Perinatal inflammation is a significant risk factor for lifelong neurodevelopmental impairments such as cerebral *** clinical and preclinical evidence links the severity and pattern of perinatal inflammation to impaired maturation of white and grey matters and reduced brain *** pathways are involved in the pathogenesis of perinatal ***,studies of human and experimental perinatal encephalopathy have demonstrated a strong causative link between perinatal encephalopathy and excessive production of the pro-inflammatory effector cytokine *** this review,we summarize clinical and preclinical evidence that underpins interleukin-1 as a critical factor in initiating and perpatuating systemic and central nervous system inflammation and subsequent perinatal brain *** also highlight the important role of endogenous interleukin-1 receptor antagonist in mitigating interleukin-1-driven neuroinflammation and tissue damage,and summarize outcomes from clinical and mechanistic animal studies that establish the commercially available interleukin-1 receptor antagonist,anakinra,as a safe and effective therapeutic *** reflect on the evidence supporting clinical translation of interleukin-1 receptor antagonist for infants at the greatest risk of perinatal inflammation and impaired neurodevelopment,and suggest a path to advance interleukin-1 receptor antagonist along the translational path for perinatal neuroprotection.

Acute lymphoblastic leukemia (ALL) is the most common malignancy in children, with the maj.rity of cases being of precursor B-cell phenoltype. Conventional cytogenetic analysis plays an important role in the diagnosis of B-cell ALL, identifying characteristic chromosomal abnormalities associated with a given prognosis therein facilitating optimized treatment. The more recent introduction of microarray technology to the analysis of B-cell ALL has afforded both higher resolution for the detection of known abnormalities and an ability to identify novel copy number abnormalities (CNAs) with potential clinical relevance. In the current study, microarray analysis was performed on 20 cytogenetically abnormal B-cell ALL cases (10 pediatric and 10 adult), while a novel microarray-based balanced-translocation detection methodology (translocation CGH or tCGH) was applied to that subset of cases with a known or suspected recurrent balanced translocation. Standard microarray analysis identified that CNAs was not detected by previous conventional cytogenetics in 75% (15/20) cases. tCGH identified 9/9 (100%) balanced translocations defining BCR/ABL1 (x4), ETV6/RUNX1 (x3), and MLL/AFF1 (x2) breakpoints with high resolution. The results illustrate the improved molecular detail afforded by these technologies and a comparison of translocation breakpoints, CNAs and patient age offers new insights into tumor biology with potential prognostic significance.

Metabolizable Amino Acid Requirements of Feedlot Calves

One hundred ninety-two crossbred steers (229 kg) were used to evaluate the influence of metabolizable amino acid intake on growth performance and health during the initial 42-d receiving period. Treatments consisted of four levels of metabolizable lysine (23, 24, 25 and 26 g/kg diet DM). Morbidity averaged 36%, and was not affected (P > 0.20) by treatments. No steers died during the study. Increasing the metabolizable lysine supply increased DMI, ADG, gain efficiency, and dietary NE (linear effect, P < 0.01). Metabolizable amino acid supply of the basal diet was determined using 6 steers (214 kg) with cannulas in the rumen and proximal duodenum. Metabolizable amino acid supply of the diet was in close agreement with expected (R2 = 0.99;P < 0.0001) based on NRC (2000) [12] Level 1 model. The metabolizable amino acid supplies for treatments in Trial 1 were estimated by adding tabular metabolizable amino acid values for the respective supplemental proteins (cottonseed meal and fish meal) to the observed metabolizable amino acid supply of the basal diet. Treatment effects on metabolizable lysine supply explained 99% of the variation (P < 0.01) in ADG, and 91% of the variation in observed versus expected dietary NE. The biological value for the intestinal chyme was determined based on chemical score technique, using bovine tissue as the reference protein. Accordingly, methionine and lysine were closely co-limiting amino acids having ratios of 77% and 79%, respectively. We conclude that current NRC standards reliably predict both requirements and supplies of metabolizable amino acids for feedlot calves. Diet formulations that do not meet the metabolizable amino acid requirements may depress both ADG and the partial efficiency of utilization of metabolizable energy for maintenance and gain.

Basal diet and indigestible marker influence apparent digestibilities of nitrogen and amino acids of cottonseed meal and soybean meal in pigs

This study was to determine the apparent ileal nitrogen(N)and amino acid digestibilities of cottonseed meal(CSM)and soybean meal(SBM)in simple carbohydrate-based and more complex wheat-based *** five Large White × Landrace boars(40.9 kg)were randomly allocated to one of 5 dietary treatments:a wheat diet,40% CSM in either a sugar:starch(1:1)or wheat-based diet,or 40% SBM in either a sugar:starch or wheat-based diet for 14 *** diets contained vitamins,minerals,and contained acid-insoluble ash(AIA)and Cr2 O3 as indigestible *** were offered(1,800 g/pig per d)in3 meals/d on d 1-11 and 8 meals/d on d *** d 14,the pigs were fed hourly for 8 *** the 8 th meal,pigs were anaesthetised with isoflurothane,and ileal and faecal digesta sampled from the terminal ileum and rectum before lethal inj.ction of *** ileal digestibility(AID)of N was greater(12.1%)when CSM was fed in the wheat-based diet as compared to the simple carbohydrate-based diet,whereas apparent ileal N digestibility of SBM was slightly lower(-4.5%)in the wheat-based diet as compared to the sugar:starch-based *** ileal amino acid digestibility generally respondeds similarly to ***,while there was a wide difference in apparent N and amino acid digestibilities of SBM and CSM when they were fed in the sugar:starch-based diets,these differences were less apparent when they were fed the wheat-based *** was an apparent net release of N into the hindgut of pigs fed CSM in both base ***,there was quite substantial apparent digestion of N in the hindgut of pigs fed SBM in both base *** use of Cr2 O3 as an indigestible marker resulted in lower apparent ileal and faecal digestibilities than using AIA,particularly for diets containing *** data demonstrate that the basal diet and choice of indigestible marker can substantially influence the estimate of apparent N digestibility,this response can differ for different protein meals.

No evidence for a genetic association between female mating preference and male secondary sexual trait in a Lake victoria cichlid fish

Sexual selection by female mating preference for male nuptial coloration has been suggested as a driving force in therapid speciation of Lake victoria cichlid fish. This process could have been facilitated or accelerated by genetic associations betweenfemale preference loci and male coloration loci. Preferences, as well as coloration, are heritable traits and are probably determinedby more than one gene. However, little is known about potential genetic associations between these traits. In turbid water,we found a population that is variable in male nuptial coloration from blue to yellow to red. Males at the extreme ends of thephenotype distribution resemble a reproductively isolated species pair in clear water that has diverged into one species withblue-grey males and one species with bright red males. Females of the turbid water population vary in mating preference coincidingwith the male phenotype distribution. For the current study, these females were mated to blue males. We measured the colorationof the sires and male offspring. Parents-offspring regression showed that the sires did not affect male offspring coloration,which confirms earlier findings that the blue species breeds true. In contrast, male offspring coloration was determined by theidentity of the dams, which suggests that there is heritable variation in male color genes between females. However, we found thatmating preferences of the dams were not correlated with male offspring coloration. Thus, there is no evidence for strong geneticlinkage between mating preference and the preferred trait in this

Seasonal tropical cyclone(TC)forecasting has evolved substantially since its commencement in the early ***,present operational seasonal TC forecasting services still do not meet the requirements of society and stakeholders:current operational products are mainly basin-scale information,while more detailed sub-basin scale information such as potential risks of TC landfall is anticipated for decision *** fill this gap and make the TC science and services move forward,this paper reviews recent research and development in seasonal tropical cyclone(TC)*** particular,this paper features new research topics on seasonal TC predictability in neutral conditions of El Ni˜no–Southern Oscillation(ENSO),emerging forecasting techniques of seasonal TC activity including Machine Learning/Artificial Intelligence,and multi-annual TC *** also review the skill of forecast systems at predicting landfalling statistics for certain regions of the North Atlantic,Western North Pacific and South Indian oceans and discuss the gap that remains between current products and potential user's *** knowledge and advanced forecasting techniques are expected to further enhance the capability of seasonal TC forecasting and lead to more actionable and fit-for-purpose products.
