Effect of Helicobacter pylori-eradication therapy on hepatic steatosis in patients with non-alcoholic fatty liver disease:a randomized–controlled pilot study

Background:Helicobacter pylori infection has been associated with insulin resistance and non-alcoholic fatty liver disease(NAFlD).This study was done to evaluate the effect of ***-eradication therapy(HPET)in patients with NAFlD compared to standard management therapy(SMT).Methods:Eighty NAFlD patients with *** co-infection were randomized into SMT(diet and exercise,n=36)and HPET(SMT plus amoxicillin,clarithromycin,and pantoprazole,n=44)*** controlled attenuation parameter(CAP),anthropometric parameters,liver enzymes,lipid profile,and glycemic parameters including homeostasis model assessment-insulin resistance(HOMA-IR)were measured and compared between two groups at the baseline and 24 ***:Sixty-four participants(SMT group[n=28]and HPET group[n=36])were included in amodified intention-to-treat *** the SMT group and the HPET group had a significant reduction in CAP scores at 24weeks(P=0.002 and Ply),but the change between the groups was insignificant(P=0.213).Successful eradication of *** occurred in 68%of the HPET group and led to greater improvement in HOMA-IR at 24weeks compared to SMT or non-responder patients(P=0.007).The liver enzymes reduced significantly at 24weeks in both groups,but the changes between the groups were *** lipid parameters remained unchanged within the groups and between the groups at 24weeks.A significant increase in the levels of reduced glutathione was noted in the HPET group,but the change between the two groups was not statistically ***:HPET was found to be comparable to SMT alone in reducing hepatic steatosis and liver enzymes at 24 weeks in NAFlD ***,successful eradication of *** led to greater improvement in HOMA-IR(Trial registration CTRI/2017/05/008608).

AIM： To evaluate the anticancer property of the dried latex （Dl） of Calotropis procera, a tropical medicinal plant, in the X15-myc transgenic mouse model of hepatocellular carcinoma and to elucidate its mechanism of action in cell culture. METHODS： The young transgenic mice were orally fed with the aqueous suspension of Dl （400 mg/kg for 5 d/wk） for 15 wk and their liver was examined for histopathological changes at 20 wk. Serum levels of vascu- lar endothelial growth factor （VEGF） were also measured in these animals. To characterize the active fraction, Dl was extracted with petroleum ether followed by methanol. The methanolic extract was sub-fractionated on a silica gel G column using a combination of non-polar and polar solvents and eleven fractions were obtained. Each fraction was analysed for cytotoxic effect on hepatoma （Huh7） and non-hepatoma （COS-1） cell lines and nontransformed hepatocytes （AMl12） using tetrazolium （MTT） assay. Finally, the mechanism of cell death was investigated by measuring the levels of Bcl2, caspase 3 and DNA fragmentation. RESUlTS： Dl treatment of mice showed a complete protection against hepatocarcinogenesis. No adverse effect was observed in these animals. The serum VEGF level was significantly lowered in the treated mice as compared to control animals. Cell culture studies revealed that the methanolic extract of Dl as well as its fraction 8 induced extensive cell death in both Huh-7 and COS-1 cells while AMl12 cells were spared. This was accompanied by extensive fragmentation of DNA in Huh-7 and COS-1 cells. No change in the levels of canonical markers of apoptosis such as Bcl2 and caspase 3 was observed. CONClUSION： Dl of C. procera has the potential for anti-cancer therapy due to its differentJable targets and non-interference with regular pathway of apoptosis.

The present research aimed to improve the dissolution rate and bioavailability of curcumin using the potential of liquisolid technology. Twelve drug-loaded liquisolid systems (lS-1 to lS-12) were prepared using different vehicles (PEG 200, PEG 400 and Tween 80) and curcumin concentrations in vehicle (40%, 50%, 60% and 70%, w/w). The carrier [microcrystalline cellulose (MCC) PH102] to coat (Aerosil®) ratio was 20 in all formulations. The systems were screened for pre-compression properties before being compressed to liquisolid tablets (lT-1 to lT-12). Post compression tests and in vitro dissolution of lTs were conducted and the results compared with those obtained for a directly compressed tablet (DCT) made of curcumin, MCC PH102 and Aerosil®. lTs exhibited higher cumulative drug release (CDR) than the DCT and the optimum formulation, lT-9 (made using Tween 80), was studied by powder XRD, DSC, SEM and FTIR. Ex-vivo permeation of curcumin from lT-9 through goat gastrointestinal mucosa was significantly (Pl bioavailability was increased 18.6-fold in New Zealand rabbits. In vitro cytotoxicity (IC50) of lT-9 towards NCl 87 cancer cells was 40.2 µmol/l substantiating its anticancer efficacy. Accelerated stability studies revealed insignificant effects of temperature and humidity on lT-9. In summary, solubility enhancement of curcumin in lTs produced significant improvements in its permeation and bioavailability.

The present article studies the stability conditions of central control artificial equilibrium generalized restricted problem of three bodies. It is generalized in the sense that here we have taken the larger primary body to be in shape of an oblate spheroid. The equilibrium points are sought by the application of the propellant for which it would just balance the gravitational forces. The launching flight of such a satellite is seen to be applicable for having arbitrary space stations for these different missions. Specialty of the result of the investigation lies in the fact that an arbitrary space station can be formed to attain any specified mission.
