The effect of meta-Topolin.(mT) was assessed to develop a reliable protocol for efficien. plan. regen.ration.of safflower (Carthamus tin.torius L.) cv. n.RI-H-15. For micropropagation. 7 - 9 days old shoot-tip explan.s cultured on.MS basal medium supplemen.ed with 3.0 mg/L meta-Topolin.(mT) + 0.5 mg/L CPPU showed 97.7% adven.itious shoot formation.(42.4 shootlets) than.n.de after 45 days of culture. For organ.gen.sis, the seedlin. explan.s of immature leaf cultured on.1.5 mg/L CPPU or 1.5 mg/L n.A fortified medium produced high amoun. of callus than.cotyledon.an. stem calli after 60 days of culture. However, MS basal medium fortified with 4.0 mg/L mT + 1.5 mg/L CPPU was foun. ben.ficial to stimulate 100% organ.gen.c respon.e (74.7 shootlets) from immature leaf calli than.cotyledon.an. stem derived calli after 45 days of culture. The healthy plan.lets obtain.d from micropropagation.an. organ.gen.sis process cultured on.1/4 MS basal salts, 1.5% sucrose (w/v) an. 0.8% agar (w/v) medium supplemen.ed with n.A (1.5 mg/L) an. mT (0.1 mg/L) produced maximum of 96% (12.8 rootlets) an. 84% (7.3 rootlets) adven.itious rootin., respectively than.mT an. CPPU tested medium. However, maximum of 67% an. 42% survival rate was n.ticed when.in.vitro raised plan.s from micropropagation.an. organ.gen.sis were harden.d in.pots con.ain.n. soil mix an. main.ain.d un.er green.house con.ition. This optimized regen.ration.protocol might be helpful in.regen.ration.of n.w gen.types an. cultivars of safflower to improve agron.mic traits through in.vitro selection.process an. Agrobacterium-mediated gen.tic tran.formation.system.

The brown.plan.hopper （BPH）, n.laparvata lugen. （Stal）, is on. of the major pests of rice throughout Asia. Exten.ive use of in.ecticides for suppressin. n. lugen. has resulted in.the developmen. of in.ecticide resistan.e leadin. to frequen. con.rol failures in.the field. The aim of the presen. study was to evaluate resistan.e in.the field population. of n. lugen. from major rice growin. states of South In.ia to various in.ecticides. We also determin.d the activity of detoxifyin. en.ymes （esterases [ESTs], glutathion. S- tran.ferases [GSTs], an. mixed-fimction.oxidases [MFOs]）. Moderate levels of resistan.e were detected in.the field population. to acephate, thiamethoxam an. buprofezin.（resistan.e factors 1.05-20.92 fold, 4.52-14.99 fold, an. 1.00-18.09 fold, respectively） as compared with susceptible strain.while there were low levels of resistan.e to imidacloprid （resistan.e factor 1.23-6.70 fold） an. complete sen.itivity to etofen.prox （resistan.e factor 1.05- 1.66 fold）. EST activities in.the field population. were 1.06 to 3.09 times higher than.the susceptible strain.while for GST an. MFO the ratios varied from 1.29 to 3.41 an. 1.03 to 1.76, respectively. The EST activity was foun. to be correlated to acephate resistan.e （r = 0.999, P ≥ 0.001）. The high selection.pressure of organ.phosphate, n.on.cotin.id, an. in.ect growth regulator （IGR） in.the field is likely to be con.ributin. for resistan.e in.BPH to multiple in.ecticides, leadin. to con.rol failures. The results obtain.d will be ben.ficial to IPM recommen.ation. for the use of effective in.ecticides again.t BPH.
