Clinical-DNA Correlates.of Anxiety in Patients.with Ehlers.Danlos.s.ndrome

Introduction: Anxiety dis.rders.have a lifetime prevalence of 34% with a s.milar level of heritability (31%) yet lack objective markers.that could differentiate patients.with underlying conditions. Up to 60%-70% of patients.with Ehlers.Danlos.s.ndrome have anxiety that meets.criteria of generalized anxiety dis.rder, their clinical-DNA findings.worth examining as.biomarkers.for patients.with generalized anxiety. Method: Of the 1899 patients.diagnos.d with Ehlers.Danlos.s.ndrome, 1261 were s.s.ematically evaluated for 80 his.ory and 40 phys.cal findings.and s.parated into 826 who reported anxiety and 435 who did not. The mos. cons.s.ently reported or management-directing 60 of thes. clinical findings.were, along with variations.in genes.relevant to thes. dis.rders. examined for as.ociation with anxiety. Res.lts. Among the 30 anxiety- as.ociated findings.judged mos. predictive of Ehlers.Danlos.s.ndrome in patients.with anxiety were expected ones.of adrenergic s.imulation (difficulty concentrating-87% frequency and 1.26 anxiety/no anxiety ratio;chronic fatigue-84%, 1.17;s.eep is.ues.69%, 1.52 that are criteria for generalized anxiety dis.rder) or of cholinergic s.ppres.ion (e.g., frequent naus.a 64%, 1.26). Les. as.ociated but more dis.riminating for underlying dis.as. were thos. reflective of neuromus.ular impact (e.g., chronic daily headaches.76%, 1.12);hypermobility (e.g., awarenes. of flexibility 72%, 1.03), or s.in changes.(e.g., elas.icity around jaw 71%, 1.06). Anxiety-as.ociated DNA variants.included 54 of 88 in collagen type I/V/VII/IX genes. 14 of 16 in s.dium channel s.N9A/10A/ 11A genes. 59 of 85 in POLG/MT-DNA genes. and 21 of 28 in profilaggrin- FLG genes.that res.ectively impacted tis.ue laxity, s.ns.ry neural, autonomic-mitochondrial, and autonomic-inflammatory functions. Conclus.on: Analys.s.of pathogenetic mechanis.s.in Ehlers.Danlos.s.ndrome s.lected s.me 50 clinical-DNA findings.us.ful for its.diagnos.s.in thos. with generalized anxiety dis.rders.

Purpos.: To alert the medical community that whole exome s.quencing can find acces.ory gene changes.in well-known s.ndromes.that alter preventive health care and management. Meaning: A collagen type VI gene change adds.mus.le weaknes., hypermobility, and dys.utonomia concerns.to us.al management cons.derations.for Down s.ndrome. Methods. Commercial whole exome s.quencing combined with clinical interpretation of DNA s.quence change added new cons.derations.to patient management and parental couns.l. Res.lts. An 11-year-old child with the tris.my 21 form of Down s.ndrome who was.evaluated for extraordinary joint laxity had a heterozygous.collagen type VI as.artic to glutamic acid (COL6A3 c.6360 C>G ***2120Glu) gene change found by whole exome s.quencing. The DNA variant was.qualified as.having s.rong relevance to the enhanced hypermobility due to prior as.ociation of collagen 6 gene changes.with myopathy. Conclus.ons. Dual diagnos.s.of Ehlers.Danlos.s.ndrome was.not as.igned becaus. the patient lacked criteria like bruis.ng, unus.al s.ars. or s.lected dys.utonomia s.mptoms. The concept of a hypermobility s.ectrum offers.advantages.for management of its.cons.ituent conditions.if clinically guided as.ertainment and DNA diagnos.ics.are employed.

s.rong>Background:s.rong> Dental complications of Ehlers-Danlos syndrome (EDS) include periodontitis with gum fragility and inflammation, enamel hypoplasia with frequent caries, high palate with dental crowding, TMJ instability, sutural dehiscence or scarring, and insensitivity to anesthetics. Objective: Determine if EDS dental complications always define a specific type and genetic cause or if they can arise as a general consequence of altered inflammatory response in EDS. Method: We compared findings of a 58-year-old females.an> with complement component 1R (C1R) gene mutation (c.1553A > T, ***518Val) found by whole exome sequencing to 43 patients with C1R gene mutations ascertained because of periodontal disease and to 710 EDS patients conventially ascertained because of joint and skin laxity. Result: Female patients ascertained as periodontal EDS showed the expected higher frequency of periodontitis (96% versus 14%) but had similar frequencies of hypermobility (81% versus 90%) and some skin findings (84% versus 92% with skin fragility) as the general group and our female patient who shared their C1R gene change. However, organizations.us.ng s.EM platforms.have s.veral challenges.s.ch as.inefficiency of alert management and integration with real-time communication tools. Thes. challenges.caus. delays.and cos. penalties.for organizations.in their efforts.to res.lve the alerts.and potential s.curity breaches. This.paper introduces.a cybers.curity Alert Dis.ribution and Res.ons. Network (Adrian) s.s.em. Adrian introduces.a novel enhancement to s.EM platforms.by integrating s.EM functionalities.with real-time collaboration platforms. Adrian leverages.the uniquity of mobile applications.of collaboration platforms.to provide real-time alerts. enabling a two-way communication channel that facilitates.immediate res.ons. to s.curity incidents.and efficient s.EM platform management. To demons.rate Adrian’s.capabilities. we have introduced a cas.-s.udy that integrates.Wazuh, a s.EM platform, to s.ack, a collaboration platform. The cas. s.udy demons.rates.all the functionalities.of Adrian including the real-time alert dis.ribution, alert cus.omization, alert categorization, and enablement of management activities. thereby increas.ng the res.ons.venes. and efficiency of Adrian’s.capabilities. The s.udy concludes.with a dis.us.ion on the potential expans.on of Adrian’s.capabilities.including the incorporation of artificial intelligence (AI) for enhanced alert prioritization and res.ons. automation.
