and clinical data analysis has provided us with greater understanding of the behavior of this virus and the associated host *** new knowledge will allow us to formulate scientiﬁcally sound and evidence-based management plans.
