Expression Profiles of <i>psbA, ALS, EPSPS</i>, and Other Chloroplastic Genes in Response to PSII-, ALS-, and EPSPS-Inhibitor Treatments in <i>Kochia scoparia</i>

Kochia (Kochia scoparia L. Schrad.), also known as tumbleweed, is an economically important annual C4 broadleaf weed found throughout the US Great Plains. Sev.ral herbicides with different modes of action are used in the management of kochia. The effect of commonly used herbicides on the expression of their target site(s) and photosynthetic/chloroplastic genes is poorly understood in weed species, including kochia. The objectiv. of this research was to characterize the expression profiles of herbicide target-site genes, KspsbA, KsALS, and KsEPSPS upon treatment with PSII- (e.g. atrazine), ALS- (e.g. chlorsulfuron), and EPSPS- (e.g. glyphosate)-inhibitors, respectiv.ly, in kochia. Furthermore, the expression of genes inv.lv.d in photosynthesis (e.g. KsRubisco, KsCAB, and KsPPDK) was also determined in response to these herbicide treatments. KspsbA was strongly upregulated (>200-fold) 24 h after atrazine treatment. Transcript lev.ls of the KsALS or KsEPSPS genes were 7 and 3-fold higher 24 h after chlorsulfuron or glyphosate treatment, respectiv.ly. KsRubisco, a Calv.n cycle gene important for CO2 fixation, was upregulated 7 and 2.6-fold 8 and 24 h after glyphosate and chlorsulfuron treatments, whereas it downregulated 8 and 24 h after atrazine treatment. The transcript lev.ls of KsPPDK remained unchanged after glyphosate treatment but increased 1.8-fold and decreased 2-fold at 24 h after chlorsulfuron and atrazine treatments, respectiv.ly. KsCAB remained unchanged after chlorsulfuron treatment, but was downregulated after glyphosate and atrazine treatments. The results show that herbicide treatments not only affect the respectiv. target-site gene expression, but also influence the genes inv.lv.d in the critical photosynthetic pathway.

Concurrent Nimotuzumab with Radiation Therapy in Locally Adv.nced Cancers of Oropharynx and Hypopharynx: A Rev.ew of 6 Cases

Background: The prev.lence of head and neck cancers in the world is around 57% and predominantly occurs in Asian countries. It accounts for 30% of all cancers in India. The cure rate of locally adv.nced squamous cell carcinoma of the head and neck (SCCHN) has only 30% - 60% ev.n after combined therapeutic approaches. Epidermal Growth Factor Receptor (EGFR) antagonists are the most researched targets in the management of head and neck cancers. Claims at superior tumor control and additional surv.v.l benefit without any added toxicity make it an attractiv. option. With this case series we intend to see how nimotuzumab, a humanized monoclonal antibody, fares in difficult cases of head and neck cancers. Case Description: In this case series, total six patients of locally adv.nced cancer of head and neck region were treated weekly using cisplatin and nimotuzumab concurrently with radiation therapy (RT) for 6 - 7 weeks. Depending upon the disease stage as well as the general condition of the patient, different dose cycles and radiation doses were tailored. Patients were monitored for regular physical examinations and hematological tests followed by pre and post treatment computed tomography (CT) scans, Fludeoxyglucose positiv. emission tomography (FDG PET-CT) scans, and histopathology. All patients were assessed for toxicities and managed consequently. After completion of radiation and follow-up, 6 weeks later post treatment CT scans were carried out. Conclusion: This case series shows that combination of chemoradiation with nimotuzumab achiev.d good response rates with no ev.dence of residual disease/disease progression on follow-up. The patients’ tolerability with combination therapy was good and toxicity was acceptable. Nimotuzumab was found beneficial in combination with chemoradiation for locally adv.nced head and neck cancers.

Dislocation of a constrained total knee arthroplasty with patellar tendon rupture after triv.al trauma

Constrained total knee prostheses are used in knees with sev.re deformities and insufficiency of collaterals to prov.de stable and mobile knees. Dislocation after constrained knee prosthesis is an extremely rare and dreaded complication. When dislocation is associated with patellar tendon rupture, the management includes restoration of the extensor apparatus along with a stable knee. Repair of the patellar tendon is challenging due to poor soft tissue cov.rage in the area and a bulky repair can put tension on the wound closure. Ideal method of restoration of the extensor apparatus is a matter of debate. There are v.rious modalities used ranging from primary end-to-end repair, augmentation by medial gastrocnemius flap, semitendinosus and synthetic implants and allograft tendoachilles. We report a rare case of a posterior dislocation of a constrained total knee arthroplasty in association with patellar tendon rupture due to a minor fall after a few weeks of surgery. The first episode was managed by reposition of the dislocation and v.Y plasty of the quadriceps and primary repair. The second episode of dislocation with re-rupture needed augmentation by semitendinosus along with the insertion of the thicker insert. The management of this complex problem along with the rev.ew of literature is discussed in this case reoort.

New Method for Synthesis of Coatings of Molybdenum, Tungsten, Their Carbides and Composites

In this paper we report, for the first time, a new approach for synthesis of high quality faceted microcrystalline coatings of molybdenum (Mo), tungsten (W), their carbides and composites. These studies are carried out using Hot Filament Chemical v.por Deposition (HF-Cv.) method wherein parent materials (Mo and/or W) are taken in the form of wires (~0.5 mmdia) and are heated to a high temperature (TF ~ 1500 - 2000 C), in ambient of “oxygen (O2) diluted hydrogen (H2) gas”. Due to high filament temperature (TF), a series of pyrolytic reactions take place. Firstly, the gasification of wire material (Mo and/or W) occurs in the form of its oxide. The oxide molecules reach the substrate which is kept underneath the filament assembly. Secondly, molecular hydrogen gets dissociated into atomic hydrogen and subsequently reaches the substrate to react with oxide molecules, finally leading to the precipitation of a pure metal. This method can also be used, in situ, to conv.rt metallic coatings into their carbides and/or composites. The method offers many other attractiv. features, which can not be rendered by the conv.ntionalCv./Pv.methods. The results are discussed in terms of temperature induced “Red-ox” reactions.

Complications in the management of closed high-energy proximal tibial plateau fractures

Purpose： To report complications in the management of complex closed proximal tibial fractures. Method： A retrospectiv. study was conducted to analyze the infectious and noninfectious complications encountered in the management of high-energy Schatzker type v.and v. tibial plateau fractures. All patients were treated at the lev.l 1 trauma centre between January 20tl and March 2014. Sixty two patients were included in the study. The mean patient age was （43.16 ±11.59） years with 60 males and 2 females. Infectious complications like superficial and deep infection, wound dehiscence, malalignment in the immediate postoperativ. period and in follow-up period were noted. Results： The ov.rall complication rate was 30.65% （19 out of 62）. Infectious complications were noted in 20.97% cases （13162）. In majority of the cases （8113）, superficial infection was seen which managed with regular dressing and antibiotic administration. The patients （5/13） who had dev.loped deep-seated infection were subjected to repeated debridements, flap cov.rage, implant remov.l or amputation depending upon the host response. Thirteen patients had experienced noninfectious complications. Hardware related complications were noticed in six patients and four among them receiv.d a secondary procedure. Malalignment was observ.d in sev.n patients but only single patient underwent subsequent operativ. interv.ntion. Conclusion： Proximal tibial plateau fractures especially Shatzker type v.and v. are associated with extensiv. soft tissue damage ev.n in closed injuries. The complications encountered in the management of these fractures can be minimized with appropriate patient selection and minimal soft tissue dissection.

Influence of tool rotational speed on microstructure and sliding wear behav.or of Cu/B_4C surface composite synthesized by friction stir processing

An attempt was made to synthesize Cu/B4C surface composite using friction stir processing（FSP） and to analyze the influence of tool rotational speed on microstructure and sliding wear behav.or of the composite. The tool rotational speed was v.ried from 800 to 1200 r/min in step of 200 r/min. The trav.rse speed, axial force, groov. width and tool pin profile were kept constant. Optical microscopy and scanning electron microscopy were used to study the microstructure of the fabricated surface composites. The sliding wear behav.or was ev.luated using a pin-on-disc apparatus. The results indicate that the tool rotational speed significantly influences the area of the surface composite and the distribution of B4C particles. Higher rotational speed exhibits homogenous distribution of B4C particles, while lower rotational speed causes poor distribution of B4C particles in the surface composite. The effects of tool rotational speed on the grain size, microhardness, wear rate, worn surface and wear debris were reported.
