Systemic gemcitabine combined with intra-arterial low-dose cisplatin and 5-fluorouracil for advanced hepatocellular carcinoma: Seven cases

The combination of intra-arterial low-dose cisplatin and 5-fluorouracil (5-FU) is effective against advanced hepatocellular carcinoma (HCC). Systemic gemcitabine chemotherapy seems effective in many cancers. We report the results of combination therapy with systemic gemcitabine, intra-arterial low-dose cisplatin and 5-FU (GEmFP). Seven patients with non-resectable advanced HCC were treated with GEmFP. One course of chemotherapy consisted of daily intra-arterial cisplatin (20 mg/body weight/hour on d 1, 10 mg/body weight per 0.5 h on d 2-5 and 8-12), followed by 5-FU (250 mg/body weight per 5 h on d 1-5 and 8-12) via an injection port. Gemcitabine at 1000 mg/m2 was administered intravenously at 0.5 h on d 1 and 8. The objective response was 57%. The response to GEmFP was as follows: complete response (no patients), partial response (four patients), stable disease (three patients), and progressive disease (no patients). The median survival period was 8 mo (range, 5-55). With regard to the National Cancer Institute Common Toxicity Criteria (NCI-CTC) grade 3 or 4 adverse reactions, seven (100%), seven, six (86%) and one (14%) patients developed leukopenia, neutropenia, thrombocytopenia and anemia, respectively. GEmFP may potentially be effective for non- resectable advanced HCC, but it has severe hematologic toxicity.

目的探讨m1巨噬细胞对乳腺癌细胞上皮细胞-间充质转换(EmT)的影响及钙蛋白酶(Calpain)在其中的介导作用。方法以人乳腺癌细胞系雌激素受体阳性(ER+)细胞mCF-7、T47D及雌激素受体阴性(ER-)细胞mDA-mB-231为模型细胞,实验分为mCF-7细胞及T47D细胞的RPmI-1640组(RPmI-1640纯培养基培养)、m1巨噬细胞条件培养液培养组(m1-Cm组)、m1-Cm+Calp组[m1-Cm联合Calpain抑制剂Calpeptin(50μmoL/L)处理]、m1-Cm+CI-Ⅲ组[m1-Cm联合Calpain抑制剂Calpain inhibitorⅢ(50μmoL/L)处理],mDA-mB-231细胞的RPmI-1640组、m1-Cm组;采用划痕实验和侵袭实验检测各组中mCF-7细胞、T47D细胞、mDA-mB-231细胞的迁移能力及mCF-7细胞的侵袭能力,采用qRT-PCR检测m0巨噬细胞、m1巨噬细胞趋化因子受体7CCR7、趋化因子配体10(CXCL10)及白细胞介素-12(IL-12) mRNA表达水平,采用Western blot实验检测mCF-7细胞、T47D细胞及mDA-mB-231细胞中上皮-钙黏素(E-cad)、纤连蛋白(FN)、波形蛋白(vim)蛋白水平表达。结果与RPmI-1640组相比,m1-Cm组ER+细胞mCF-7及T47D迁移能力增强(PmCF-7细胞侵袭能力增强(PmCF-7及T47D E-cad蛋白表达水平下调,而FN、vim表达上调(PmDA-mB-231中只有E-cad表达上调(Pm1-Cm组相比,ER+细胞mCF-7及T47D的m1-Cm+Calp组、m1-Cm+CI-Ⅲ组中m1-Cm诱导的ER+乳腺癌细胞EmT效应被逆转(Pm1巨噬细胞条件培养液可以促进ER+细胞人乳腺细胞的EmT,但对ER-细胞EmT无明显影响,其促进ER+乳腺癌细胞EmT信号机制可能与Calpain通路有关。

Knowledge and Utilization of mushroom as a Food Supplement among Families in Selected Local Government Areas of Cross River State, Nigeria

The demand for health consciousness to prevent malnutrition, a high risk factor in developing countries especially in the rural areas is increasing. This study was designed to assess the level of knowledge and utilization of mushroom as a food supplement in six Local Government Areas of Cross River State, one predominantly urban and the other five mostly rural. Data were collected using a structured questionnaire from a total of 250 participants. Findings revealed that 178 (71.2%) of respondents had good knowledge of mushrooms as food supplement 205 (82%) utilized them as such, while majority of the participants 243 (93.6%) 20.1% said it is part of the family diet if available. Chi-square analysis showed statically significant association between knowledge and utilization (X2 cal = 89.62a, X2 tab = 3.84;df = 1, p mushrooms as the most cost-effective source of a healthier diet tackling food insecurity, diversification and malnutrition for families in Cross River.

aim: To evaluate the functional differences between the 2 liver lobes in non-cirrhotic patients by using computed tomography/99mTc-galactosyl human serum albumin (CT/99mTc-GSA) single-photon emission computed tomography (SPECT) fusion images. mETHODS: Between December 2008 and march 2012, 264 non-cirrhotic patients underwent preoperative liver function assessment using CT/99mTc-GSA SPECT fusion images. Of these, 30 patients, in whom the influence of a tumor on the liver parenchyma was estimated to be negligible, were selected. Specifically, the selected patients were required to meet either of the following criteria: (1) the presence of an extrahepatic tumor; or (2) presence of a single small intrahepatic tumor. These 30 patients were retrospectively analyzed to calculate the percentage volume (%Volume) and the percentage function (%Function) of each lobe. The ratio between the %Function and %Volume (function-to-volume ratio) of each lobe was also calculated, and the ratios were compared between the 2 lobes. Furthermore, the correlations between the function-to-volume ratio and each of 2 liver parameters [lobe volume and diameter ratio of the left portal vein to the right portal vein (LPV-to-RPV diameter ratio)] were investigated. RESULTS: The median values of %Volume and %Function were 62.6% and 67.1% in the right lobe, with %Function being significantly higher than %Volume (P median values of %Volume and %Function were 31.0% and 28.7% in the left lobe, with %Function being significantly lower than %Volume (P me ratios of the right lobe (1.04-1.14) were significantly higher than those of the left lobe (0.74-0.99) (P me ratio showed no significant correlation between the lobe volume in either lobe. In contrast, the function-to-volume ratio showed significant correlations with the LPV-to-RPV diameter ratio in both lobes (right lobe: negative correlation, rs = -0.37, P = 0.048; left lobe: positive

Effects of combined drug therapy on HIV-1 infection dynamics

Infection of human immunodeficiency virus （HIV） is determined through the decay of healthy CD44- T-cells in a well-mixed compartment, such as a bloodstream. A mathe- matical model is considered to illustrate the effects of combined drug therapy, i.e. reverse transcription plus protease inhibitor, on viral growth and T-cell population dynamics. This model is used to explain the existence and stability of infected and uninfected steady states in HIV growth. An analytical technique, called variational iteration method （vim）, is used to solve the mathematical model. This method is modified to obtain the rapidly convergent successive approximations of the exact solution. These approximations are obtained without any restrictions or the transformations that may change the physical behavior of the problem. Numerical simulations are computed and exhibited to illustrate the effects of proposed drug therapy on the growth or decay of infection.

目的探讨m1巨噬细胞条件培养液对乳腺癌细胞上皮-间质转换(EmT)的影响、及EmK/Notch1通路在其中的介导作用。方法将mCF-7细胞分为RPmI-1640组(RPmI-1640纯培养基)、m1-Cm组(m1巨噬细胞条件培养液)、m1-Cm+DAPT组(m1-Cm联合50μmol/L DAPT)及m1-Cm+U0126组(m1-Cm联合10μmol/L U0126),采用划痕实验和侵袭实验(Transwell)检测mCF-7细胞迁移和侵袭能力,采用蛋白印迹(Western blot)实验检测mCF-7细胞蛋白水平表达。结果与RPmI-1640组相比,m1-Cm组mCF-7细胞迁移率及侵袭率均增加(Pvim蛋白表达水平上调(Pm1-Cm组相比,m1-Cm+U0126组及m1-Cm+DAPT组mCF-7细胞迁移率及侵袭率均降低、E-cad蛋白表达水平上调(Pvim蛋白表达水平下调(Pm1-Cm组比较,m1-Cm+U0126组Notch信号通路Notch1蛋白、其靶基因Hes1蛋白表达均被显著抑制(Pm1-Cm+DAPT组ERK磷酸化未被抑制(P>0.05)。结论m1巨噬细胞条件培养液促进mCF-7细胞EmT,该效应可能与ERK/Notch1信号通路活动有关。

mICROSTRUCTURE AND PROPERTIES OF CuCr25 ALLOYS WITH DIFFERENT Ni CONTENT

CuCr25 alloys containing different Ni content were prepared by vacuum induction melting (vim). The micro structure and properties were tested. The results show that with the increase of Ni content in CuCr25 alloys, the Cr phase changed from developed dendrite into nodular grains and was drastically refined; the electrical conductivity significantly decrease, but still reach the level of conventional CuCr50 when the Ni content is below 0.5%. The Ni content had little influence on their breakdown strength. The first breakdown sites transferred to the boundary of Cu and Cr phase for CuCr25Ni compared to the Cr phase for CuCr25 without Ni.

A Study of Some Nonlinear Partial Differential Equations by Using Adomian Decomposition method and Variational Iteration method

In this paper, a numerical solution of nonlinear partial differential equation, Benjamin-Bona-mahony (BBm) and Cahn-Hilliard equation is presented by using Adomain Decomposition method (ADm) and Variational Iteration method (vim). The results reveal that the two methods are very effective, simple and very close to the exact solution.
